Hsp 72 In Rat Skeletal Muscle: Fibre Type Differences And Effect Of Exercise, Marius Locke

Western blot analyses of SDS-PAGE separated proteins from various hindlimb muscles from non-exercised control animals probed with an HSP 72 monoclonal antibody, demonstrated the constitutive expression of HSP 72 in rat muscles comprised primarily of Type I muscle fibres (soleus), but not in muscles comprised primarily of Type IIB/X fibres (white gastrocnemius-WG). In muscles of mixed fibre type, (plantaris and red gastrocnemius-RG) HSP 72 content was proportional to the percentage of Type I fibres.;Immunolocalization of HSP 72 in cross-sections from the soleus muscle demonstrated that both Type I and Type IIA fibres expressed HSP 72, but Type I fibres expressed …

Monoclonal antibodies (mAbs), raised against whole, fixed, uncoated, culture forms of Trypanosoma (Duttonella) vivax, were used to identify two invariant membrane proteins of this protozoan parasite. Since non-variant membrane proteins of the cell surface, flagellar pocket and endocytic pathway are potential targets for the control of trypanosomiasis of livestock by immunization, the identification and characterization of invariant membrane proteins is a necessary preliminary step.;A 65 kDA invariant membrane glycoprotein (gp65), identified using mAb 4E1, was the main focus of this study. Sex-hormone binding globulin (SHBG) and corticosteroid binding globulin (CBG) are the major plasma transport proteins for sex-steroids and glucocorticoids, respectively, and may impact directly upon steroid hormone bioavailability to target cells. Androgen binding protein (ABP) is closely related to SHBG and is believed to maintain an androgenic environment for sperm maturation. The isolation of SHBG-related cDNAs from human testis and a corresponding transcription unit indicates that SHBG and ABP are encoded by the same gene. In addition, the SHBG/ABP gene gives rise to two novel testicular transcripts through a process of alternative splicing. The putative proteins encoded by these transcripts …

Pseudomonas syringae pv. tomato is the causal agent of bacterial speck on tomato. An essential feature of this organism, in the pathogenic process, is its ability to enter the intercellular spaces of its host and use the available plant substrates for growth. Tomato leaves contain quantities of carbon energy in the form of carboxylates. A Tn5 mutant, DC3481, cannot utilize carboxylates as a source of carbon energy for growth (CA{dollar}\sp-{dollar}) and is nonpathogenic (Path{dollar}\sp-{dollar}) on tomato. Convict cichlids (Cichlasoma nigrofasciatum) form monogamous pair bonds and have cooperative biparental care of their eggs, larvae and fry for up to six weeks. This fish is widely used in laboratory studies as a model of biparental care. However, very little is known of its habits in its natural habitat. I studied four natural populations of convict cichlids, in 'pool' and 'stream' habitat, in Costa Rica during the long dry season, December to June in 1990 and 1991. The number of pairs guarding broods increased from December to a peak in mid-March, then rapidly declined. Twenty-eight percent of 175 males …

Gap junctional communication in preimplantation mouse embryos was explored by investigating: (1) the expression pattern of the connexin (Cx: gap junction protein) multigene family in maturing mouse oocytes and preimplantation embryos; (2) the relative role of Cx43 in gap junctional communication in embryos; and (3) the control of the acquisition of gap junctional coupling.;Both Cx43 and Cx32 transcripts were detected in maturing oocytes but these transcripts declined to undetectable levels shortly after ovulation. Cx43 transcripts were detected again at the 4-cell stage and accumulated steadily thereafter to reach a maximum in blastocysts. Cx26 and Cx46 transcripts could not be detected …

Treatment of Lewis rats with intracardiac injections of neuroantigen-containing liposomes significantly reduced the clinical severity of spinal cord-induced experimental allergic/autoimmune encephalomyelitis (EAE). Liposomes containing myelin basic protein (MBP), galactocerebroside (GC), or MBP + GC were effective in this respect, while liposomes containing proteolipid protein were not. The best protection was afforded by liposomes composed of whole myelin, as these ameliorated both the acute and relapsing phases of the disease, with a concomitant reduction in CNS pathology.;Neuroantigen-specific proliferative responses of lymphnode cells (LNC) taken from liposome-treated rats were markedly attenuated in comparison to control LNC, while delayed type hypersensitivity (DTH) reactions …

This thesis examines the determinants of childbearing among young Gambian women aged 13 to 24 and ranks the respective components (or groups of variables) in order of importance. An empirical analysis is done using bivariate and multi-variate statistical techniques. Based on these analyses, it is found that the factors of the empirical model (combining the reference group theory and a Bongaarts-type framework) vary in their effect on the fertility of young women. This study presents an analysis of demographic, socio-economic and environmental characteristics that may influence the quantum and timing of first, second and third births among Iranian women. This study focuses on the role that these variables played in influencing the fertility behaviour before, during and after the Islamic Revolution. The birth history data gathered by the 1988 Pars Province Growth Rate Survey-Iran are used in this study. An in-depth fertility questionnaire permits comparisons before and after the Islamic Revolution. The sample contains 2511 ever married women aged 15 years and above. Direct synaptic contact between substance P (SP)-immunoreactive terminals and glutamic acid decarboxylase (GAD)-positive neurons was more often observed in the substantia nigra pars lateralis (SNL) than in the substantia nigra pars reticulata (SNR) and was rarely observed in the substantia nigra pars compacta (SNC). Substance P terminals also formed synapses with cell bodies and dendrites of tyrosine hydroxylase (TH)-positive, dopaminergic neurons in the SNC and SNR. Multiple SP terminals were often observed with symmetrical and, less frequently, asymmetrical synapses on individual TH-containing dendrites. Control of tonic firing of sympathetic nerves by the rostral ventrolateral medulla (RVLM) and the pontine reticular formation (PRF) was investigated. Electrical activity of renal, splenic, mesenteric and preganglionic greater splanchnic nerves and T{dollar}\sb{lcub}13{rcub}{dollar} white rami was recorded in artificially respired, urethane-anaesthetized rats. Glycine (1.0M;180nl) was microinjected into the RVLM to inhibit neuronal discharge and effects on these nerves were compared. To study descending inhibitory influences on these nerves, effects of RVLM blockade were compared to effects of cervical spinal cord transection. RVLM blockade caused greater decreases in discharge of renal ({dollar}-44\ \pm\ 3\%{dollar}) than splenic ({dollar}-31\ \pm\ 2\%{dollar}) nerves, …
