After vanishing from the public eye for more than 50 years, bed bugs have resurged to become one of the most widely discussed and heavily researched insect pests in the westernized world. Our inability to prevent and successfully treat infestations has been the driving force behind this wave of research. I addressed gaps in our understanding of bed bugs by examining behavioral and life history characteristics, as well as insecticide application responses. I showed that natural-based products are generally ineffective against bed bugs, particularly when used as a residual treatment. I also found that bed bugs may be killed through …

To investigate how soil microbial diversity is influenced by the formation of an experimental edge-creating gap within a southern New England oak-hickory forest, I used a molecular fingerprinting technique known as terminal restriction fragment length polymorphism (TRFLP). Sequence variability in the 16S ribosomal RNA (rRNA) gene in soil bacterial communities is detected by differences in the length and abundance of fragments produced by digesting PCR products amplified from rRNA genes. The different patterns observed are assumed to represent unique phylotypes.

Fusion genes occur due to chromosomal instability where two previously separate genes rearrange and fuse together, forming a hybrid gene. The first fusions were reported in leukemias; however, with the advent of more powerful sequencing technologies, fusions have recently been reported in several solid tumors. Using next-generation deep sequencing approaches, we discovered a fusion gene connecting the fibroblast growth factor receptor 3 (FGFR3) gene to the transforming coiled-coil containing protein 3 (TACC3) gene in glioblastoma multiforme. Biologically relevant short sequences of DNA have previously been shown to possess high affinity and specificity for binding of small polyamide structures to their minor groove. These structures, or minor groove binders, have the potential to control access to specific regulatory regions of DNA. Anti-platelet drugs are the mainstay treatment because of the direct involvement of platelets in the initiation and propagation of thrombosis. However, the currently available anti-platelet drugs, such as antagonists of platelet receptors or of effector systems participating in platelet activation, have their own limitations. A new mode of affecting platelet reactivity may prove to offer unique advantages in a host of clinical settings. Heart disease is the major cause of mortality in the United States and other parts of the world. Heart transplantation is the treatment of choice for patients with end stage heart failure. However, transplanted organs fail due to either acute or chronic rejection. This acute and chronic rejection impacts distinct compartments of cardiac allografts. Acute rejection is characterized by infiltration of mononuclear cells whereas chronic rejection is characterized by progressive narrowing of coronary arteries. Trypanosoma brucei is a protozoan parasite that causes sleeping sickness in humans and Nagana in cattle. They evade the host's immune defense by periodically switching their major surface antigen, variant surface glycoprotein (VSG), a phenomenon termed antigenic variation. Inside its mammalian host, bloodstream form (BF) T. brucei monoallelically expresses its major surface molecule VSG from the VSG Expression Sites (ESs) located at subtelomeric loci. Monoallelic VSG expression ensures effective antigenic variation and maximizes the efficiency of T. brucei pathogenesis. Inflammation is an obligatory attempt of the host immune system to protect the body against infection. However, unregulated synthesis of pro-inflammatory products can have detrimental effects. Though mechanisms which contribute to inflammation are well appreciated, those that resolve inflammation are poorly understood. Therefore, understanding the molecular basis of such pathways will provide an entirely novel approach to treat and prevent inflammatory diseases. Deregulated mTORC1 contributes to tumorigenesis and chemoresistance. However, mTORC1-specific inhibitors (rapalogs) show modest efficacy in the clinic, as they unleash the feedback inhibition on upstream, prosurvival pathways. An alternative approach is to target downstream functions of mTORC1. We investigated acquired resistance to fludarabine (Flu), a purine analog, active agent for chronic lymphocytic leukemia (CLL). Elevated phospho-p70S6k (T389), an mTORC1 activation marker, predicted Flu resistance in a panel of B-cell tumor lines, and primary CLL cells. Talin is a cytosolic protein which is known to be one of the key players involved in integrin mediated cell adhesion dependent processes, including blood coagulation, and tissue remodeling. It connects the extracellular matrix with the actin cytoskeleton. Talin comprises of a head domain (talin-H) and a rod domain (talin-R). Talin-H is further subdivided in F0, F1, F2 and F3 domains. Talin-R contains 13 contiguous helical bundle domains (R1-R13) followed by an actin binding dimerization domain (DD). Invasion by non-native species has had significant ecological and economic impacts on a global scale. In the state of Florida, Old World climbing fern (Lygodium microphyllum) is an invasive plant listed by FLEPPC as a category one invader with significant ecological impacts that threaten native plant diversity. The objective of this study was to conduct an avian survey of five water sources (four ephemeral former cattle tanks and one perennial spring), determine how many species inhabit or use Indio Mountains Research Station, and show a hierarchal similarity between the survey sites. This survey was conducted in a Chihuahuan Desert scrub landscape on Indio Mountains Research Station (IMRS) in Hudspeth County, Texas. There have been no previous formal studies of the avifauna at IMRS. All species accounts or sightings have been from casual sightings.

The black sea bass, Centropristis striata, is a member of the family Serranidae that is commercially important throughout its range, which extends throughout the western Atlantic (from Cape Cod, Massachusetts to Cape Canaveral, Florida) and Gulf of Mexico (from Mobile Bay, Alabama to Tampa Bay, Florida). There are two known subspecies, C. striata striata in the Atlantic and C. striata melana in the Gulf, and through behavioral and morphological evidence two separate stocks are managed in the Atlantic, north and south of Cape Hatteras, NC. Quarterly samples were used to estimate assemblage-level (all species combined) fish production within three minimally-impacted, southern West Virginia streams. The total annual fish production estimate was highest in Slaunch Fork (37.52 kg∙ha-1∙y-1), a tributary of the Tug Fork River, and lowest in Cabin Creek (10.59 kg∙ha-1∙y-1), a Guyandotte River tributary. Creek Chub Semotilus atromaculatus, Mottled Sculpin Cottus bairdii and Blacknose Dace Rhinicthys atratulus were the most abundant species among sites, accounting for >90% of all sampled individuals. The HIV protein Rev regulates the expression of essential viral proteins during the course of infection by a mechanism that is well understood. It promotes nuclear export of viral transcripts, normally retained in the nucleus owing to the presence of introns, by interacting with host cell transport factors. However, over-expression of Rev in cells leads to defects in cell cycle progression, specifically slowing growth and impairing progression through mitosis (43). While it is possible that Rev may be altering the proteins in transport pathways, cell cycle defects may be attributed to Rev's interactions with other proteins.

Nuclear and mitochondrial genomes have co-adapted through evolution. Interactions between these genomes are required for mitochondrial functions. My goal was to determine how divergence of these interactions affected mitochondrial function and organismal fitness. The impact of divergence between Caenorhabditis briggsae populations and between C. briggsae and its sister species, C. nigoni was assessed. To accomplish this, four sets of hybrid lines were constructed. Each hybrid line possessed the nuclear genome of one parental strain and mitochondria from another parental strain. Mutations are the ultimate source of the biological diversity on which natural selection acts, but the vast majority of these mutations are harmful. As such, mutations lead to disease states like cancer, extinction of small populations, and can drive pathogen evolution. Unfortunately, because mutations are rare and past studies have been subject to detection biases, very little is known about the distribution of fitness effects from naturally occurring mutations.
