Cell migration in complex micro-environments, that are similar to tissue pores, is important for predicting locations of tissue nucleation and optimizing scaffold architectures. Firstly, how fibroblast cells - relevant to tissue engineering, affect each other’s directional decisions when encountered with a bifurcation of different channel lengths was investigated. It was found that cell sequence and cell mitosis influence the directional choices that the cells made while chemotaxing. Specifically, the fibroblasts chose to alternate between two possible paths - one longer and the other shorter - at a bifurcation. The challenges involved in modeling biological systems are significant and push the boundaries of conventional modeling. This is because biological systems are distinctly complex, and their emergent properties are results of the interplay of numerous components/processes. Unfortunately, conventional modeling approaches are often limited by their inability to capture all these complexities. By using in vivo data derived from biomedical imaging, image-based modeling is able to overcome this limitation.

Targeted nanoparticle drug delivery has the potential to replace current forms of cancer therapy with previously unparalleled efficiency. Upon introduction into the human body, nanoparticles exhibit a substantial increase in diameter due to a biomolecular corona formation caused by interactions between blood plasma proteins and the nanoparticles. These interactions must be analyzed and understood for targeted delivery to reach its potential in both feasibility and efficiency.

In the coating of paper, properties like strength, durability, and print quality are affected not only by the binder type, but the final location of the binder relative to the pigments and the fibers. The purpose of this study was to develop and validate an instrument that can measure how senior chemical engineering students make process safety decisions. The Engineering Process Safety Research Instrument (EPSRI) contains dilemmas that represent process safety scenarios, followed by three decision options, and 12-15 considerations that fall into pre-conventional, conventional, or post-conventional forms of reasoning. Three studies were completed as a part of this research. The content validation study ensured the dilemmas represented process safety scenarios, the considerations matched their perceived theoretical definitions, and that no content areas were omitted. Recent developments in downhole temperature measurements open new alternatives contributing to reservoir characterization. In this dissertation, novel forward and inverse models to analyze production- and injection-induced temperature signals are developed for conventional and unconventional reservoir applications. Important limitations of the proposed models are addressed by accounting for fluid property variations and complex production strategies.
Forward modeling approaches involve making relevant assumptions that allow rigorous analytical solutions to be constructed using Laplace transform, Method of Characteristics, and control volume analysis. Cyanobacteria have the potential to be low-cost and sustainable cell factories for bio-products; however, many challenges face cyanobacteria as biorefineries. This dissertation seeks to advance non-model photosynthetic organisms for biotechnology applications by characterizing central carbon metabolism and its regulations. Cyanobacteria phenotypes for bio-production are examined and their intracellular metabolism is quantified. Using isotopic labeling experiments, phenotypic relationships between biomass composition, central carbon fluxes, and metabolite pool sizes are investigated. Metabolic analyses of cyanobacteria led to new investigations of flux regulation mechanisms via protein spatial organizations or metabolite channeling. Recently discovered, graphene and graphene oxide materials have drawn considerable research attention due to outstanding and novel properties, which underpin wide material potential for a number of advanced applications including supercapacitors, solar cells, sensors, catalysts, semiconductors, sorbents, and membranes, among others. Graphene oxides (GO), which are considered as a family of oxidized graphene materials (derivatives), is a key precursor to the synthesis of free-standing graphene via oxidation-exfoliation-reduction pathways. With the advancements of modern technology, computers and softwares make simulation models possible such as fire models to simulate fire and smoke movements. In this paper, a computational fluid dynamic (CFD) software Fire Dynamic Simulator (FDS) is used to conduct a series of atrium tests to investigate the effectiveness of smoke exhaust systems. FDS solves the Navier-Stokes equations appropriate for low speed flows (Ma < 0.3) with an emphasis on smoke, heat transport and CO2 concentrations from fires. The default turbulence model used in FDS simulation is the Large Eddy Simulation (LES), which is the solution of Navier-Stokes equations at low speed.

Bones, in addition to bearing loads, play an important role in balancing P levels in our bodies. The main objective of this work is toimprove the accuracy on calculations of design set pressure and performance curves (sizing equations) for Gas-Lift Valves (GLVs) at high pressure (real conditions). This improved calculation method is demonstrated to have an important effect on well unloading and production operations.

Remediation of textile wastewater, particularly concerning dyes, has been a longstanding concern. With the rise of more stringent regulations, the study of advanced methods of waste treatment is becoming necessary. The objective of this study was to synthesize and observe the use of a heterogeneous catalyst in a Fenton reaction by doping an SBA-15 catalyst support with iron. The catalyst was successfully synthesized, was easily filterable, exhibited resistance to acidic environments, and showed thermal stability. Along-wind dispersion, or stretching of the cloud in the direction of the wind, plays an important role in the concentration and modeling of contaminant releases in the atmosphere. Theoretical and empirical derivations were compared for appropriate parameterization of along-wind dispersion. Available field data were analyzed to evaluate and improve previous parameterizations of the along-wind dispersion coefficient, σ_x. Current insulation solutions across multiple industries, especially the commercial sector, can be bulky and ineffective when considering their volume. Aerogels are excellent insulators, exhibiting low thermal conductivities and low densities with a porosity of around 95%. Such characteristics make aerogels effective in decreasing conductive heat transfer within a solid. These requirements are crucial for aerospace and spaceflight applications, where sensitive components exist among extreme temperature environments. At Amgen, the Automated Vision Inspection (AVI) systems capture the movement of unwanted particles in Amgen's drug product containers. For quality inspection, the AVI system must detect these undesired particles using a high speed spin-stop agitation process. Woo Pig Sulfate (WPS) has developed two methods for reducing levels of calcium sulfate in mine impacted waters, a “hot process” and a two-stage nanofiltration process. Both processes were designed to treat gypsum saturated water that is currently being pumped via interceptor wells to prevent the spread of a sulfate plume into a nearby community. Each solution was designed for Freeport McMoran’s Sierrita mine in Green Valley, Arizona, which was visited in order to gain insight about the problem. Below 250 ppm on the treated stream, nanofiltration and the hot process achieve overall recoveries of 84 and 99%, respectively. The …

Heart failure is the main cause of death for both men and women in the United States. The only proven treatment for patients with heart failure is heart transplantation. The goal of this research is to create patches of tissue that could mimic the function of the native heart to repair the damaged portions of the heart. Wildland fire, which includes both planned (prescribed fire) and unplanned (wildfire) fires, is an important component of many ecosystems. Prescribed burning (controlled burning) is used as an effective tool in managing a variety of ecosystems in the United States to reduce accumulation of hazardous fuels, manage wildlife habitats, mimic natural fire occurrence, manage traditional native foods, and provide other ecological and societal benefits. During wildland fires, both live and dead (biomass) plants undergo a two-step thermal degradation process (pyrolysis and combustion) when exposed to high temperatures. Optimization has enabled automated applications in chemical manufacturing such as advanced control and scheduling. These applications have demonstrated enormous benefit over the last few decades and continue to be researched and refined. However, these applications have been developed separately with uncoordinated objectives. This dissertation investigates the unification of scheduling and control optimization schemes. The current practice is compared to early-concept, light integrations, and deeper integrations. This quantitative comparison of economic impacts encourages further investigation and tighter integration. Sepsis is a severe blood infection caused by bacteria entering the blood stream. Sepsis caused by antibiotic resistant bacteria is very dangerous with a high mortality rate. The current clinical diagnostic methods for sepsis require culturing the blood sample prior to other steps of the diagnosis procedure. Culturing the blood samples is a time-consuming step which increases the time required for the diagnostic procedure. This research applies techniques from the field of optimization to spatial positioning and energy management in Unmanned Aerial Vehicles (UAVs). Two specific areas are treated: optimization of UAV view plans for 3D modeling of infrastructure, and trajectory optimization of solar powered high-altitude long-endurance (HALE) UAVs. Structure-from-Motion (SfM) is a computer vision technique for creating 3D models from 2D images. View planning is the process of planning image sets that will effectively model a given scene.
