Part I. 1,3-Oxathiolan-5-ones can be reduced to 2-mercaptoaldehydes which are synthetically important in the preparation of 2,5-dihydro-thiophenes and dienes.('1,2) Alkylation of 1,3-oxathiolan-5-ones was examined as a route to structurally more complex 2-mercaptoaldehydes. Alkylations were successful only with very reactive alkylating agents.('2,3) Aldol reactions were more successful. 1,3-Oxathiolan-5-ones were reduced with lithium aluminum hydride to 2-mercaptoalcohols, which could be transformed into the 2-(methylthio)alcohols. Part II. Camphor imines of glycinates were prepared from camphorthione and alkylated with various alkylating agents differing in steric requirements and reactivity. Both glucose- and succinate-grown whole cells of P. putida transport 2-keto-D-gluconate (2KGA) by a saturable process. The transport of 2KGA by membrane vesicles prepared from glucose-grown P. putida occurs by a facilitated diffusion process, with an apparent Km of 111.0 (+OR-) 2.9 (mu)M and a Vmax of 0.55 (+OR-) 0.04 nmol.mg protein.('-1)min.('-1). The provision of artificial and natural electron donors results in a significant decrease in Km value, while the Vmax remains virtually unchanged. 2KGA transport is inhibited by antimycin A, rotenone and DNP in the presence of L-malate/FAD. 4F2KGA and 3F2KGA also inhibit 2KGA transport competitively. Part I. A new fluorometric, coupled, enzymatic method for the determination of adenosine triphosphate is described. The procedure uses the hexokinase reaction coupled with three other enzymes in a system of phosphorylation and double reduction to form NADPH. The NADPH is measured in a reaction catalyzed by diaphorase in which the nonfluorescent material, resazurin, is converted to the highly fluorescent compound, resorufin. This kinetic method can determine adenosine triphosphate at the picomole level. Intra- and inter-assay CV's were both less than 3% and recoveries were quantitative. No significant interference was observed from EDTA or heparin. Correlation with an established method …
