Adverse childhood experiences (ACEs) include various forms of physical and emotional abuse that lead to detrimental long term effects. Agalychnis callidryas (red-eyed tree frogs) have been identified as a novel biomedical model for ACEs. Red-eyed tree frogs demonstrate an escape hatch mechanism under stressors including those from predators, flooding and hypoxia where the embryo undergoes an accelerated developmental process similar to what is observed in the human population regarding precocious puberty. In order to assess this model for neurobiological outcomes of ACEs, morphological and genomic protocols must be developed and validated which is this study’s focus. In terms of metabolism, eating disorders manifest in two extreme directions: overnutrition, which can lead to obesity, and malnutrition, which can result in underweight or even starvation. Both extremes compromise the quality of life. According to the Diagnostic and Statistical Manual of Mental Disorders-5 (DSM-5) standard, eating disorders affect up to 17.9% of young women and 2.4% of young men. Although eating disorders are primarily defined as mental disorders rather than metabolic disorders, they are intertwined with complex emotions and sensory perceptions. Chronic stress has detrimental effects on psychological and physiological health. Incapability to adapt to chronic stress can lead to depression, anxiety disorders, and addiction. The brain regions critical for reward processing share diverse physiological and molecular changes linked to depressive-like behaviors as a result of chronic stress. Cumulative evidence shows that counteraction or prevention of these alterations in neurons, occasionally in a projection-specific manner, can enhance stress adaptation or resilience.

The locus coeruleus (LC), the primary site of brain norepinephrine (NE), is a key anatomical brain region implicated in the stress response. Stress is a neuroendocrine physiologic response to a stressor that promotes organism survival through adaptive change and restoration of homeostasis. The central stress response, which drives behavioral and physiological change, is primarily mediated by activating the hypothalamic-pituitary-adrenal (HPA) axis. This study aimed to investigate the potential impact of negative social media content on athletes' cortisol levels and subsequent performance. The study focused on the change in cortisol levels and differences in free throw performance, based on previous research findings. We hypothesized that negative social media postings would increase the stress experienced by student-athletes, resulting in elevated cortisol levels and decreased performance. Additionally, participants (n = 8) completed a questionnaire to examine the interaction between preexisting fear and the biological stress response. For women, two of the greatest risk factors for affective disorders are adversity experienced during puberty and later becoming pregnant. We have created a translationally relevant mouse model where we address these complex risk factors. Previously, we discovered that pregnant mice (dams) that experienced chronic variable stress (CVS) during puberty display a blunted hypothalamic-pituitary-adrenal (HPA) response when exposed to an acute stressor. Interestingly, this alteration only first becomes apparent during pregnancy, which is a sensitive period for these effects due to normative neuroendocrine changes. Huntington Disease (HD) is a fatal neurodegenerative disorder that is characterized by motor, cognitive, and psychiatric symptoms. Although HD onset is determined by motor symptoms, psychiatric symptoms, like depression and aggression, can develop earlier, have a larger impact on quality of life, and are understudied due to stigma. Our lab has observed hyper aggression in our humanized HD mouse model (Hu97/18) compared to our knock-in HD mouse model (Q175FDN). We characterized these differences and found that the Hu97/18 mice overreact in neutral situations, behaving as if they are in threatening situations. The brain senses inflammatory signals and drives the release of glucocorticoids (GCs) — potent immunosuppressants — via the activation of the hypothalamic-pituitary-adrenal (HPA) axis. This inflammation-induced HPA axis activation is largely mediated by prostaglandin E2 (PGE2), acting on two subtypes of the PGE2 receptor, EP1 and EP3. Major depressive disorder is one of the most pervasive psychiatric illnesses in the United States. Women are at greater risk for developing depression, particularly during their childbearing years. Approximately 17% of new mothers develop postpartum depression within 4 weeks after parturition. The risk for postpartum depression is even greater in women who do not breastfeed or stop breastfeeding early. Repeated exposure to stress is known to have a myriad of effects on the brain, contributing to the development of psychiatric disorders, such as anxiety, depression, and drug addiction. For example, rats undergoing repeated social stress develop increased cocaine self-administration. Despite the apparent sex difference in prevalence of anxiety in humans, pre-clinical studies that have led to anxiolytic drug discoveries between 1960 and 2012 used male animals and approximately 6% of 10,000 studies used female animals. The generalizability of the efficacy of these drugs to both sexes may be limited if data derived are predominantly based on the male brain. The purpose of this dissertation, therefore, was to investigate potential underlying neuronal mechanisms that could be contributing to the sex differences in stress-related mood disorder prevalence and to focus on shedding light on female brains.

The common marmoset is a good model for research because they are easy to house and have complex social relationships (French et al., 2019). Marmosets are sensitive to social isolation, and when introduced to a stressor, the HPA axis is activated (Saltzman & Abbott, 2011). The purpose of this experiment is to validate marmosets as a translational model for stress due to social relationships in humans. This is done by validating biomarker concentration levels at baseline, then comparing the concentration when introduced to a stressor. This dissertation describes the use of Drosophila neuroblasts (NBs) to model human ribosomopathies; the overall goal is to understand why specific stem cell and progenitor cell populations are the primary targets in nucleolar stress as seen in the ribosomopathies. Chapter 1 provides an overview of relevant literature. Chapter 2 describes nucleolar stress in Drosophila neuroblasts as a model for human ribosomopathies. For this, we induce nucleolar stress by using the UAS-GAL4 system to express RNAi that depletes Nopp140 transcripts, and we also employ homozygous, CRISPR-Cas9-generated Nopp140 gene disruptions with a systemic null phenotype (Nopp140-/-). The stress response is essential for survival, but it can be detrimental when persistently activated during chronic stress. Therefore, the magnitude of the stress response must be finely-tuned in order to be adaptive. Indeed, an animal’s stress response system normally undergoes habituation that decreases responses to a repeated, familiar stressor. Importantly, the same animal still responds normally, or even becomes sensitized, to an unfamiliar (novel) stressor. This indicates that chronic stress simultaneously induces two opposing types of plasticity, one that underlies habituated responses and another that drives sensitized responses. Early Life Immune And Physical Stress Directly Influences Anxiety-Like Behaviour In Adolescent Rats: Examining Sex Differences, Jordan M. Ward

This thesis examined the effects of neonatal acute immune activation with the endotoxin, lipopolysaccharide (LPS) on postnatal days 3 and 5 on adolescent anxiety-like behaviour in rats before and after a stress period. Previous research has shown that adults rats exposed to LPS during the neonatal stage show anxiety-like behaviour following a period of stress. This thesis investigated this effect in adolescence. The present results showed significantly higher anxiety-like behaviour in saline controls, and a potential neuroprotective effect of low dose LPS (15 µg/kg) contrary to what was reported in adult rats. An increasing number of studies show that people employed as first responders in high trauma service jobs tend to experience a high level of stress, at work and after hours. Studies suggest that constant exposure to job related stress leads to both physical and mental dysregulation. This study looks at the effects of implementing mindfulness tools and techniques to those working in law enforcement. It is well known that arginine vasotocin (AVT) in birds is involved in physiological homeostasis such as cardiovascular, osmotic regulation as well as reproductive functions. Pertinent to these physiological functions, AVT immunoreactive (-ir) neurons in the hypothalamus have been found associated with hemorrhage, dehydration, oviposition and other physiological regulation. Evidence, however, suggests that AVT also plays significant roles in modulating behavior, memory, stress, and food intake. This dissertation research addresses the latter two neuroendocrine functions of AVT in detail within the chicken brain.
