Chronic inflammation is a major cause of the pathogenesis of musculoskeletal diseases such as fragility, fracture, and nonunion. Studies have shown that modulating the immune phenotype of macrophages from proinflammatory to pro-healing can heal recalcitrant bone defects. Current therapeutic strategies predominantly apply biochemical cues, which often lack target specificity, and controlling their release kinetics in vivo is challenging spatially and temporally. We have developed a magnetic iron-oxide nanocomplexes (MNC)-based therapy for resolving chronic inflammation in the context of promoting fracture healing. Wearable technologies for continuous monitoring of cerebral hemodynamics in freely behaving subjects not only advance our understanding of cognitive processing and adaptive behavior, but also provide vital information for diagnosis and therapeutic assessment of cerebral diseases associated with hypoxia/ischemia. Wearable near-infrared diffuse optical techniques have been used at the bedside to probe deep cerebral hemodynamics, including near-infrared spectroscopy (NIRS) for cerebral oxygenation measurement and diffuse correlation spectroscopy (DCS) for cerebral blood flow (CBF) measurement, respectively. Currently, there is no standard in vitro model for studying the effects of mechanical stimulation on muscle in type II diabetes. Existing models primarily utilize electrical stimulation, which does not fully recapitulate the effects of exercise. In this thesis, we create a standardized in vitro model of murine muscle that can recapitulate the benefits seen in exercise when mechanically stimulated. Moreover, we show that a type II diabetes environment has similar effects on the muscle in vitro as well as in vivo.

Almost one in every three people worldwide is infected with Toxoplasma gondii (T. gondii). The biology and growth of the parasite’s bradyzoite form in host tissue cysts are not well understood. T. gondii’s metabolic state influences the morphology of its single mitochondrion, which can be visualized using fluorescence microscopy with specific dyes. Hence, fluorescence microscopy images of cysts purified from infected mouse brains carry biological information about bradyzoites, the poorly understood form of the parasite within them. The equine racing industry is one of the main proponents in Kentucky’s economic infrastructure. In this industry there has come a need to investigate the nature of the proximal sesamoid bone (PSB). Breakdowns involving the PSBs are the leading cause in racehorse deaths in the industry, with still little known about what causes this bone to fracture. This study seeks to shed insight by investigating the internal structure of the PSB. Using microCT scanning, the internal porous structure was captured. We investigated if listening to songs changes breathing pattern which changes autonomic responses such as heart rate (HR) and heart rate variability (HRV) or change in breathing pattern is a byproduct of listening to songs or change in breathing pattern as well as listening to songs causes changes in autonomic responses. Seven subjects (4 males and 3 females) participated in a pilot study where they listened to two types of songs and used a custom developed biofeedback program to control their breathing pattern to match the one recorded during listening to the songs.

Approximately a third of the population worldwide is chronically infected by the parasite Toxoplasma gondii. During chronic infection the parasite resides within tissue cysts as the poorly understood bradyzoite form. Observation of these bradyzoites via microscopic imaging within tissue cyst purified from infected mouse brains has shown metabolic activity with heterogeneous replication potential. The objectives of this study were to analyze effects of day-long exposure to LBP risk factors on lumbo-pelvic coordination (LPC) in nursing occupations and to verify if physical activity level affects diurnal work-related changes in LPC. Thirty-three nurses were recruited into three groups based on workplace physical demands and each completed two data collection sessions, one before and one after their 8-12 hour work shift. Surgical resection may be an option for some of these patients, but localization of the epileptogenic zone is difficult and expensive. Individuals with low back pain (LBP) demonstrate an abnormal lumbopelvic coordination compared to back-healthy individuals. This abnormal coordination presents itself as a reduction in lumbar contribution and an increase in pelvic contribution to trunk motion. This study investigated the ability of a hip orthosis to correct such an abnormal lumbo-pelvic coordination by restricting pelvic rotation and, as a result, increasing lumbar contributions. The effects of the hip orthosis on the magnitude and timing characteristics of lumbopelvic coordination were investigated in 20 patients with LBP and 20 asymptomatic controls. Normal movement execution requires proper coupling of motor and sensory activation. An increasing body of literature supports the idea that incorporation of sensory stimulation into motor rehabilitation practices increases its effectiveness. Paired associative stimulation (PAS) studies, in which afferent and efferent pathways are activated in tandem, have brought attention to the importance of well-timed stimulation rather than non-associative (i.e., open-loop) activation. Abnormal lumbar movement has been observed in individuals who have a history of low back pain (LBP). Affected individuals display a reduction in lumbar spine rotation during trunk movement tasks, while pelvic rotation increases to compensate. Measurement of tissue hemodynamics provides vital information for the assessment of tissue viability. This thesis reports three noninvasive near-infrared diffuse optical systems for spectroscopic measurements and tomographic imaging of tissue hemodynamics in vulnerable tissues with the goal of disease diagnosis and treatment monitoring. A hybrid near-infrared spectroscopy/diffuse correlation spectroscopy (NIRS/DCS) instrument with a contact fiber-optic probe was developed and utilized for simultaneous and continuous monitoring of blood flow (BF), blood oxygenation, and oxidative metabolism in exercising gastrocnemius. The high prevalence and mortality of cerebrovascular disease has led to the development of several methods to measure cerebral blood flow (CBF) in vivo. One of these, arterial spin labeling (ASL), is a quantitative magnetic resonance imaging (MRI) technique with the advantage that it is completely non-invasive. The quantification of CBF using ASL requires correction for a tissue specific parameter called the brain-blood partition coefficient (BBPC). Despite regional and inter-subject variability in BBPC, the current recommended implementation of ASL uses a constant assumed value of 0.9 mL/g for all regions of the brain, all subjects, and even all species.

Abnormalities in lumbopelvic rhythm (LPR) play a role in occurrence/recurrence of low back pain (LBP). The LPR before spinal fusion surgery and its changes following the surgery are not understood. A repeated measure study was designed to investigate timing and magnitude aspects of LPR in a group of patients (n = 5) with LBP before and after a spinal fusion surgery. Participants completed a forward bending and backward return task at their preferred pace in the sagittal plane. Volumetric muscle loss (VML) is a debilitating injury which results in full or partial loss of function. Current clinical options utilize tissue grafts and bracing to restore function. Tissue graft implantation oftentimes leads to serious complications, some of which end in graft rejection and thereby necessitate further surgeries and procedures. Polymeric scaffolds show promise as scaffolding systems due to their mechanical properties and overall degradation profiles. Scaffolds need appropriate mechanical properties, 10-60 kPa modulus, and overall degradation times, five days to two weeks, to initiate tissue regeneration. Heavy backpacks have been suggested to have a pathogenic role in experience of low back pain among children. We have conducted a repeated-measure study to investigate the backpack-induced changes in lumbo-pelvic coordination of forty gender-balanced college age students when they walked and jogged on a treadmill with two different types of backpacks: normal and ergonomically modified. The backpack-induced changes in lumbo-pelvic coordination were larger when carrying an ergonomically modified vs. a normal backpack as well as when jogging versus walking. The larger changes in lumbo-pelvic coordination when carrying an ergonomically modified backpack were likely due to kinematic restraints imposed by …
