Faculty Publications and Other Works -- Biomedical and Diagnostic Sciences

Background

Antimicrobial resistance is becoming increasingly important in both human and veterinary medicine. This study investigated the proportion of antimicrobial resistant samples and resistance patterns of Staphylococcus isolates from cats presented at a veterinary teaching hospital in South Africa. Records of 216 samples from cats that were submitted to the bacteriology laboratory of the University of Pretoria academic veterinary hospital between 2007 and 2012 were evaluated. Isolates were subjected to antimicrobial susceptibility testing against a panel of 15 drugs using the disc diffusion method. Chi square and Fisher’s exact tests were used to assess simple associations between antimicrobial resistance and …

Molecular Basis Of Surface Anchored Protein A Deficiency In The Staphylococcus Aureus Strain Wood 46, Manasi Balachandran, Richard J. Giannone, David A. Bemis, Stephen A. Kania Aug 2017

Molecular Basis Of Surface Anchored Protein A Deficiency In The Staphylococcus Aureus Strain Wood 46, Manasi Balachandran, Richard J. Giannone, David A. Bemis, Stephen A. Kania

Protein A in Staphylococcus aureus is encoded by the spa (staphylococcal protein A) gene and binds to immunoglobulin (Ig). The S. aureus strain Wood 46 has been variously reported as protein A-deficient and/or spa negative and used as a control in animal models of staphylococcal infections. The results of this study indicate that Wood 46 has normal spaexpression but transcribes very low levels of the srtA gene which encodes the sortase A (SrtA) enzyme. This is consistent with unique mutations in the srtA promoter. In this study, a low level of sortase A explains …

Complete Genome Sequence Of Staphylococcus Pseudintermedius Type Strain Lmg 22219, Mohamed Adel Salaheldin Abouelkhair, Stephen Kania, D A. Bemis, Matthew C. Riley Feb 2017

Complete Genome Sequence Of Staphylococcus Pseudintermedius Type Strain Lmg 22219, Mohamed Adel Salaheldin Abouelkhair, Stephen Kania, D A. Bemis, Matthew C. Riley

Faculty Publications and Other Works -- Biomedical and Diagnostic Sciences

We report the first complete genome sequence of LMG 22219 (=ON 86T = CCUG 49543T), the Staphylococcus pseudintermedius type strain isolated from feline lung tissue. This sequence information will facilitate phylogenetic comparisons of staphylococcal species and other bacteria at the genome level.

An Exploratory Descriptive Study Of Antimicrobial Resistance Patterns Of Staphylococcus Spp. Isolated From Horses Presented At A Veterinary Teaching Hospital, James Wabwire Oguttu, Daniel Nenene Qekwana, Agricola Odoi Jan 2017

An Exploratory Descriptive Study Of Antimicrobial Resistance Patterns Of Staphylococcus Spp. Isolated From Horses Presented At A Veterinary Teaching Hospital, James Wabwire Oguttu, Daniel Nenene Qekwana, Agricola Odoi

Faculty Publications and Other Works -- Biomedical and Diagnostic Sciences

Background

Antimicrobial resistant Staphylococcus are becoming increasingly important in horses because of the zoonotic nature of the pathogens and the associated risks to caregivers and owners. Knowledge of the burden and their antimicrobial resistance patterns are important to inform control strategies. This study is an exploratory descriptive investigation of the burden and antimicrobial drug resistance patterns of Staphylococcus isolates from horses presented at a veterinary teaching hospital in South Africa.

Methods

Retrospective laboratory clinical records of 1027 horses presented at the University of Pretoria veterinary teaching hospital between 2007 and 2012 were included in the study. Crude and factor-specific percentages …

Determination Of Meloxicam In Egg Whites And Yolks Using Reverse Phase Chromatography, Sherry Cox, Joan Bailey, Molly White, Kristen Gordon, Marcy Souza Jan 2017

Determination Of Meloxicam In Egg Whites And Yolks Using Reverse Phase Chromatography, Sherry Cox, Joan Bailey, Molly White, Kristen Gordon, Marcy Souza

Faculty Publications and Other Works -- Biomedical and Diagnostic Sciences

A new method of analysis has been developed and validated for the determination of meloxicam in egg whites and yolks. Following a liquid extraction for the whites and a solid phase extraction for the yolks, samples were separated on an XBridge C18 column and quantified using ultraviolet detection at 360 nm. The mobile phase was a mixture of water with glacial acetic acid and aceto- nitrile, with a flow rate of 1 mL/min. The procedure produced a linear graph over the concentration range 5–1500 ng/mL with a lower limit of quantification of 5 ng/mL. Intra- and inter-assay variabil- ity was …

Patterns And Predictors Of Antimicrobial Resistance Among Staphylococcus Spp. From Canine Clinical Cases Presented At A Veterinary Academic Hospital In South Africa, Daniel N. Qekwana, James W. Oguttu, Fortune Sithole, Agricola Odoi

Faculty Publications and Other Works -- Biomedical and Diagnostic Sciences

Background

Antimicrobial resistance in staphylococci, often associated with treatment failure, is increasingly reported in veterinary medicine. The aim of this study was to investigate patterns and predictors of antimicrobial resistance among Staphylococcus spp. isolates from canine samples submitted to the bacteriology laboratory at the University of Pretoria academic veterinary hospital between 2007 and 2012. Retrospective data of 334 Staphylococcus isolates were used to calculate the proportion of samples resistant to 15 antimicrobial agents. The Cochran-Armitage trend test was used to investigate temporal trends and logistic regression models were used to investigate predictors of antimicrobial resistance in Staphylococcus aureus and Staphylococcus …

The Influence Of Storage Time And Temperature On Propofol Concentrations In Canine Blood And Plasma., Sherry Cox Jan 2017

The Influence Of Storage Time And Temperature On Propofol Concentrations In Canine Blood And Plasma., Sherry Cox

Faculty Publications and Other Works -- Biomedical and Diagnostic Sciences

Propofol is an intravenous anesthetic commonly used due to its favorable pharmacokinetic and pharmacodynamic profile. There are discrepancies in the literature about the most appropriate sample for determining propofol concentrations. Although plasma has been used for determining propofol concentrations, whole blood has been the preferred sample. There is also a lack of consistency in the literature on the effect of storage time and temperature on propofol concentrations and this may lead to errors in the design of pharmacokinetic/pharmacodynamics studies. The purpose of this study was to determine the difference in propofol concentrations in whole blood versus plasma and to evaluate …
