Understanding basic neuronal mechanisms hold the hope for future treatment of brain disease. The 1st international conference on synapse, memory, drug addiction and pain was held in beautiful downtown Toronto, Canada on August 21-23, 2006. Unlike other traditional conferences, this new meeting focused on three major aims: (1) to promote new and cutting edge research in neuroscience; (2) to encourage international information exchange and scientific collaborations; and (3) to provide a platform for active scientists to discuss new findings. It has been considered a "law" that a lost structure cannot reappear in evolution. The common explanation, that genes required for the development of the lost structure degrade by mutation, remains largely theoretical, however. Additionally, the extent to which this mechanism applies to systems of repeated parts, where individual modules are likely to exhibit few unique aspects of genetic control, is unclear. We investigated reversibility of evolution in one such system, the vertebrate dentition, using as a model loss of oral teeth in cypriniform fishes, which include the zebra fish. Background: Comparative genomics has become an essential approach for identifying homologous gene candidates and their functions, and for studying genome evolution. There are many tools available for genome comparisons. Unfortunately, most of them are not applicable for the identification of unique genes and the inference of phylogenetic relationships in a given set of genomes.

Results: GenomeBlast is a Web tool developed for comparative analysis of multiple small genomes. A new parameter called "coverage" was introduced and used along with sequence identity to evaluate global similarity between genes. Karyotypes of eight species of Leptodactylus (Anura, Leptodactylidae) with a description of a new karyotype for the genus. Eight species of the Neotropical genus Leptodactylus were karyologically studied: seven of them (L. gracilis, L. mystacinus, L. petersii, L. pustulatus, L. macrosternum, L. ocellatus, L. labyrinthicus) presented 2n=22 and L. silvanimbus showed a distinctive karyotype with 2n=24. Genetic Resolution Of The Enigmatic Lesser Antillean Distribution Of The Frog Leptodactylus Validus (Anura, Leptodactylidae), Keneth Yanek, W. R. Heyer, Rafael O. De Sá

Leptodactylus validus has an unusual distribution, inhabiting Trinidad, Tobago, and the Lesser Antilles, but not the mainland of South America. This distribution is inconsistent with other distribution patterns observed for these islands. Although slight variation in adult morphology has been observed among the different island populations of L. validus, call data suggest the presence of a single species. Calls of L. pallidirostris from Venezuela and Brazil suggested that this taxon might be conspecific with L. validus. Sequence data from the 12S and 16S mt rDNA genes indicate that L. validus represents a single species throughout its distribution and is conspecific …

Total and intra-mitochondrial gonadal cholesterol concentrations are decreased in fish exposed to the phytoestrogen beta-sitosterol (beta-sit). The present study examined the potential for beta-sit to disrupt de novo cholesterol synthesis in the gonads of goldfish exposed to 200 microgram/g beta-sit and 10 microgram/g 17beta-estradiol (E2; estrogenic control) by intra-peritoneal Silastic® implants for 21 days. The de novo cholesterol synthetic capacity was estimated by incubating gonadal tissue with 14C-acetate for a period of 18 hours, followed by chloroform/methanol lipid extraction and thin layer chromatography (TLC) lipid separation. MicroRNAs (miRNAs) are capable of controlling gene expression by targeting complimentary sequences in many mRNAs. Thus, a small number of miRNAs are capable of regulating expression of many different genes. miRNAs have been found in all animals from Drosophila to human and they are highly conserved. This work was undertaken in order to identify such RNAs in the newt eye.

Organogenesis involves an initial surge of cell proliferation, leading to differentiation. This is followed by cell death in order to remove extra cells. During early development, there is little or no cell death. However, there is a lack of information concerning the genes required for survival during the early cell-proliferation phase. Here, we show that Lobe (L) and the Notch (N) ligand Serrate (Ser), which are both involved in ventral eye growth, are required for cell survival in the early eye disc. Background: The Hippo tumor-suppressor pathway has emerged as a key signaling pathway that controls tissue size in Drosophila. Merlin, the Drosophila homolog of the human Neurofibromatosis type-2 (NF2) tumor-suppressor gene, and the related protein Expanded are the most upstream components of the Hippo pathway identified so far. However, components acting upstream of Expanded and Merlin, such as transmembrane receptors, have not yet been identified.

Results: Here, we report that the protocadherin Fat acts as an upstream component in the Hippo pathway. Fat is a known tumor-suppressor gene in Drosophila, and fat mutants have severely overgrown imaginal discs. We …

Recent field work and a review of catalogued specimens in the herpetology collections at the Yale Peabody has yielded 170 new town records for amphibians and reptiles in Connecticut. These are reported here, along with observations on the status of the red-ear slider (Trachemys scripta) in Connecticut.

We previously showed that the Bvg virulence control system regulates biofilm formation in Bordetella bronchiseptica (Y. Irie, S. Mattoo, and M. H. Yuk, J. Bacteriol. 186:5692-5698, 2004). Analyses of the extracellular components of B. bronchiseptica biofilm matrix revealed that the major sugar component in the matrix was xylose, and linkage analysis indicated a majority of it to be in a 4-linked polymeric form. The production of xylose was independent of Bvg regulation but instead was dependent on bacterial growth phase. In addition, N-acetyl-glucosamine in the matrix was found to be important for the initial development of the biofilm. These …

The purpose of this application, under Article 75.6 of the Code, is to conserve the usage of the specific name of Rana ocellata Linnaeus, 1758 for a species of leptodactylid frog from South America by the designation of a neotype. Prevailing usage of the name is threatened by the identity of the type specimen which is a different species than that which is currently known as Leptodactylus ocellatus. It is proposed that all name-bearing types be set aside and a neotype designated in accord with prevailing usage.

BACKGROUND: Because amphibians arise from a branch of the vertebrate evolutionary tree that is juxtaposed between fishes and amniotes, they provide important comparative perspective for reconstructing character changes that have occurred during vertebrate evolution. Here, we report the first comparative study of vertebrate genome structure that includes a representative amphibian. We used 491 transcribed sequences from a salamander (Ambystoma) genetic map and whole genome assemblies for human, mouse, rat, dog, chicken, zebrafish, and the freshwater pufferfish Tetraodon nigroviridis to compare gene orders and rearrangement rates.

The fossil record indicates that cypriniform fishes, a group including the zebrafish, lost oral teeth over 50 million years ago. Despite subsequent diversification of feeding modes, no cypriniform has regained oral teeth, suggesting the zebrafish as a model for studying the developmental genetic basis of evolutionary constraint. To investigate the mechanism of cypriniform tooth loss, we compared the oral expression of seven genes whose mammalian orthologs are involved in tooth initiation in the zebrafish and the Mexican tetra, Astyanax mexicanus, a related species retaining oral teeth. The MyoD family of basic helix-loop-helix transcription factors function as heterodimers with members of the E-protein family to induce myogenic gene activation. The E-protein HEB is alternatively spliced to generate alpha and beta isoforms. While the function of these molecules has been studied in other cell types, questions persist regarding the molecular functions of HEB proteins in skeletal muscle. Our data demonstrate that HEB alpha expression remains unchanged in both myoblasts and myotubes, whereas HEB beta is upregulated during the early phases of terminal differentiation. Red imported fire ant, Solenopsis invicta Buren, is a serious pest in the southeastern United States. It has caused economic losses in agricultural products, stings can cause anaphylactic shock, and it has reduced biodiversity. S. invicta has displaced native ant species throughout its range, presumably by competitive exclusion. In 1998, S. invicta populations were confirmed in southern New Mexico, resulting in a quarantine of Doña Ana County. Although large populations of the native southern fire ant, S. xyloniMcCook, are common in the Mesilla Valley, only two small populations of S. invicta have been found since the initial quarantine. At my rural household, the most vexatious insect pest has got to be the ubiquitous Japanese beetle. Native to Japan, these pests have infested most of eastern North America, with isolated infestations appearing in some western states. One reason that Japanese beetles are so bad is that they deliver a double-whammy: the larvae (grubs) consume roots and are particularly destructive of turf and pasture grasses while the adults consume leaves and flowers of a wide variety of plants, leaving behind skeletonized versions of the plant parts consumed.

A PCR based strategy was developed to screen a Xenopus oocyte λgt10 cDNA library. The PCR-based lift pool (LP) method follows the same two tiered strategy as conventional screening of phage libraries by filter hybridization. Two rounds of plating, one at high density to detect the clone, and one at low density to purify the clone to homogeneity, are performed. In the first round, lysates from high density plates, termed plate pools (PP), serve as template for PCR. 1. Lake sturgeon (Acipenser fulvescens) are threatened or endangered throughout much of their range. Juvenile sturgeon utilize sandy and silty habitats extensively during their growth. Invasive zebra mussels change the nature of sandy and silty habitats because they settle on and coat the habitat with the shells of living and dead individuals. The potential impacts of this increased habitat complexity on lake sturgeon is unknown. 2. Position effects can complicate transgene analyses. This is especially true when comparing transgenes that have inserted randomly into different genomic positions and are therefore subject to varying position effects. Here, we introduce a method for the precise targeting of transgenic constructs to predetermined genomic sites in Drosophila using the φC31 integrase system in conjunction with recombinase-mediated cassette exchange (RMCE). We demonstrate the feasibility of this system using two donor cassettes, one carrying the yellow gene and the other carrying GFP. The Arabidopsis thaliana wall-associated kinases (WAKs) bind to pectin with an extracellular domain and also contain a cytoplasmic protein kinase domain. WAKs are required for cell elongation and modulate sugar metabolism. This work shows that in leaf protoplasts a WAK1-GFP fusion protein accumulates in a cytoplasmic compartment that contains pectin. The WAK compartment contains markers for the Golgi, the site of pectin synthesis. The migration of WAK1-GFP to the cell surface is far slower than that of a cell surface receptor not associated with the cell wall, is influenced by the presence of fucose side chains on one or more …

Invasives like kudzu, Japanese honeysuckle, tree-of-heaven, and oh-so-many others, seem ubiquitous, crowding out native plants and altering all manner of ecological interactions. The wall-associated kinases (WAK), a family of five proteins that contain extracellular domains that can be linked to pectin molecules of the cell wall, span the plasma membrane and have a cytoplasmic serine/threonine kinase domain. Previous work has shown that a reduction in WAK protein levels leads to a loss of cell expansion, indicating that these receptor-like proteins have a role in cell shape formation. Here it is shown that a single wak2 mutation exhibits a dependence on sugars and salts for seedling growth. Nine microsatellite loci were isolated from the insect Dolichogenidea homoeosomae (Hymenoptera: Braconidae), an important parasitoid of the sunflower moth Homosoeosoma electellum (Lepidoptera: Pyralidae), and assayed for polymorphism. All nine loci were polymorphic within the five populations tested, with two to 14 alleles per locus. Expected and observed heterozygosities ranged from 0.39 to 0.90 and 0.25 to 0.72 respectively. These are the first microsatellite primers developed for D. homeosomae and will be useful for studies of population dynamics and connectivity. © 2006 Blackwell Publishing Ltd.

Solenopsis invicta Buren populations were confirmed in southern New Mexico, resulting in a quarantine of Doña Ana County in 1998; however, Solenopsis xyloni McCook remains the dominant fire ant. This study compared the queen weights and egg production of polygyne colonies of both species. There was no difference in live weights of the major worker caste; however, queens were very different in weight. In this study, S. invicta queens weighed almost twice as much as S. xyloni (S. invicta = 11.32 ± 0.25mg vs. S. xyloni = 6.40 ± 0.30mg) and produced over 50% as many eggs as S. xyloni. …

We describe the distress calls of Leptodactylus chaquensis and Leptodactylus elenae. They each consist of a single, long, tonal sound with complex harmonic structure and complex frequency modulation. The distress calls of both species are different from their respective advertisement calls. Moreover, there are also clear interspecific differences. Harmonic frequencies in L. elenae are higher, there are fewer emphasized harmonics and the call is shorter and repeated at a higher rate (sometimes paired) than the call of L. chaquensis. Leptodactylus fuscus is a neotropical frog ranging from Panamá to Argentina, to the east of the Andes mountains, and also inhabiting Margarita, Trinidad, and the Tobago islands. We performed phylogenetic analyses of 12S rRNA, 16S rRNA, tRNA-Leu, and ND1 mitochondrial (mt) DNA sequences from specimens collected across the geographic distribution of L. fuscus to examine two alternative hypotheses: (i) L. fuscus is a single, widely distributed species, or (ii) L. fuscus is a species complex. The single-strand overhang present at telomeres plays a critical role in mediating both the capping and telomerase regulation functions of telomeres. The telomere end-binding proteins, Cdc13 in Saccharomyces cerevisiae, Pot1 in higher eukaryotes, and TEBP in the ciliated protozoan Oxytricha nova, exhibit sequence-specific binding to their respective single-strand overhangs. S. cerevisiae telomeres are composed of a heterogeneous mixture of GT-rich telomeric sequence, unlike in higher eukaryotes which have a simple repeat that is maintained with high fidelity. In yeast, the telomeric overhang is recognized by the essential protein Cdc13, which coordinates end-capping and telomerase activities at the telomere. The Cdc13 …

Neuronal pentraxins (NPs) define a family of proteins that are homologous to C-reactive and acute-phase proteins in the immune system and have been hypothesized to be involved in activity-dependent synaptic plasticity. To investigate the role of NPs in vivo , we generated mice that lack one, two, or all three NPs. NP1/2 knock-out mice exhibited defects in the segregation of eye-specific retinal ganglion cell (RGC) projections to the dorsal lateral geniculate nucleus, a process that involves activity-dependent synapse formation and elimination.
