Consolidated Chamber Design And Protocol For Olfactory Conditioning Assay With Drosophila Melanogaster, Sasha Bronovitskiy, Andres Castillo, Michael Yan, Fang Ju Lin

The olfactory conditioning assay is widely used in Alzheimer’s disease research to quantify learning and memory in Drosophila melanogaster. The assay tests ability to recall an aversive conditioned stimulus of scent paired with electrical shock when presented a choice between shock-associated and unrelated scents. The T-maze, a commonly used apparatus for olfactory conditioning assays, employs an elevator mechanism to transfer live flies from the shock-delivering training chamber to the scent selection point. This elevator mechanism is known to cause fly casualty. T-mazes are not commercially available and often difficult to reproduce. Other existing variations of olfactory conditioning apparatuses use …

Antibiotics have been our most effective weapon against bacterial infections since their discovery in the early 1900s. Their use has been critical in reducing mortality rate from infectious diseases. However, in the last few decades, the overuse of antibiotics, beginning at an early age and into adulthood, has become a growing concern globally. Penicillin is one of many extensively used antibiotics in early childhood that has been used to treat childhood infections. The purpose of this research is to provide an alternative to naturally derived bone grafts. There is a gap in the supply of donors and the demand of bone tissue. Artificial scaffold creation can work as an implant and decrease the shortage of bone grafts and increase the range of injuries that can be repaired. Current research focuses on optimizing mechanical properties such as porosity, improving vascularization using cells, and generating osteoconductivity. For osteodifferentiation, mesenchymal stem cells (MSCs) can differentiate into mesodermal lineages such as chondrocytes, osteoblasts, adipocytes, and tenocytes by supplementing cultures with lineage-specific soluble factors (Marchetti). The photosynthetic efficiency of Cornus controversa leaves was decreased significantly under Cd treatment while it was not affected by Pb exposure. Cd decreased while Pb treatment increased the chlorophyll contents of Cornus controversa leaves. Furthermore, the peroxidase (GPX) activities were decreased after Cd treatment while elevated by Pb exposure in Cornus controvera seedlings. In addition, both Cd and Pb exposures increased the malondialdehyde (MDA) and proline contents and elevated the superoxide dismutase (SOD) activities of Cornus controvera seedlings. Collectively, these results indicated that Cornus controversa may be more tolerant to Pb than Cd toxicity. This finding will contribute to the …

Sperm whale myoglobin (Mb) has weak dehaloperoxidase activity and catalyzes the peroxidative dehalogenation of 2,4,6-trichlorophenol (TCP) to 2,6-dichloroquinone. Crystals of Mb and of its more active G65T variant were used to study the binding of TCP, 4-iodophenol (4-IP) and phenol. The structures of crystals soaked overnight in a 10 mM solution of phenol revealed that a phenol molecule binds in the proximal cavity, forming a hydrogen bond to the hydroxyl of Tyr146 and hydrophobic contacts which include interactions with C and C of the proximal histidine His93. The phenol position corresponds to the strongest xenon binding site, Xe1. It …
