The Redox State Of The [2fe-2s] Clusters In Soxr Protein Regulates Its Activity As A Transcription Factor, Huangen Ding, Elena Hidalgo, Bruce Demple

SoxR protein is a redox-responsive transcription factor that governs a regulon of oxidative stress and antibiotic resistance genes in Escherichia coli. Purified SoxR contains oxidized [2Fe-2S] clusters and stimulates in vitro transcription of its target gene soxS up to 100-fold. SoxR transcriptional activity, but not DNA binding, is completely dependent on the [2Fe-2S] clusters; apo-SoxR prepared in vitro binds the soxS promoter with unchanged affinity but does not have transcription activity. Thus, modulation of the SoxR [2Fe-2S] clusters was proposed to control the protein's function in transcription. Here, we provide evidence that SoxR with reduced [2Fe-2S] clusters is inactive. We report performance measurements of a sub-millimeter resolution CdZnTe strip detector developed as a prototype for astronomical instruments operating with good efficiency in the 30-300 keV photon energy range. The prototype is a 1.4 mm thick, 64 × 64 contact stripe CdZnTe array of 0.375 mm pitch in both dimensions. Pulse height spectra were recorded in orthogonal-stripe coincidence mode which demonstrate room-temperature energy resolution < 10 keV (FWHM) for 122 keV photons with a peak-to-valley ratio > 5:1. Good response is also demonstrated at higher energies using a coplanar grid readout configuration. Spatial resolution capabilities finer than the stripe pitch are demonstrated. Wetland research in northern Belize provides the earliest evidence for development of agriculture in the Maya Lowlands. Pollen data confirm the introduction of maize and manioc before 3000 B.C. Dramatic deforestation, beginning ca. 2500 B.C. and intensifying in wetland environments ca. 1500-1300 B.C., marks an expansion of agriculture, which occurred in the context of a mixed foraging economy. By 1000 B.C. a rise in groundwater levels led farmers to construct drainage ditches coeval with the emergence of Maya complex society ca. 1000-400 B.C. Measurements of NO, NO2, and total reactive oxidized nitrogen (NOy) were added to ongoing measurements of aerosols, CO, and O3 at Sable Island (43°55′N, 60°01′W), Nova Scotia, during the North Atlantic Regional Experiment (NARE) 1993 summer intensive. Ambient levels of NOx and NOy were found to be highly variable, and elevated levels can be attributed to the transport of polluted continental air or presumably to relatively fresh emissions from sources upwind (e.g., ship traffic). The median values for NOx and NOy are 98 and 266 parts per trillion by volume (pptv), respectively. A multiday pollution episode occurred during which elevated …

Multifragment decays of central collisions in 84Kr+197Au at E/A=70MeV are studied. By utilizing a technique sensitive to the emission order of fragments, it is deduced that carbon fragments are emitted prior to beryllium fragments when these fragments have the same velocity. This observation is consistent with the cooling of a thermally decaying source.

Two critical steps in the assembly of yeast and mammalian glycosylphosphatidylinositol (GPI) anchor precursors are palmitoylation of the inositol residue and mannosylation of the glucosamine residue of the glucosaminyl phosphatidylinositol (GlcNα-PI) intermediate. Palmitoylation has been reported to be acyl-CoA dependent in yeast membranes (Costello, L. C., and Orlean, P. (1992) J. Biol. Chem. 267, 8599-8603) but strictly acyl- CoA independent in rodent membranes (Stevens, V. L., and Zhang, H. (1994) J. Biol. Chem. 269, 31397-31403), and thus poorly conserved. Within the framework of classical molecular dynamics, we study the collision Au+Au at an incident energy of 35 MeV/nucleon. It is found that the system shows a critical behavior at peripheral impact parameters, revealed through the analysis of conditional moments of charge distributions, Campi scatter plot, and the occurrence of large fluctuations in the region of the Campi plot where this critical behavior is expected. When applying the experimental filters of the MULTICS-MINIBALL apparatus, it is found that criticality signals can be hidden due to the inefficiency of the experimental apparatus. Energy dispersive spectrometry line scan and ac impedance spectroscopy were used in this study to investigate the chemical reactions between two cathode materials, La_0.84Sr_0.16MnO₃ (LSM), La_0.5Sr_0.5CoO_3-δ (LSC), and the electrolyte La_0.9Sr_0.1Ga_0.8Mg_0.2O_2.85 (LSGM). Significant interdiffusions of Co into LSGM and Ga into LSC were found at an LSC/LSGM interface even at relatively low fabrication temperatures. In contrast, only small interdiffusions of Mn into LSGM and Ga into LSM were detected at the LSM/LSGM interface even though it was fired at 1470°C. The …

Sidewards directed fragment flow has been extracted for 84Kr+197Au collisions at E/A=200MeV, using techniques that are free of reaction plane dispersion. The fragment flow per nucleon increases with mass, following a thermal or coalescencelike behavior, and attains roughly constant limiting values at 4≤A≤12. Comparisons of the impact parameter dependences of the measured coalescence-invariant proton flow to Boltzmann-Uehling-Uhlenbeck calculations clearly favor a momentum dependent nuclear mean field.

A major revision of Article 8 of the Uniform Commercial Code was passed by the New York Assembly on July 2, 1996, but did not receive Senate consideration prior to adjustment. In light of the strong support given Proposed Article 8 by the banking and securities industries, the New York State Legislature will probably give serious consideration to passage when the next legislative session begins.

Managed-care advocates praise its cost controls on treatments for beginning to tame the health care beast, which devoured nearly 14 percent of the nation's gross domestic product in 1994, according to the U.S. Department of Health and. Human Services. Such belt-tightening is necessary to allocate health care dollars rationally, advocates contend, pointing to direct patient costs as proof that a healthy balance has been achieved. But critics contend that managed care is more about making money than saving it. Even though costs have gone down, they argue, premiums have remained high and corporate profits have soared. In marine soft-sediment habitats, the sediment surface is altered by activities of sediment dwellers (infauna). Such biogenic disturbance can influence recruitment success if settling larvae and juveniles avoid disturbed sites or if juveniles die as a result of disturbance after settling. Because infauna commonly lose exposed body parts to browsing predators and disturb less sediment as a result, we developed a simulation model to examine the interactions between browsing predation, infaunal adult activity, and recruitment. Sediment disturbance in the model was based on data for the polychaete Abarenicola pacifica. We simulated the activity of two general types of predators: prey …

A new, sensitive method allows one to search for the enhancement of events with nearly equal-sized fragments as predicted by theoretical calculations based on volume or surface instabilities. Simulations have been performed to investigate the sensitivity of the procedure. Experimentally, charge correlations of intermediate mass fragments emitted from heavy ion reactions at intermediate energies have been studied. No evidence for a preferred breakup into equal-sized fragments has been found.

Continuous measurements of SO42− and electrical conductivity (ECM) along the GISP2 ice core record the Toba mega‐eruption at a depth 2590.95 to 2091.25 m (71,000±5000 years ago). Major chemical species were analyzed at a resolution of 1 cm per sample for this section. An ∼6‐year long period with extremely high volcanic SO42− coincident with a 94% depletion of nitrate and 63% depletion of chloride is observed at the depth of the Toba horizon. Such a reduction of chloride in a volcanic layer preserved in an ice core has not been observed in any previous studies. The nearly complete depletion of …

A comprehensive graphical modeling environment has been developed to address the needs of the computer simulation of groundwater flow and transport. The Department of Defense Groundwater Modeling Systems (GMS), developed at the Engineering Computer Graphics Laboratory at Brigham Young University, is part of a multi-year project funded through the Department of Defense, Department of Energy and Environmental Protection Agency. GMS is a graphically based software tool providing facility through all aspects of the groundwater flow and transport modeling process. We show how information about the elastic stiffness and compliance of an orthotropic polycrystal may be obtained from measurements of the statistical properties of the microstructure. We begin by discussing the statistical properties of the governing equations and the hierarchy that results when the equations are averaged. Perturbation solutions are obtained in terms of low-order statistical information. Using methods previously developed, we derive bounds for the elastic stiffnesses and compliances. A heirarchy of bounds is derived using the statistical information obtained from measurements of the microstructure. We discuss the methods used to obtain the correlation functions of the microstructure and …

While stories of Idaho's Mormon polygamists' capture and trial are well known, the prison experiences of these men are not. Since prison overcrowding was a nineteenth-century problem too, they served their sentences in several states. Many kept diaries or journals, and from these personal accounts we can learn a good deal about their life in prison.
