Research was conducted to develop methods for improving rice through somaclonal variation using plant tissue culture techniques. Experiments were directed toward developing an efficient culture system by which plants could be efficiently regenerated in rice somaculture. Histological studies of the process of callus formation and plant regeneration were also carried out. Explant sources evaluated for their potential for callus induction and plant regeneration included seedling mesocotyl, root-tips, embryos from mature seeds, nodes, immature panicles, and mature panicles. The thermal death time of Clostridium botulinum type E spores in crawfish tailmeat was determined at temperatures ranging from 80 to 95(DEGREES)C. Crawfish paste containing 10('6) spores per gram was dispensed into 16 x 125 mm test tubes, which were subsequently placed in water baths preset at the test temperatures. Sample tubes were removed from the baths at predetermined time intervals and incubated anaerobically for up to two months. The D value for the spores in crawfish tailmeat was determined from the corrected time of exposure to the heat and survival data using three methods of calculation. D values were …

Three different treatments of salt 6, 8 and 10% were used in the process of aging hot pepper mash in oak and plastic barrels over a period of two years. The chemical characteristics of the changes during the aging process of these mashes were determined. These characteristics included pH, titratable acidity, salt content, color changes and total pectic substances. The pectic substances were degraded by the hot pepper pectic enzymes very rapidly after only a few days of aging. This degradation could be retarded by increasing the salt content of the mash or by aging in oak barrels. However, the …

This research was conducted to examine the nature of the difference in resistance to blast under different water-management systems. Two cultivars; M-201, susceptible, and Brazos, partially resistant to blast, grown in flooded and upland (unflooded) field plots were inoculated with race IH-1 of Pyricularia oryzae in 1984 and 1985. Final disease ratings on M-201 and Brazos averaged 77.6% and 39.8% under upland conditions and 1.1% and 0.1% under flooded conditions, respectively. When blasted plants in upland plots were flooded, the rate of blast development decreased on Brazos but changed little on M-201. No increase in disease was observed when flooded …

Heritability and other genetic effects for the date of first flowering, date of maximum flowering, average flowering date, percent flowering, pith and brix in sugarcane were estimated in a diallel cross experiment. A three year study indicated that the date of first flowering, date of maximum flowering, average flowering date, and percent flowering were highly heritable characters. The heritability estimates of pith and brix were very low and variable. Partitioning of total direct response in parental lines into average direct and average maternal genetic effects indicated that average maternal genetic effects of the early and late lines were larger in …

Diplodia gossypina was only isolated from the basal portion of sweet potato sprouts growing from mother roots which were bedded adjacent to diseased roots, bedded in infested soil or inoculated with conidial suspension before bedding. It was recovered 1-2 cm above and 2-4 cm below the point of inoculation 2 months after inoculation of wounded established vines. It was not isolated from the proximal or distal ends of roots harvested from field plots in which vine cuttings or established vines were artificially inoculated or when vine cuttings were planted in artificially infested soil. Isolates of Cercospora oryzae were readily obtained by incubating pieces of surface sterilized diseased leaf or leaf sheath samples in a moist chamber at 28 C. Sporulation on culture media was significantly higher following incubation at 28 C on cornmeal agar than on water agar, V-8 juice agar, and potato-dextrose agar. Narrow brown leaf spot lesions developed on leaves when inoculated plants were placed inside a humidified chamber for 1 to 16 days. Whole cottonseed extruded at temperatures ((DEGREES)C) and rates (kg/hour) of 131, 314; 135, 182; 146, 195; and 156, 286 was evaluated for protein solubility, in vitro ammonia production, and in situ disappearance to estimate bypass protein potential. Whole cottonseed, soybean meal, corn gluten meal, and soybean meal and whole cottonseed heated for 4 hours at 127, 138, and 149(DEGREES)C were also evaluated for comparison. Solubility was determined by three methods: hot water-refluxed 1 hour; cold water-mixed 30 minutes; and cold water-homogenized 5 seconds. This study was conducted to identify those plant growth regulators that cause the greatest increase in shoot number of basic genotypes of the genus Saccharum and at the same time identify those genotypes that exhibit a high degree of response to certain growth regulators which have been proposed as tillering agents. A series of three factorial experiments was conducted under controlled conditions in the greenhouse utilizing fourteen cultivars representing the spectrum of the genus Saccharum along with five synthetic growth regulators proposed as tillering agents. Five Thailand soils belonging to the Rangsit Very Acid (Rsa), Rangsit (Rs), Mahaphot (Ma), Bang Pakong (Bg), and Bangkok (Bk) series were employed in redox-pH-ion chemistry studies. Decreases in soil redox potential upon submergence were relatively slower and less intense in the acid sulfate soils than in the non-acid marine soil. Among the acid sulfate soils, the Rsa soil was most sluggish with respect to decreases in redox potential. A series of studies were conducted from 1983-1985 under greenhouse, field, and laboratory conditions. Activities of asulam methyl (4-aminophenyl)sulfonyl carbamate , dalapon (2,2-dichloropropionic acid) and asulam/dalapon combinations on itchgrass (Rottboellia exaltata L.f.) and johnsongrass Sorghum halepense (L.) Pers. were examined. Combinations consisted of tank mixes and sequential applications. Visual evaluations were taken 3 and 6 weeks after treatment (WAT) in 1983 and 4 and 8 WAT in 1984. Twelve WAT plant height, stem number, seedhead number and biomass were measured. Visual evaluation results paralleled quantitative measurements. Planting soybeans between early May and early June provides the highest yield. Planting either before or after this optimal time can significantly reduce yields. Most of the yield from determinate soybeans comes from the branches. Branch development and yield are reduced at nonoptimal planting dates, while main axis yield is not. The relatively short photoperiods of nonoptimal planting dates appears to cause the reduction of both the vegetative and reproductive growth periods resulting in reduced development and yield. Hemarthrosis in rhesus monkeys was studied to: (1) provide information regarding the possible relationship between traumatic hemarthrosis and naturally occurring calcium pyrophosphate dihydrate-deposition disease (CPPD-DD) in rhesus monkeys, (2) fill a void in the literature regarding experimental hemarthrosis in primates, (3) and study the early changes that occur in blood-induced cartilage destruction by adding biochemical, metabolic and morphometric analyses to the traditional morphologic evaluations. Three ml of autologous blood were injected into the left knee of 16 anesthetized monkeys; the right knee was an untreated control. A sensitive, rapid, and accurate method for extraction and analysis of gossypol in bovine plasma and tissue by high-performance liquid chromatography was developed. A three day old Guernsey bull calf absorbed gossypol after single oral dose of gossypol acetic acid in corn oil (5 mg/kg of body weight). Maximum level of plasma gossypol (5.5 ug/ml) was absorbed at 6 hours after treatment. Average gossypol recoveries from plasma and tissue samples spiked before extraction were 88.2% and 82.9%, respectively. Calli derived from 'Tangi' (susceptible) and MSUS 42 and LA 8318 (resistant) exhibited differential responses when inoculated with Colletotrichum fragariae, isolate CF1. The best differential in terms of growth and multiplication of the fungus was observed at 15(DEGREES)C and this differential decreased with increasing incubation temperature and time. Level of benzyladenine in the medium did not affect disease expression. Susceptible tissues exhibited extreme sensitivity, characterized by intense discoloration and cessation of growth, when subcultured onto a medium containing the culture filtrate of C. fragariae while the resistant tissues were affected less. Raw, broiled and fried beef liver samples were stored at -20(DEGREES)C in either vacuum or air-saturated packages. At periods of 30, 60 and 90 days the folates were extracted and analyzed by reverse phase high performance liquid chromatography (HPLC). Tetrahydrofolic acid (THF), dihydrofolic acid (DHF), 5-CH(,3)-THF, 5-CHO-THF and pteroylglutamic acid (PGA) were identified and quantitated. Two major folate oxidation products, pterin-6-carboxylic acid (pt-6-COOH) and p-aminobenzoylglutamic acid (p-ABG) were also detected. The total folate content of beef liver was higher than previously reported. PGA was not found in any of the samples but a large peak corresponding to the retention time …

Three cultivars of Lilium longiflorum, Thunb., 'Ace', 'Nellie White' and 'Harson' were forced for the spring of 1983 and 1984 in greenhouse sections with minimum temperatures of 10, 14.5 or 18.9(DEGREES)C until 5 weeks after emergence (phase 1). Plants from each temperature treatment were then divided equally among the 3 temperature regimes (phase 2) to give 9 temperature histories. The design was based on results from a preliminary experiment in 1982. Fewer flowers, fewer leaves and shorter terminal height were associated with higher minimum temperatures. Flower and leaf number were most affected by early (phase 1) temperature treatment. 'Ace' produced …

As part of a chemical-ecological study of the Florida scrub community, Ceratiola ericoides (Empetraceae) and Calamintha ashei (Lamiaceae) were chemically investigated for natural products with possible allelochemical activities. The chromatographic separations and chemical investigations were guided by germination and radicle growth bioassays performed on seeds of commercial lettuce (Lactuca sativa) and Schizachyrium scoparium, a native grass of the Florida sandhill community. From the dichloromethane extract of aerial parts of C. ericoides, 2', 4'-dihydroxychalcone, the dihydrochalcones angoletin and 2',6'-dihydroxy-4-methoxy-3',5'-dimethyldihydrochalcone, the flavanones 8-methylpinocembrin, 6,8-dimethylpinocembrin and 7-hydroxyflavanone were isolated along with the triterpenes erytrodiol and ursolic acid. One Landsat Thematic Mapper (TM), and two Multispectral Scanner (MSS) data sets were digitally analyzed for forest stand and type mapping of the Kisatchie Ranger District, Kisatchie National Forest, Louisiana. Detailed ground-verification maps were produced from interpretation of 1:12,000 and 1:58,000 color-infrared (CIR) aerial photography of nine test compartments in the study area. Stand boundary and soils maps of the study area were obtained and input to the digital geographic information system along with the satellite and ground-verification data.
