Sulfhydryl Modification Of The Calmodulin-Stimulated Phosphatase, Calcineurin., Jerome David. Wagner

Calcineurin (CaN), a calmodulin stimulated protein phosphatase is a heterodimer composed of a 61kD catalytic subunit and a 19kD regulatory subunit. The larger subunit has three binding sites for transition metal ions and also contains the intrinsic metal ions, Zn$\sp{2+}$ and Fe$\sp{3+}$. The smaller subunit contains four "EF-hand" Ca$\sp{2+}$ binding sites. All ten of calcineurin's cysteine residues are found on the catalytic subunit. This study explores the metal ion dependence of CaN using Ca$\sp{2+}$, Mg$\sp{2+}$, Mn$\sp{2+}$ and Ni$\sp{2+}$ as well as its thiol chemistry using Ellman's reagent, iodoacetate, iodoacetamide, glutathione and a novel thiol reagent introduced by this study, 1-p-chlorophenyl-4,4-dimethyl-5-diethylamino-1-penten-3-one …

Surface enhanced Raman (SERS), surface enhanced Resonance Raman (SERRS) of mono and multilayer films of three N,N$\sp\prime$-unsymmetrical dialkyl-3,4:9,10-perylenebis (dicarboximides)s, namely N-octyl-3,4:9,10-perylenetetracarboxylic monoanhydride(Oc-PTCDO), N-octyl-3,4:9,10-perylenetetracarboxylic monoimide(Oc-PTCDNH) and N-octyl, N$\sp\prime$-isobutyl-3,4:9,10-perylene-bis (dicarboximide) (PBDC) have been studied and vibrational analyses have been attempted. SERS and SERRS of evaporated thin films of perylene, 3,4:9,10 perylene tetracarboxylic dianhydride (PTCDA) and 3,4:9,10 perylene tetracarboxylic N,N$\sp\prime$ bis (methyl) diimide (PTCDIMe) on tin sphere, Ag island and Au island films are used to characterize the vibrations of the three unsymmetrical perylene derivatives. Further Studies On The Metabolism Of 4-Deoxy-4-Fluoro-D-Glucose In Pseudomonas Putida., John Watcyn. Williams

Following a 24 h incubation of glucose-grown whole cells of Pseudomonas putida with D- (U-$\sp{14}$C) 4-deoxy-4-fluoro-D-glucose (4FG), there is an extensive release of fluoride ion, (95 $\pm$ 5%). The isolated peptidoglycan (PG) fraction contains less than 1% of the radiolabel. This small, but significant amount of radioactivity is not due to the presence of a novel carbohydrate derived from 4FG. HPLC analysis with an Aminex HPX-87C (Bio-Rad) carbohydrate analytical column reveals the presence of the radioactive amino acids threonine, serine, glutamate and aspartate. These are considered to have arisen from the observed $\sp{14}$CO$\sb2$ from 4FG. When the incubation is performed …

Part I. N-sulfinyl-n-butylamine 26 and N-sulfinyl-n-pentylamine 34 were prepared according to the literature. Alkylation reactions using LDA as the base were performed on compounds 26 and 34 using benzylic and simple alkyl halides as the alkylating agent. No useful products were obtained using simple alkyl halides, but compounds 32, 33, 35, and 36 were isolated when benzyl bromide was used. These are the result of sulfur anion attack on the alkyl halide. Compounds 32, 33, 35, and 36 were subjected separately to cycloaddition reactions with 2,3-dimethylbutadiene. Part 1: Palladium catalyzed cross-coupling reaction. Many aryl bromides will undergo a palladium catalyzed cross-coupling reaction with 3- ((t-butyldimethylsilyl) oxymethyl) -4-(tri-n-butylstannyl)furan to form a new class of 3,4-disubstituted furans in excellent yields. The reaction is fast and easy to perform. It also represents an expansion of the scope of the cross-coupling reaction as it provides examples of rare aryl tin-aryl bromide couplings. Part 2: Lead tetraacetate oxidations of 2-(trialkylsilyl)furans. The treatment of 2-(t-butyldimethylsilyl)furan with lead tetraacetate in warm (70$\sp\circ$C) acetic acid provides 2-acetoxy-3-(t-butyldimethylsilyl)-$\Delta\sp3$-butenolide and 4-acetoxy-4-(t-butyldimethylsilyl)-$\Delta\sp2$-butenolide in a 2:1 ratio and 90% yield. The outer membrane of Pseudomonas putida is prepared using both lysozyme digestion and sucrose density gradient separation of French pressed whole cell suspensions of the bacteria. The sucrose density gradient preparation gives greater yields and is more suited to large scale preparations of the outer membrane of P. putida. Examination of the outer membrane preparations by sodium dodecyl sulfate polyacrylamide gel electrophoresis revealed that it contains 9 major protein bands. These proteins are similar but not identical to the outer membrane proteins of P. aeruginosa. One particular protein, Band E (42.9 kDa), is induced by growth of P. putida on …

Purification And Characterization Of Ubiquitin-Associated And Free Thiol Protease Inhibitors From Dormant Artemia Embryos., Margaret J. Sonnenfeld-Karcz

Encysted embryos of the brine shrimp Artemia contain a mixture of thiol protease inhibitors (TPI's) which appear to be important in the regulation of a cathepsin B-like protease. These TPI's were subjected to gel filtration, anion exchange chromatography, two types of cation exchange procedures (fast protein liquid chromatography and CM-cellulose chromatography) and high performance liquid chromatography (HPLC). Cation exchange chromatography of inhibitor preparations on a Mono S column fractionated six TPI peaks. Using HPLC to purify the proteins the TPI's eluted between 39% and 41% acetonitrile from a C-18 column revealing considerable hydrophobicity of each inhibitor. SDS-urea PAGE of these …
