Kaposi’s sarcoma-associated herpesvirus (KSHV) is considered the etiologic agent of Kaposi’s sarcoma and several lymphoproliferative disorders. Recently, the full-length KSHV genome has been cloned into a bacterial artificial chromosome (BAC) and successfully recovered in 293 cells. The herpesviral glycoproteins are structural components of the KSHV particle and are thought to facilitate virus entry, egress and virus-induced cell fusion. Investigations, described herein, have focused on the genetic manipulation of the KSHV-BAC36 in order to address the role of K8.1 glycoprotein in virus entry and the role of the carboxyl tail a-helices of glycoprotein (gB) in virus-induced cell fusion. One of the defining features of mental retardation is a problem in adaptive functioning. An area of adaptive skills commonly deficient in this population is social functioning, often characterized as behaviors that provide individuals with the means to interact effectively and appropriately with others. Researchers in this area have generally focused on improving appropriate social behavior and/or decreasing behavioral excesses that interfere with social interactions. Pore geometry and topology are important determinants of sediment physical properties, such as porosity and permeability. They also influence processes that occur in the sediment, such as acoustic propagation, attenuation, and dispersion, single- and multi-phase fluid flow, and hydrodynamic dispersion. This study uses images to evaluate pore geometry and topology of ooid (subspherical particles) and siliclastic (angular quartz) sand that was collected from the marine environment south of Bimni Bahamas and Ft. Walton Beach, FL, respectively. Behaviors that are appropriate, inappropriate, or a combination of both can be linked together in a behavior chain. When the contingencies for one response change (e.g., reinforcement is withheld for the last response in the chain; the last response is reinforced even if it occurs without the other responses in the chain) or the environmental conditions change (e.g., access to the reinforcer is available independent of responding), the other responses in the chain may be affected. This study investigated if accounting graduates from Louisiana State University, a public institution, perceived that their accounting curriculum enhanced the development of their personal competencies of professional demeanor, problem solving/decision making, interaction, leadership, and communication. A survey mailed to accounting graduates who graduated from Fall 1999 to Spring 2002 was used to gather the data for the study. Responses from graduates with different curricular paths were compared to determine if significant differences existed in their perceptions of the development of their personal competencies. This study will analyze the relationship between myth and the fantasy rhetoric of charismatic leadership by employing Fantasy Theme Analysis to examine the pervasive discourses invoking this enduring folk belief. Fantasies of the Charismatic Superhero are explored within the popular leadership treatises of successful “management gurus” and in our popular culture entertainments. The rhetorical visions of Stephen Covey’s “Principle-Centered Leadership,” Jim Collins’ “Level 5 Leadership,” and Manz and Sims' “SuperLeadership” are examined for their displacement of charismatic leadership in favor of the empowered crypto-charisma of self-leading memberships. The purpose of this qualitative study was to provide an examination of beliefs, context factors, and practices of exemplary teachers that lead to a technology-enriched curriculum. Three middle school teachers participated. Using both direct and participant observation the Spradley model was followed with three rounds of observations: (1) descriptive, (2) focused, and (3) selective. Interviews were conducted with open-ended questions and documents were collected from the parish website. This content analysis study examined 36 plant-centered children’s science picture books that have a plot and characters published from 1950 to present. Botanical subject matter and learning opportunities offered by these books were analyzed, along with the range and frequency of the National Science Education Standards-consistent and age-appropriate plant science concepts and principles. Turbine blade tip leakage flow from blade pressure side to suction side over the tip surface increases the thermal loading to the blade tip, leading to a high local temperature and thus, is considered one of the primary sources of blade failure. Leakage flow can be reduced by using a recessed or squealer tip blade or by cooling the blade tip to incorporate film cooling. Little attention has been given to estimating dynamic travel demand in transportation planning in the past. However, when factors influencing travel are changing significantly over time – such as with an approaching hurricane - dynamic demand and the resulting variation in traffic flow on the network become important. In this study, dynamic travel demand models for hurricane evacuation were developed with two methodologies: survival analysis and sequential choice model. The objectives of these experiments were to determine the effect of an early low dose administration of P4 or altrenogest (ALT) on pregnancy rates in repeat breeder (RPB) females, if an increase in pregnancy rates could result from a direct effect of P4 on the embryo and if ALT could support pregnancy in the absence of a functional CL. The four-parameter kappa distribution (Hosking 1994) was analyzed with respect to the various possible shapes of the probability density function. The general form for the cumulative distribution function when both h and k are non-zero is: F(x) = { 1 - h [ 1 - k ( x - ξ ) / α ]1/k }1/h. DNA binding of the DNA polymerase I from Thermus aquaticus (Taq and Klentaq fragment) and E. coli (Klenow fragment) have been studied as a function of [salt] and temperature in order to understand their DNA binding thermodynamics. Binding of the two different species of polymerases occurs with sub-micromolar affinities in very different salt concentration ranges. Thus, at similar [KCl] the binding of Klenow is ~ 3kcal/mol (150X) tighter than the binding of Taq/Klentaq. Linkage analysis of the [KCl] dependence of DNA binding at 25°C reveals a net release of 2-3 ions for Taq/Klentaq and 4-5 ions for Klenow. DNA binding …

Since Ashford and Cummings' (1983) seminal article, situational and individual difference variables have predominantly been examined to predict how often employees will seek feedback. Sodium undecenoyl-L-Leucinate (L-SUL) was synthesized and modified by use of alkali metal counterions, alcohols, and Triton X-102. Characterization of the modified poly-L-SUL was performed in order to elucidate the structural changes on the surfactants by use of four techniques. Two new methods for the efficient parallel computation of the unsteady incompressible Navier-Stokes equations are presented. Such efficient methods are desired for large scale parallel computations of unsteady turbulent flows such as Direct Numerical Simulations (DNS). The performance of the new methods has a distinct advantage over the artificial compressibility method, in that the methods exhibit robust convergence for a variety of flow problems without extensive need for tuning computational parameters. This investigation examined the effects of age, exercise and test condition on hemodynamic variables, autonomic and vascular function in relation to resistance exercise (RE). The associations among these variables were also examined. METHODS: Sixteen young (21.4+1.4 yrs) and 16 older (69.7+3.9 yrs) women performed 5- and 15- repetition maximal (RM) of knee extension RE. This work demonstrates peptide-based aggregation mediation of fibril forming peptides, notably β-amyloid (1-40), Aβ1-40. To achieve the goals of this work, Aβ1-40 aggregation was governed by ionic strength, temperature, and pH. After controlling and reproducing Aβ1-40 aggregate formation (fibrils), each peptide-based aggregation mediator was evaluated for its ability to self-aggregate and to possess surface-active properties. The incorporation of these mediators in Aβ1-40 solutions changed the surface tension and produced aggregate complexes of mediator and Aβ1-40. Studies were conducted to test the hypothesis, significant benefits can be achieved for the management of rice water weevil, Lissorhoptrus oryzophilus Kuschel, by the use of rice varieties with low levels of host plant resistance coupled with the judicious use of cultural practices and insecticides. Tolerant traits of a current commercial variety of rice, "Bengal", was manifested consistently over multiple years. The purpose of this study was to identify elements present in effective literacy instruction, i.e., those elements that fostered reading, speaking, listening, thinking, writing, and metacognitive skills. By examining variables that promoted literacy acquisition and achievement, a detailed account resulted which described classroom practice as related to teacher beliefs, strategies, materials, and classroom climate. The Histone-like protein HU is ubiquitous in eubacteria. Usually with a length of ~90 amino acids, they are predominantly homodimeric, with sequence and structural homology. Escherichia coli HU is involved in DNA repair and recombination. The crystal structure of Anabaena HU shows that it binds DNA with prolines intercalating into the DNA backbone, introducing two kinks at a spacing of 9 bp and bending the DNA through a variable angle of 105-140°. We present results from investigations of the nonlinear development of the secular bar-mode instability that is driven by gravitational radiation-reaction (GRR) forces in rotating neutron stars. Our fully three-dimensional hydrodynamical simulations have shown that, in the absence of any competing viscous effects, initially uniformly rotating axisymmetric n=1/2 polytropic stars with a ratio of rotational to gravitational potential energy greater than the critical limit are driven by GRR forces to a bar-like structure, as predicted by linear theory. Foreland basin subsidence through time is reproduced in this study, as the flexure of an elastic beam in an inviscid fluid under the vertical stress, caused by discrete-distributed loads. Thus, seismostratigraphic data from the Timor Sea peripheral foreland basin, in northwestern Australia, and the Putumayo retroarc foreland basin in the Colombian Andes, are forward modeled, at chronostratigraphic intervals, to assess the evolving geodynamic conditions of the basins. Results show that the accommodation in foreland basins varies as the depositional basement is vertically adjusted according to the regionally isostatic compensation of the lithosphere. Since supernatural elements are unknown and unfamiliar, they easily arouse anxiety, fear, and even result in terror. Recently an efficient methodology, a hetero Pauson-Khand reaction, based on titanium-catalyzed cyclocarbonylation of tethered enals for the general preparation of ¥ã-butyrolactone rings, which are typically embedded in polycyclic systems of many natural products was reported. To demonstrate this new strategy, the total syntheses of the natural products asteriscanolide and ginkgolide were investigated. The first part of this work is dedicated to synthetic efforts toward the total synthesis of asteriscanolide. Approaches highlighted by the [2,3]-Wittig rearrangement, the thermal silyloxy-Cope rearrangement, and a titanium-catalyzed cyclocarbonylation, which is the pivotal step to afford the ¥ã-butyrolactone ring.
