Magnetic resonance imaging (MRI) is one of the non-invasive imaging modalities used in longitudinal cell tracking. Previous studies suggest that MagA, a putative iron transport protein from magnetotactic bacteria, is a useful gene-based magnetic resonance contrast agent. Hemagglutinin (HA)-tagged MagA was stably expressed in undifferentiated embryonic mouse teratocarcinoma, multipotent P19 stem cells to provide a suitable model for tracking these cells during differentiation. Western blot and immunocytochemistry confirmed the expression and membrane localization of MagA-HA in P19 cells. Many vascular and cardiac diseases are diagnosed and treated using a medical technique known as percutaneous transluminal catheter intervention (PTC). In PTC, the interventionalist inserts a catheter into the vasculature, and using the vessel as the guiding passageway, the catheter is navigated to desired anatomical targets where it would be used for various purposes such as catheter ablation for the treatment/management of cardiac arrhythmias. The catheterization procedure is conventionally guided with x-ray fluoroscopic imaging and more recently, but rarely, with Magnetic Resonance Imaging (MRI). Transcranial Doppler ultrasound (TCD) is used for the assessment of cerebral blood flow velocity (CBFV) at the middle cerebral artery (MCA) with the assumption that diameter of the artery does not change. Thus, CBFV is equivalent to cerebral blood flow (CBF). The purpose of this thesis was determine if the MCA dilates during hypercapnia (HC) and/or constricts during hypocapnia (HO) in healthy young and older adults using 3T magnetic resonance imaging (MRI). Development of methods to monitor cellular activity with magnetic resonance imaging (MRI) provides a powerful tool for studying disease progression and monitoring therapy. This thesis investigates components necessary for development of reporter gene expression for MRI, to track Nkx2.5 transcription factor activity during tumour growth and, in the future, to calibrate MR contrast against a well-known optical reporter gene, like firefly luciferase. In human MCF-7 breast cancer cells, Nkx2.5 is induced by all-trans retinoic acid (tRA) and activates a minimal promoter of the rat sodium iodide symporter (rNISmin) gene.
