Technique And Performance Level Comparisons Of Male And Female Hammer Throwers, Suzanne M. Konz

The aim of this study was two-fold: 1) what the hammer throwing technique differences between sexes are and 2) what technique parameters help determine throw distance. The performances of the top 16 male and female throwers at the 2003 World Athletic Final and the top 13 male and female throwers from the 2003 USA Track and Field Nationals were examined. Video was captured using three Canon 60 Hz cameras. The best throws of each athlete were digitized and analyzed using the Peak Motus 8.2 motion analysis system. Expression And Function Of Alpha3 And Beta2 Neuronal Nicotinic Acetylcholine Receptor Subunits In Hek-293 Cells, Nathan W. Steinhafel

Single-cell real-time quantitative RT-PCR was used to characterize the mRNA expression of rat neuronal nicotinic acetylcholine receptor (nAChR) subunits α3 and β2 in CA1 hippocampus stratum radiatum and stratum oriens interneurons. α3β2 co-expression was detected in 43% of interneurons analyzed. The nAChR subtype α3β2 was transiently expressed in cells derived from the human embryonic kidney cell line 293 at mRNA levels found in the CA1. The functional properties of α3β2 in HEK-293 cells were characterized by whole-cell patch clamping using acetylcholine (ACh) as an agonist. Although it is well-established that animals will self-stimulate electric current to various diverse brain structures, the neural substrate of brain stimulation reward (BSR) has eluded identification since its discovery more than a half-century ago. We show that GABA neurons in the midbrain, hypothalamus and thalamus express connexin-36 (Cx36) gap junctions and couple electrically with dopamine application or by stimulation of the internal capsule (IC), which also supports self-stimulation. In this collection of papers I have summarized my investigations into the field of evolutionary genetics and more specifically into patterns of biodiversity and evolutionary processes. The lizards (and frogs) studied here share common features in that they are largely present in unique environments, which are also regions that are biologically understudied. Most of these taxa show high degrees of endemism, interesting natural history characteristics, and each group manifests distinctive adaptations of general evolutionary interest. The harsh environmental and poor economic conditions of the Bolivian Altiplano require intervention to assist many of those that live there to become economically self-sufficient. We attempted to find introduced dry season reserve forage grasses that could produce enough biomass to be useful as feed for livestock, and that could also produce enough seed to distribute to farmers. While some of the grasses produced reasonable amounts of biomass, none produced seed in quantities that would be even close to being economically viable. Several novel polymer monoliths for the analysis of peptides and proteins were synthesized using polyethylene glycol diacrylate (PEGDA) as crosslinker. Photo-initiated copolymerization of polyethylene glycol methyl ether acrylate and PEGDA yielded an inert monolith that could be used for size exclusion liquid chromatography of peptides and proteins. This macroscopically uniform monolith did not shrink or swell in either water or tetrahydrofuran. More importantly, it was found to resist adsorption of both acidic and basic proteins in aqueous buffer without any organic solvent additives. A strong cation-exchange polymer monolith was synthesized by copolymerization of 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS) and PEGDA. A ternary …

Cranial placodes are ectodermal regions that contribute extensively to the vertebrate peripheral nervous system. The development of the ophthalmic trigeminal (opV) placode, which gives rise only to sensory neurons of the ophthalmic lobe of the trigeminal ganglion, is a well-studied model of sensory neuron development. While key differentiation processes have been characterized at the tissue and cellular levels, the molecules governing opV placode development have not been well described. Photovoltaic cells and fluorescence sensing are two important areas of research in chemistry. The combination of photon-activated electron donors with electron acceptors provides a strong platform for the study of optical devices. A series of four oligothiophene-ruthenium complexes has been synthesized. Variation in oligothiophene length and bipyridine substitution allowed comparison of these variables on electronic properties. The longer oligothiophenes display lower energy absorption and emission compared to the shorter ones. Aromatic conjugation appears more complete with para-, rather than meta-, substitution. Oligothiophenes and Ru(bpy)32+ are highly fluorescent individually, but fluorescence is quenched when connected. Bonds of carbon to fluorine are …

Objective: To determine the extent to which coaching over the telephone is an effective method in promoting the loss of body weight and body fat percentage. Research Methods and Procedures: Over a period of four months, in a randomized, double blind, placebo-controlled study, 120 overweight and obese (BMI 25-35 kg/m²) adults either received telephone coaching or no coaching. In addition, each participant was randomly assigned to take a supplement or placebo daily. Body weight and body fat percentage were measured at baseline, two months, and four months. Characterization of phosphoproteins-including detection, identification of phosphoproteins and identification of phosphorylation sites-is mostly done with radiolabeling and proteomic techniques. Three main topics related to phosphoprotein characterization are included in this dissertation. First, large-scale characterization of the CHO (Chinese hamster ovary) cell phosphoproteome was done using two dimensional gel electrophoresis (2DE) separation, visualization of phosphoproteins by radiolabeling or a phosphoprotein specific dye, followed by MALDI-TOF identification. Because radiolabeling of phosphoproteins is very sensitive and straightforward to quantify, such analysis can give a clear picture of the relative phosphosphorylation of proteins present in a sample. Exposure of human erythrocytes to elevated intracellular calcium causes alterations in cell shape and stimulates shedding of the cell membrane in the form of microvesicles. We hypothesized that both the shape transition and microvesiculation are influenced by microscopic membrane physical properties such as lipid order. To test this hypothesis, membrane properties were manipulated by varying the experimental temperature, membrane cholesterol content, and the internal ionic environment. Changes in membrane order were assessed using steady-state fluorescence spectroscopy with an environment-sensitive probe, laurdan. The focus of this study was to elucidate the molecular and cellular mechanisms whereby fibroblast growth factors (FGFs) mediate outgrowth of the limb. Specifically, we examined the epistatic relationship between FGF and Wnt/Planar cell polarity (PCP) signaling in establishing cell polarity as a mechanism for outgrowth. By implanting beads into embryonic limbs and lateral plate mesoderm, we established that FGF activates Wnt5a in a gradient fashion. Once it was established that Wnt5a was expressed at the right time and place to turn on PCP signaling, we investigated the ability of Wnt5a to influence cell migration and/or cell polarity. Critical mule deer (Odocoileus hemionus) winter ranges in southeastern Utah dominated by Wyoming big sagebrush (Artemisia tridentata var. wyomingensis [Beetle and A. Young] Welsh) have developed dense cheatgrass (Bromus tectorum L.) understories. These communities are currently characterized by predominately mature to decadent stands of sagebrush with few perennial grasses and forbs. Sagebrush seedlings and perennial grasses compete for limited resources with annual grasses and forbs. The analysis of proteins in biological fluids by capillary electrophoresis (CE) is of interest in clinical chemistry. However, due to low analyte concentrations and poor concentration limits of detection (CLOD), protein analysis by this technique is frequently challenging. Coupling preconcentration techniques with CE greatly improves the CLOD. This thesis uses a background subtraction to produce high-quality silhouettes for use in human identification by human gait recognition, an identification method which does not require contact with an individual and which can be done from a distance. A statistical method which reduces the noise level is employed resulting in cleaner silhouettes which facilitate identification. The thesis starts with gathering video data of individuals walking normally across a background scene. Background: Cartilage disorders affect millions of people in the United States alone, with effects ranging from poor skeletal development and joint pain to shortened lifespan and perinatal lethality. Many of these disorders have their root in defects of collagen, type XI collagen being among the most important. A mouse model of such a type XI collagen defect is the chondrodysplasia (cho) mutant. Mice homozygous for this null mutation in the Col11a1 gene do not express the α1 chain of type XI collagen. This results in a functional knockout of type XI collagen, leading to insufficient skeletal development and perinatal lethality. …

Purpose: Reducing body temperature before exercise is called precooling. Past research suggests that reducing body core temperature (Tc) slightly can result in improved running performance. This study evaluated the effects that warming up, while wearing a cooling vest prior to an interval workout, had on Tc, and interval time in long-distance runners. Methods: Nineteen healthy male collegiate cross-country runners were recruited for this study. Ferritins are ubiquitous iron storage proteins in living systems. Although much is known about the iron deposition process in ferritin and a mechanism has been developed, several important issues still remain unknown. One lingering question is the less than stoichiometric quantities of hydrogen peroxide detected in previous studies on animal ferritins. The mountain whitefish (Prosopium williamsoni) has been largely overlooked in population genetic analyses despite its wide distribution in discrete drainage basins in western North America for over four million years. Its closest sister taxa the Bear Lake whitefish (P. abyssicola), Bonneville cisco (P. gemmifer), and Bonneville whitefish (P. spilonotus) are found only in Bear Lake Idaho-Utah and were also included in the analyses. Objective: To investigate the potential etiology of a loss of neck control injury in female standard ballroom dancers. The median frequency (MF) as measured by electromyography (EMG) of the left upper trapezius (UT), left splenius capitius (SPL), and right sternocleidomastoid (SCM) of injured dancers was compared to non-injured dancers. This comparison was performed to identify whether dancers with a history of loss of neck control have a greater amount of fatigue than those with no history of this particular injury. A key component to conservation is an accurate understanding of genetic subdivision within a species. Despite their ecological and economic importance, relatively little is understood about the genetic structuring of Yellowstone cutthroat trout in Yellowstone National Park. Here, we use traditional (Fst, Rst, Nm, and AMOVA) and modern (Bayesian assignment tests, coalescent theory, and nested clade analysis) analytical approaches to describe the population genetic subdivision of cutthroat trout spawning populations in Yellowstone Lake and to identify genetically distinct population segments throughout Yellowstone National Park. In this study, the single-cell relative quantitative mRNA expression of three Calcium-binding proteins (CaBPs; calbindin, calretinin, parvalbumin) and eight nicotinic acetylcholine receptor (nAChR) subunits (alpha2-alpha5, alpha7, beta2-beta4) from interneurons from the stratum radiatum or stratum oriens within the CA1 region of rat hippocampi was analyzed using quantitative real time RT-PCR. Eighty-seven percent of the interneurons examined expressed CaBP mRNA. Parvalbumin mRNA was detected in 64%, while calbindin and calretinin expression was detected in 26% and 40% of interneurons, respectively. The inaccessibility and cost of soil testing reduce effectiveness of fertilizer use on small-scale subsistence farms, and inadequate funding promotes adoption of soil tests in developing countries with minimal validation. For example, Mehlich-I extraction of phosphorus (P) currently used extensively in Guatemala may not be suitable for its broad range of soils. At least four alternatives are available but are relatively untested [Bray 1, Mehlich III, Olsen and pressurized hot water (PHW)]. Glycosphingolipids (GSLs), composed of a polar saccharide head and a lipophilic ceramide tail, are ubiquitous components of the plasma membrane of eukaryotic cells. They serve in many regulatory capacities and have antigenic properties towards natural killer T (NKT) cells of the innate immune system. Critical to the recognition of glycosylceramides by NKT cells are antigen presenting cells (APC), such as dendritic cells, which are responsible for binding, processing, and delivery of ligands to these lymphocytes. Quinoa (Chenopodium quinoa) is an Andean crop adapted to harsh environmental conditions and containing a high and well-balanced protein composition. Two seed storage proteins, the 2S albumin and 11S globulin, are the major amino acid reservoirs for the developing seedling. An in-depth study of the genes encoding these proteins is necessary to understand the source of quinoa's protein quality. Our objectives include identification and sequencing of the 11S genomic and cDNA clones, analysis of 11S expression profiles in different quinoa accessions and evaluation of evolutionary relationships between the sequence of the 11S gene in quinoa and homologous genes in other …
