The prospects of nanoparticle-based drug delivery and imaging have been hindered by insufficient understanding of the effects of nanoparticle physicochemical properties on their in vivo disposition. Here, we present an integrative mathematical modeling and in vivo imaging approach to quantify the relationship between nanoparticle physicochemical properties, namely, size, surface charge, and surface chemistry, on their in vivo disposition kinetics in healthy rats. We developed simple master equations in closed-form to accurately represent the time-dependent concentration of nanoparticles in different regions of the body and obtain functional relationships for predictive purpose to support rational design of nanomedicine. Colorectal cancer (CRC) is the third most common malignancy in the United States for men and women. A major risk factor for CRC development and progression is inflammation, in particular the inflammatory cytokines IL-1, IL-6, and TNF-α. These proteins promote critical cancer functions, such as proliferation, invasion, survival, and migration. Control of inflammation is one potential area of therapeutic intervention for some cancers. Because signaling of these cytokines overlaps in many of the same pathways, targets upstream of protein production could prove an effective strategy to reduce inflammation. Human papillomaviruses (HPVs) are the most common sexually transmitted infectious agents. They are responsible for >99% of all cervical cancers as well as subsets of other anogenital cancers. HPVs are also the causative agents of a growing number of head and neck cancers. As such, they present a significant health problem both in the U.S. and developing countries. Much is still unknown regarding the factors underlying progression from HPV infection to cancer or maintenance of viral oncoprotein expression following oncogenic transformation. Breast cancer is the second leading cause of cancer related deaths in women. Advanced breast cancer can metastasize to the lungs, liver, bones and brain becoming fatal conditions for many patients. There is a dire need for metastasis preventing medications, however the process required for a medication to become FDA approved for clinical use is long and arduous. Studies have found promising benefits for breast cancer patients given ToradolTM, or racemic ketorolac, as an NSAID during resection surgery. Our recent studies have shown that Tumor Adjacent Histologically Normal (TAHN) breast tissue demonstrates many of the characteristics of breast tumors. For example, through immunohistochemical staining with markers such as alpha smooth muscle actin and TGF-β, we have shown the accumulation of myofibroblasts in TAHN tissues 1 cm from the tumor margin (TAHN-1). Additionally, TAHN-1 epithelia stain positive for epithelial to mesenchymal transition (EMT) associated proteins, such as transforming growth factor beta and alpha smooth muscle actin. The purpose of this study was to determine if TAHN-1 fibroblasts retained myofibroblast characteristics and if they could induce EMT in primary culture. …
