Low volume fungicide smoke particle deposits were studied with scanning electron microscopy and energy dispersive X-ray analysis. Under greenhouse conditions, particles deposited on Rhododendron leaf surfaces and onto inert, carbon planchets adjacent to Rhododendron leaves were characterized by electron beam analysis. Angular particles containing chlorine, identified as vinclozolin deposition, were quantified on a per unit area basis. Leaf samples collected from plants immediately adjacent to the smoke applicator had more particles than leaf samples collected elsewhere. More deposition was measured on leaf surfaces than on inert planchet surfaces. This study increased our knowledge about the effectiveness of using less fungicide …

Embryonic blood vessels have not been grown in organ culture, in a way which might easily submit them to studies of vascular organogenesis. The chick embryo dorsal aorta is easily accessible and relatively simple to explant to culture. Its organ culture may provide a model for wounding and repair of the intima and/or media and provide a model for studies of growth (or maintenance) and differentiation. Fresh dorsal aorta and its organ culture from the chick embryo was characterized morphologically by light microscopy scanning (SEM) and transmission electron microscopy (TEM) from days 4 to 18. Calcium oxalate crystals occur on the sporangiolar appendages and aerial hyphae of the two known genera (Hesseltinella, Radiomyces) of the mucoralean family Radiomycetaceae. In Hesseltinella the sporangiolar appendages are acicular with a polygonal, square, or hexagonal, discoid base. The appendage core consists of calcium oxalate and is composed of two portions: (1) a round, acicular spine with a more or less hexagonal base, and (2) a square, polygonal, or hexagonal, discoid base with a central perforation that generally contains the spine base. A simple, rapid method for visualizing the internal morphology of amphibian embryos is described. Fixed embryos of Ambystoma mexicanum are surrounded with commercial embedding material and are frozen. Internal structure is revealed by cutting the embryos with a cooled scalpel or razor blade, removing the frozen embedment with an aqueous buffer, and processing the halved embryos for scanning electron microscopy (SEM). This technique allows the internal anatomy of the embryo to be viewed but is much simpler and more rapid than previously described techniques. The vascular architecture of male gonads of Blennius pavo is studied by scanning electron microscopy of vascular corrosion casts. Arterial supply to the gonads is by a branch of the first ventral segmental artery of the tail. From the surface of the gonads, this vessel gives rise to branches which supply testes, spermatic ducts, testicular glands, blind pouches, urogenital sinus and urogenital papilla. The testis has a rope-ladder-like capillary network around the seminiferous tubules, while in the testicular gland the capillary network is irregular in form. The backscattered electron (BSE) induced secondaries (SE2) emerge from an area that is usually many orders of magnitude larger than the area in which the impinging primary probe releases secondary electrons (SE1). These SE2 secondary electrons form a) an undesired background signal in high resolution scanning micrographs and b) are responsible for the well known proximity effect in electron beam lithography. The ciliated epithelium of the rabbit's trachea was investigated after a single 5 mg dose of cis-diamminedichloroplatinum (cis-DDP). Specimens were taken for scanning electron microscopy, transmission electron microscopy and light microscopy. Examination was performed daily for 20 consecutive days. A cytotoxic effect of the drug on the ciliated epithelium was observed with bent ciliary tips, swollen tips and broken cilia. Finally the cilia were lost and large areas of the surface were covered with microvilli. However, 20 days after the drug injection, the restitution of the ciliary carpet was almost complete.

As crystals are important etiologic agents for disease, their accurate identification in tissues and body fluids is of utmost importance. This paper surveys the roles of crystals in disease process and outlines current analytical techniques for crystal detection and identification in bone tissues. The value of multiple correlated techniques is demonstrated including scanning electron microscopy, x-ray energy spectroscopy and powder diffraction analysis. The current feasibility of utilizing intermediate voltage scanning transmission analytical electron microscopy to integrate these analytical techniques on the same tissue sample is emphasized.

The backscattered electron signal can be detected using a solid state detector system. Because this signal is affected by the surface topography it is necessary to determine the optimal detection condition for a registration mark detection in the electron-beam lithography and for an automatic topographical surface reconstruction.

The BSE signal from surface marks has been measured using semiconductor detectors with a small collection angle. Calculations of the BSE signal have been carried out by means of a simplified model of electron backscattering. In order to apply SEM-based automated image analysis (AIA) to the characterization of not only minerals in coal but to the coal itself, sample preparation methods need to be developed beyond common practice. A significant consideration is the degree of contrast achievable between the mount media chosen and the coal. Four low-atomic number materials (epoxy, polyethylene, polystyrene and carnauba wax) were compared for their potential as suitable mounting media for coal samples. Epoxy is satisfactory only for characterization of mineral particles since the contrast between epoxy and coal particles is negligible. Scanning electron microscopic (SEM) autoradiography of the lung is being used to determine the distribution of inhaled, alpha particle-emitting, plutonium dioxide particles. SEM autoradiography provides high visual impact views of alpha activity. Particles irradiating the bronchiolar epithelium were detected both on the bronchiolar surface and in peribronchiolar alveoli. The technique is being used to obtain quantitative data on the clearance rates of plutonium particles from bronchi and bronchioles.

In this tutorial we describe our methods for preparing detergent-extracted cytoskeletons for observation by high resolution scanning (SEM) and scanning transmission (STEM) electron microscopy. We both discuss the theoretical background and provide practical procedures for each of the following steps: cell culture on Formvar-coated gold grids; prefixation with aldehydes or protein crosslinking reagents (homobifunctional N-hydroxy-succinimide esters); extraction with Triton X-100 or Brij 58 detergent in microtubule stabilizing buffer; postfixation in formaldehyde and glutaraldehyde; dehydration; critical point and freeze drying; sputter coating with 1-2 nm of platinum or tungsten; and examination by SEM and both normal and inverted contrast STEM. Zinc deficiency in man results in multisystem disease. It may be acquired or hereditary; the latter can be fatal if left untreated. Premature babies are particularly susceptible to zinc deficiency. Unfortunately no simple, reliable test for zinc status exists at present.

Short, newly-emerging scalp hair samples from 3 classical cases of zinc deficiency all showed the same characteristic abnormalities when examined by scanning electron microscopy, i.e. straight, blunt tips bearing scales, unusually thick cuticular scales with jagged free-edges, and very fine longitudinal corrugations in individual scales. Canine chromosomes are not only numerous (38 autosomal pairs), but they are small (compared to human chromosomes) and morphologically similar as well. Analysis of the canine karyotype by light microscopy (LM) of banded chromosomes is, thus, difficult, and the literature on the canine karyotype is scanty. In this study, we describe examination of chromosomes from normal and chronically irradiated dogs with the scanning elect ran microscope (SEM). Metaphase chromosomes from bone marrow aspirates were Giemsa-banded with either 0.025% trypsin alone or 0.1% trypsin preceded by 10% H2O2 and prepared for SEM. Concentrations of small fossil mammals are frequently encountered in Cenozoic deposits, but the causes for such accumulations have seldom been determined. In many cases the tooth, jaw, and limb fragments appear to be well-preserved under light microscopy, and it is difficult to differentiate damage due to predator digestion from breakage and abrasion due to physical agents. In order to find more specific evidence of predator digestion, we used a scanning electron microscope (SEM) to examine the surface microstructure of bones and teeth consumed by Bubo virginianus (great horned owl) and Canis latrans (coyote), which prey upon similar species. The vascular system of the sciatic nerve of chicken and rats was examined by means of the microcorrosion casting technique and freeze-broken specimens.

The main epineural vessels form two lateral and interfascicular vascular bundles which anastomose with one another and also with the peri -and endoneural plexuses. On epi- and perineural vessels one can find morphological correlates for regulative means such as sphincters. Even the endoneural vessels depict numerous anastomoses.

Heart myocytes and capillaries are enmeshed in a complex array of connective tissue structures arranged in several levels of organization: epimysium, the sheath of connective tissue that surrounds muscles; perimysium, which is associated with groups of cells; and endomysium, which surrounds and interconnects individual cells. The present paper is a review of work in this field with an emphasis on new, unpublished findings, including composition of endomysial fibers and disposition of newly described perimysial fibers. Signal mixing technique using asymetrically placed backscattered electron detectors in a scanning electron microscope is presented in this paper. Two types of detectors have been used: a low-take off angle ring scintillation detector (placed around the specimen) and a wide-angle semiconductor detector (placed above the specimen). It has been shown that the discussed configuration gives good "real" topography in all directions on the specimen surface and also reduces significantly the pseudo-topography effect of flat grain boundaries.

Six crown and bridge alloys ranging in nobility between 25-63 wt % (18-45 at %) were analyzed by optical microscopy, scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and secondary ion mass spectroscopy (SIMS), as well as by L*a*b* colorimetry before and after in vitro tarnishing in artificial saliva with and without additions of 0.00016, 0.016, and 1.6 % Na2S with a rotating wheel apparatus. Electron microscopy was used to characterize the fine-grained products formed during the electrorefining of copper and to elucidate the chemical processes occurring during this metallurgical process. Copper anodes and the anode slimes from the electrorefining of copper at three Canadian refineries are studied to determine the behaviour of lead and selenium and the formation of various silver-copper selenides during the electrorefining process. Silver occurs in solid solution in the copper metal and dissolves with the copper. Silver then reacts with Cu2Se to form Ag-Cu selenides. Lead is present in the anode mostly as complex Pb-As or Pb-As-Sb-Bi oxides. …

The post-storage quality of fruits depends on the ultrastructure of the outer tissues and on the fruit mineral composition (mostly on calcium). Pectins and their bonds with calcium play a central role in the tissues during ripening.

During senescence the middle lamella disappears from the cell walls, the fruits become soft and cell wall reticulation can be seen. There are changes within the cells too; plasmalemma separates from the wall, tonoplast from the cytoplasm, and plastids, mitochondria and nuclei are disorganized.

Two kinds of whey powder, one containing only amorphous lactose, and the other containing precrystallized lactose, and spray-dried amorphous lactose were examined for particle structure by scanning electron microscopy. Lactose crystallization under 75th relative humidity was studied by X-ray diffraction analysis. Scanning electron microscopy appeared to detect changes due to lactose crystallization on the smooth surface of the particles examined. Lactose crystallization s tarted within 30 minutes after exposure to the humid atmosphere. Both a-lactose hydrate and B-lactose crystals were detected in the s pray- dried amorphous lactose by X -ray diffraction within 40 minutes under the humid conditions. Only …

Scanning Electron Microscopy (SEM) and X-ray Microanalysis (EDS) have been used to investigate product container interaction problems in plain tinplate metal food containers and to examine glass and glass-like particles that were found in canned food.

Through the use of SEM-EDS, it was determined that: sulfate containing particles from the cannery cooling water reacted with exposed iron or tin-iron alloy on the tinplate surface causing an external rusting problem; sulfur dioxide was responsible for a container discoloration and a pitting corrosion problem in canned fruit nectar; hydrogen sulfide produced from the SO2-tinplate reaction was the cause of an associated off-odor …

Electron microscopy techniques were used to study the attachment matrix of Pseudomonas fragi used to demonstrate the presence of attachment fibrils of P. fragi adhered to stainless steel. Transmission electron microscopy (TEM) was used to examine thin sectioned cells strained with ruthenium red or alcian blue. Extracellular substances surrounding cells stained with ruthenium red or alcian blue had a mat- or spike-like morphology. This indicated that these substances were acidic mucopolysaccharides that may be involved in the attachment of P. fragi to food contact surfaces.

Immature and mature Granny Smith and Red Delicious apples (Malus domestica Borkh . ) were studied with scanning electron microscopy (SEM). Parenchyma cells were observed to form a net-like pattern in sections transverse to the stem-calyx of the apple. Bundles of about six intact cells were connected, creating large intercellular spaces. The intercellular spaces were different in shape from different perspectives to the cut surfaces ; round in stem-calyx transverse sections and elliptical in stem-calyx cross sections.
