Learning to anticipate significant events accurately is a crucial element of survival for all species. The process by which animals acquire this knowledge has been a central question of psychological research. A fundamental assumption of many learning theories is that the predictive value assigned to cues is not simply determined by their probability of reinforcement but rather by their ability to compete with other cues present during learning. Anxiety-related attention bias (AB), a type of cognitive bias underlying the development and maintenance of anxiety disorders, is typically defined as exaggerated attention towards threat; however, some individuals also demonstrate AB away from threat and in some instances, no detectable AB at all. Debate about the reliability and validity of AB behavioral measures has prompted examination of additional assessment methods. Neurophysiological methods, such electroencephalography (EEG), have yielded encouraging early findings. The steady-state visually evoked potential (SSVEP), a measure of selective visual attention derived from EEG, holds promise as a biosignature for AB. Early life adversity (ELA) is the exposure to a single or to multiple traumatic events before the age of 18 that go beyond the child’s coping. These adverse events are often exacerbated during adolescence particularly when cognitive performance is compromised. Adolescents who experienced ELA may show symptoms of post-traumatic stress disorder (PTSD), while not vividly recalling the early life trauma. These individuals show atypical connectivity between prefrontal-amygdala and hippocampus, all of which is associated with an increased risk of experiencing a traumatic event again later in life. Anxiety disorders are among the most common mental health diagnoses, affecting about a third of the population in their lifetime. However, approximately a third of individuals with anxiety do not respond to current treatment approaches, highlighting the need to identify additional potential therapeutic mechanisms. Safety learning is one such mechanism, but methodological challenges and a dearth of research have prevented the field from advancing the understanding of the role of safety learning in the etiology and remediation of anxiety disorders. Anxiety affects nearly twice as many women as it affects men across all cultures and economic groups. Importantly, girls have a higher chance of inheriting anxiety disorders than boys, and many anxiety disorders appear at a very young age. However, little is known about sex differences in brain and behavioral development and how they relate to anxiety in adulthood. Anxiety disorders will affect an estimated one in three Americans, significantly impacting emotional health and quality of life. High personal and economic costs make research on the etiology, maintenance, and treatment of anxiety a crucial public health goal. Selective and exaggerated attention towards threat, termed attention bias (AB), has been identified as a core behavioral and neurocognitive mechanism in anxiety. A novel treatment, attention bias modification training (ABMT), targets AB and aims to ameliorate anxiety. The serotonergic system has been the major candidate in the pathophysiology of mood related disorders such as anxiety and major depressive disorder (MDD). Unfortunately, current antidepressant drugs are ineffective in 50% of the population and require chronic administration for a period of 3-6 weeks before the onset of therapeutic response. 5-HT4 receptor (5-HT4R) agonists have emerged as potential candidates for fast antidepressant action, since an antidepressant response can be achieved after 3 days of pharmacological administration in rodents.
