Chronic inflammation is a major cause of the pathogenesis of musculoskeletal diseases such as fragility, fracture, and nonunion. Studies have shown that modulating the immune phenotype of macrophages from proinflammatory to pro-healing can heal recalcitrant bone defects. Current therapeutic strategies predominantly apply biochemical cues, which often lack target specificity, and controlling their release kinetics in vivo is challenging spatially and temporally. We have developed a magnetic iron-oxide nanocomplexes (MNC)-based therapy for resolving chronic inflammation in the context of promoting fracture healing. Hydrophobic deep eutectic solvents (DESs) have emerged as excellent extractants. A major challenge is the lack of an efficient tool to discover DES candidates. Currently, the search relies heavily on the researchers’ intuition or a trial-and-error process, which leads to a low success rate or bypassing of promising candidates. The emergence of deep learning models and their success in visual object recognition have fueled the medical imaging community's interest in integrating these algorithms to improve medical diagnosis. However, natural images, which have been the main focus of deep learning models and mammograms, exhibit fundamental differences. First, breast tissue abnormalities are often smaller than salient objects in natural images. Second, breast images have significantly higher resolutions but are generally heavily downsampled to fit these images to deep learning models. Experimental process simulation and quantification of microstructure development during processing are challenging due to limitations with machinery temperature capability, inadequate resolution and sampling volume of currently available characterization techniques, and difficulty characterizing material microstructures as close to processing-relevant conditions as possible. This dissertation addresses how process simulation can be performed using Gleeble thermomechanical technologies and how microstructure development during these processing simulations can be quantified both in-situ and ex-situ.

This thesis presents a new parallelized real-time path planning process. This process is an extension of the Real-Time Rapidly Exploring Random Trees* (RT-RRT*) algorithm developed by Naderi et al in 2015 [1]. The RT-RRT* algorithm was demonstrated on a simulated two-dimensional dynamic environment while finding paths to a varying target state. We demonstrate that the original algorithm is incapable of running at a sufficient rate for control of a 7-degree-of-freedom (7-DoF) robotic arm while maintaining a path planning tree in 7 dimensions. In June 2021 a series of high altitude flights were conducted in Spaceport America, NM, using a balloon launched Uncrewed Aircraft System (UAS) to assess its capability to conduct measurements of various atmospheric properties and study turbulence in the troposphere and lower stratosphere. This UAS descends using an automated flight trajectory. The instruments aboard included a NASA-developed infrasonic microphone to evaluate its remote turbulence detection capabilities and a five-hole probe capable of measuring the in situ wind vector. Currently, there is no standard in vitro model for studying the effects of mechanical stimulation on muscle in type II diabetes. Existing models primarily utilize electrical stimulation, which does not fully recapitulate the effects of exercise. In this thesis, we create a standardized in vitro model of murine muscle that can recapitulate the benefits seen in exercise when mechanically stimulated. Moreover, we show that a type II diabetes environment has similar effects on the muscle in vitro as well as in vivo.

Yttria-stabilized tetragonal zirconia polycrystal (Y-TZP) has gained popularity as the choice of material for dental prosthetics. Ivoclar Vivadent’s IPS e.max ZirCAD Prime dental ceramic incorporates a unique gradient technology that varies the yttria content over the thickness of the material. The top layer is composed of 5Y-TZP which is desired for its optical properties while the bottom layer is composed of a much stronger 3Y-TZP. The Herculaneum scrolls were buried and carbonized by the eruption of Mount Vesuvius in A.D. 79 and represent the only classical library discovered in situ. Charred by the heat of the eruption, the scrolls are extremely fragile. Since their discovery two centuries ago, some scrolls have been physically opened, leading to some textual recovery but also widespread damage.
