Grapevine vein clearing virus (GVCV) is an emerging pathogen causing severe damage to cultivated grapevines in the Midwest area of the United States. The prevalence of GVCV has been reported in native Vitis spp. and Ampelopsis cordata, a close relative of vitis in the Vitaceae family, which act as a reservoir for the virus. GVCV can be transmitted from wild A. cordata to Vitis spp. by Aphis illinoisensis (grape aphids) under greenhouse conditions, but the prevalence and transmission in native populations remains unknown. Jaeger 70 cultivar is an interspecific hybrid of Vitis linececumii x Vitis rupestris, with an interesting past. It was created by Hermann Jaeger of Neosho, MO, who is most notable for saving the French wine industry by developing phylloxera resistant rootstocks in the mid-1800s. Traditionally the Jaeger 70 variety has been grafted to French wine grape cultivars to impart disease resistance and cold hardiness while retaining desirable wine characteristics. One of the most effective methods of reducing pesticide application in vineyards is breeding hybrid varieties for disease resistance. The new varieties must be assessed for disease resistance and viticultural traits. This thesis focuses on seven new varieties from a cross of ‘Norton’ and ‘Cabernet Sauvignon’. ‘Norton’ was chosen because of its disease resistance and cold hardiness. ‘Cabernet Sauvignon’ was selected because of its high-quality wine making berries. A recent threat to the sustainability of grape production is Grapevine vein-clearing virus (GVCV), the first DNA virus discovered in grapevines. Infection with GVCV leads to vine decline, lower quality berries, and eventual death of the grapevine. Since GVCV was discovered in cultivated grapevines, research has been dedicated to investigating its range and origin. The entire genome of the first GVCV isolate from a grape cultivar ‘Chardonel' has been deposited in GenBank and is used as a reference genome.
