Many small, rural communities struggle with aging or inadequate water resource recovery facilities (WRRFs), and face challenges in constructing and operating such systems. Although existing literature has provided insight into the environmental sustainability of large systems including both the construction and operational phases, these studies have not examined small systems in adequate depth or breadth. The main objective of this study was to develop a mix design adjustment method for Fiber Reinforced Concrete (FRC) that would maintain appropriate workability while improving hardened concrete performance. A literature review was conducted to examine existing methods for adjusting mix designs to account for fiber introduction. It was found that while increasing fine aggregate and cement paste content can make up for lost workability with the addition of fibers, no rational mix design adjustment method is available. Due to the relatively high cement content and low water-to-cement ratio (w/c) used, bridge deck concrete is prone to premature cracking. Internal curing has been found to greatly reduce the chance of premature cracking as well as concrete deterioration. Conjugation is a main mechanism for the horizontal gene transfer of ARGs from wastewater microbiome to the microbiome indigenous to surface water. Some damages such as Alkali Silica Reaction (ASR) tend to start from the inside, where there is no easy way to identify or evaluate them in the early stage. Manure is applied to cropland areas with varying surface cover to meet single- or multiple-year crop nutrient requirements. The objectives of this field study were to (1) examine microbial transport following land application of manure to sites with and without wheat residue, (2) compare microbial loads following land application to meet the 0, 1, 2, 4, and 8-year P-based requirements for corn, and (3) evaluate the effects of rainfall simulation run on microbial transport. Hyperspectral reflectance data in the visible, near infrared and shortwave infrared range (VIS–NIR– SWIR, 400–2500 nm) are commonly used to nondestructively measure plant leaf properties. We investigated the usefulness of VIS–NIR–SWIR as a high-throughput tool to measure six leaf properties of maize plants including chlorophyll content (CHL), leaf water content (LWC), specific leaf area (SLA), nitrogen (N), phosphorus (P), and potassium (K). This study examines the potential for replacing the standard intersection design at two-way stop control (TWSC) and all-way stop control (AWSC) intersections along rural highways with a roundabout or a restricted crossing u-turn (RCUT) facility. The geometry of the RCUT design prohibits left and through movements from the side road, and provides a u-turn location downstream from the main crossing. This type of facility has been implemented for rural highways extensively in other states and has the potential to serve as a cost-effective solution to improve roadway safety within Nebraska.

At ethanol plants, the control of volatile organic compound containing emissions from fermentation tanks and distillers dried grains with solubles driers is accomplished through the use of air scrubbers and regenerative thermal oxidizers. The operation of these control units imposes substantial operating costs for the ethanol plant. Two dormitory halls at the University of Nebraska-Lincoln known as Cather and Pound halls were demolished via controlled implosion on December 22, 2017. Cather and Pound halls were two thirteen-story reinforced concrete structures. The demolition of these two structures included the implosion of controlled charges at selected columns on alternating floors which initiated the progressive collapse of these structures. A new steel, side-mounted, beam-and-post, bridge rail was designed, crash-tested, and evaluated according to safety performance guidelines included in the American Association of State Highway and Transportation Officials Manual for Assessing Safety Hardware (MASH) for Test Level 4 (TL-4). The new bridge rail system was designed to be compatible to multiple concrete bridge decks utilized by the States of Illinois and Ohio. Vegetative filter strips (VFS) consisting of perennial vegetation have been successfully used to reduce the transport of contaminants in runoff from land application areas. The effectiveness of a winter wheat strip, which may be more acceptable to producers, in reducing microbial transport was examined in this study. Nebraskan streams contribute excess nitrogen to the Mississippi/Atchafalaya River Basin and Gulf of Mexico, which results in major water-quality impairments. Reducing the amount of nitrogen (N) exported in these streams requires the use of best management practices (BMPs) within the landscape. The presence of clay particles increases the specific surface area of a soil and can affect the calibration of electromagnetic soil water sensors including reflectometers. To quantify and correct for this effect in two relatively new reflectometers, three TDR315 and three CS655 sensors were installed in each of five soils with clay content ranging from 5 to 49%. Crop residue is an abundant resource for the potential production of biofuels, but a better understanding of its use on net carbon emissions must be developed to mitigate climate change. Xylanases decrease the xylan content in pretreated biomass releasing it from hemicellulose, thus improving the accessibility of cellulose for cellulases. In this work, an endo-β-1,4-xylanase from Aspergillus fumigatus var. niveus (AFUMN-GH10) was successfully expressed. The structural analysis and biochemical characterization showed this AFUMN-GH10 does not contain a carbohydrate-binding module. The enzyme retained its activity in a pH range from 4.5 to 7.0, with an optimal temperature at 60°C. AFUMN-GH10 showed the highest activity in beechwood xylan. The mode of action of AFUMNGH10 was investigated by hydrolysis of APTS-labeled xylohexaose, which resulted in xylotriose and xylobiose as the main products. AFUMN-GH10 …

Land application of manure introduces gastrointestinal microbes into the environment, including bacteria carrying antibiotic resistance genes (ARGs). Measuring soil ARGs is important for active stewardship efforts to minimize gene flow from agricultural production systems; however, the variety of sampling protocols and target genes makes it difficult to compare ARG results between studies. We used polymerase chain reaction (PCR) methods to characterize and/or quantify 27 ARG targets in soils from 20 replicate, long-term no-till plots, before and after swine manure application and simulated rainfall and runoff. This study investigates the microstructural evolution and mechanical response of sputter-deposited amorphous silicon oxycarbide (SiOC)/crystalline Fe nanolaminates, a single layer SiOC film, and a single layer Fe film subjected to ion implantation at room temperature to obtain a maximum He concentration of 5 at. %. X-ray diffraction and transmission electron microscopy indicated no evidence of implantation-induced phase transformation or layer breakdown in the nanolaminates. Substrate mediated gene delivery (SMD) is a method of immobilizing DNA complexes to a substrate via covalent attachment or nonspecific adsorption, which allows for increased transgene expression with less DNA compared to traditional bolus delivery. It may also increase cells receptivity to transfection via cell-material interactions. Substrate modifications with poly(acrylic) acid (PAA) brushes may improve SMD by enhancing substrate interactions with DNA complexes via tailored surface chemistry and increasing cellular adhesion via moieties covalently bound to the brushes.
