Assessing Mitochondrial Activity In Embryos From Heat-Stressed Ova, Kimberly Ann Nagle

Elevated ambient temperatures negatively impacts pregnancy rates in agriculturally important females. An oocyte undergoing meiotic maturation exposed to a physiologically-relevant heat stress results in alterations of developmentally important processes and reportedly decreases subsequent developmental potential after fertilization. To address problems of reduced competence after exposure to heat stress for the first 12 h of meiotic maturation (hIVM), Payton (2009) assessed the transcriptome profiles of oocytes and associated cumulus cells by microarray analysis and reported heat-induced perturbations at the molecular level in the oocyte. The Hox genes are a family of highly conserved transcription factors that are critical during embryonic development. Although the function of Hox genes is to pattern the embryo from head to tail, the mechanism by which Hox genes do this is unknown. Elucidating the mechanism for Hox gene function can provide insights into how morphologies are formed in a variety of organisms. I am investigating the role of Hoxd13 in patterning the colon in the developing chick. The Musculus complexus (pipping muscle) plays a primary role in the hatching of the chick from the eggshell at the end of its embryonic life. Various metabolic and cellular changes have been associated with the pipping muscle during development. These studies profiled the pipping muscle at the molecular level by identifying proteins which are associated with the developmental changes. In the first phase of the study, protein expression profile of the Day 13 chicken embryo pipping muscle was obtained using DDF with nano HPLC mass spectrometric analyzer. Gastrulation is a fundamental process that reorganizes the primary germ layers to shape the internal and external features of an early embryo. Morphogenetic movements underlying this process can be classified into a variety of different types of cellular movements. I will focus on investigating in this thesis two types of cell movements in the dorsal mesoderm; mediolateral cell intercalation and convergence and extension. During gastrulation, mesoderm cells send protrusions to gain traction on neighboring cells and the surrounding extracellular matrix; a process called mediolateral cell intercalation. Equine embryo cryopreservation is unsuccessful in larger, more easily collected, day-7 embryos. It is imperative that methods to successfully cryopreserve large equine embryos or develop reliable methods to determine embryo size before collection. Therefore the objectives for this study were to quantify the amount of tritiated glycerol that would permeate various sizes of equine embryos and to determine if circulating progesterone concentration was correlated with in utero embryo size. Mean embryo diameter (± SEM) across treatments (1.4M and 3.4M tritiated glycerol) was 696.5µm ± 108.6µm and 925.9 µm ± 214.1µm, respectively and were not different (P=0.44). The Gulf killifish (Fundulus grandis) is a topminnow native to the tidal marshes of the Gulf of Mexico. The species is prized by anglers as effective bait for catching popular sportfish and is of interest to aquaculturists and bait dealers due to its hardy nature. Limited seasonal availability and aquaculture of this species due to low fecundity and larval cannibalism of fry has led to increased incentive to improve cultural techniques.
