Free Solution Electrophoresis Measurements And Their Theoretical Relationships To Net Protein Valence, Jennifer A. Durant Jan 2003

Free Solution Electrophoresis Measurements And Their Theoretical Relationships To Net Protein Valence, Jennifer A. Durant

Doctoral Dissertations

Biochemical solutions are composed to a large degree of charged molecules (proteins, DNA, dissolved salts, etc) which can interact and therefore exert a force on each other. Analytical determination of the valence of biological macro-ions contributes to our understanding of the forces involved in both intra- and inter-molecular interactions. This dissertation uses two free solution techniques, membrane confined electrophoresis (MCE) and capillary electrophoresis (CE), to study valence in the context of a series of charge mutants of T4 lysozyme and RNase Sa. The use of these mutants allows attribution of the changes in electrophoretic behavior entirely to changes in charge. …

Connections Between New Hampshire Superintendents' Leadership Orientations And Their Perceptions Of Selected Issues Associated With Politics In Education, Althea E. Sheaff Jan 2003

Connections Between New Hampshire Superintendents' Leadership Orientations And Their Perceptions Of Selected Issues Associated With Politics In Education, Althea E. Sheaff

Doctoral Dissertations

Many Americans, including educational professionals, eschew politics in classrooms, administrative offices, or any interaction with the educational system. The apolitical myth has it roots in the struggle for public schooling by common school crusaders like Horace Mann and Henry Barnard and continuing in the twentieth century with adoption of Frederick Taylor's scientific management principles. Despite commonly held notions of the separateness and distance between politics and education, this study concludes that politics in education is inevitable.

The purpose of the study is to uncover connections between superintendents' leadership orientations and their political behaviors and beliefs using a multiple perspectives approach. …

"Everybody Get Together": The Sixties Counterculture And Public Space, 1964--1967, Jill Katherine Silos Jan 2003

"Everybody Get Together": The Sixties Counterculture And Public Space, 1964--1967, Jill Katherine Silos

Doctoral Dissertations

Historians and cultural analysts have traditionally considered the sixties counterculture an apolitical phenomenon by historians and other analysts. Yet concentrated examination of the public activities of the counterculture in San Francisco from 1963 to 1967 reveals that they were engaged in the creation of a public political culture that challenged the power of civil authorities to regulate the uses of parks, streets and sidewalks. In doing so, the counterculture constituted a distinct community with a political agenda.

This thesis is demonstrated through an analysis of the development of an ethos toward public space in the Beat movement and Merry Prankster …

Hydrodynamic Behavior And Thermal Stability Of A Pegylated Protein: Studies With Hen Egg Lysozyme, Yatin R. Gokarn Jan 2003

Hydrodynamic Behavior And Thermal Stability Of A Pegylated Protein: Studies With Hen Egg Lysozyme, Yatin R. Gokarn

Doctoral Dissertations

We studied the effect of covalent attachment of polyethylene glycol (PEGylation) on the hydrodynamic behavior and thermal stability of a model protein---Hen Egg Lysozyme (HEL). HEL was modified with a linear, 20-kD, PEG to produce mono (PEG1-HEL), di (PEG2-HEL), and triPEGylated (PEG3-HEL) species.

The hydrodynamic properties of HEL were altered upon PEGylation. A decrease in sedimentation (s) and diffusion (D) coefficients was observed for all three PEG-HEL molecules in comparison to HEL (1.81 s). Despite differences in molecular weights of the PEG-HEL molecules (∼34, 55 and 80 kD), their s values were very close (1.0--1.1 s). Significant hydrodynamic non-ideality was …

Law Enforcement Dilemmas In The Investigation Of Internet Sex Crimes Against Minors, Melissa Wells Jan 2003

Law Enforcement Dilemmas In The Investigation Of Internet Sex Crimes Against Minors, Melissa Wells

Doctoral Dissertations

This study examines dilemmas associated with the investigation of Internet sex crimes against minors. Data collected from a national sample of law enforcement agents provide insights into complications of three types of cases: (1) Internet crimes against identified victims, (2) Internet solicitations to undercover law enforcement, and (3) Internet child pornography crimes.

A mail survey of United States law enforcement agencies identified two samples of Internet sex crimes against minors. The first sample consisted of 464 investigations in which an arrest was made, and the second sample included 68 cases in which no arrest was made. Telephone surveys were used …
