Even seasoned workers find it tedious and sometimes frustrating to remove Hülle cells and stray conidia from Aspergillus cleistothecia by rolling them with forceps across an agar surface, particularly when large numbers must be cleaned. It can be even more challenging to teach the skill to others, especially to a whole class of easily discouraged undergraduates, who may be seeing their first high-mag image of forceps tips at the same time as their first view of a cleistothecium.

Split-Marker Recombination For Efficient Targeted Deletion Of Fungal Genes, Natalie L. Catlett, Bee-Na Lee, O. C. Yoder, B. Gillian Turgeon

A commonly used method for fungal gene deletion is introduction of linear DNA consisting of a selectable marker gene flanked on both sides by short stretches of DNA that target a gene of interest (Wirsel et al 1996 Curr. Genet 29:241-249). Gene deletion in Cochliobolus heterostrophus and Gibberella zeae occurs efficiently with this approach. To facilitate deletion construct synthesis, we have applied the "split-marker” deletion strategy previously developed for Saccharomyces cerevisiae (Fairhead et al. 1996 Yeast 12:1439-57; Fairhead et al. 1998 Gene 223:33-46). A simple acetone-drying protocol was adopted to replace the lyophilization step while isolating genomic DNA from Aspergillus mycelia. This DNA is suitable for PCR, restriction enzyme digestion and Southern blot analysis with digoxigenin-labeled DNA probes. Acetone drying/ preservation can be a useful method in the molecular analysis of fungal DNA samples.

Neurospora possesses more cell types than are commonly recognized. We have been able to identify 28 morphologically distinct types. Having the cell types clearly defined will be important for genome annotation, describing new mutant phenotypes, and determining sites of gene expression.

During the evolutionary divergence of N. tetrasperma from the eight-spored Neurospora species, ascus development was reprogrammed with the result that each of the four large ascospores is heterokaryotic, containing nuclei of both mating types, and germlings are self-fertile. Unique features of genome oganization, cell biology, and population structure have attracted investigators to use this pseudohomothallic, four-spored species for a wide range of studies. A bibliography listing 164 publications was published in 1994 in Fungal Genetics Newsletter 41:72-78. Eighty-six additional publications are listed here. Podospora anserina is a coprophilous fungus growing on herbivore dung. It is a pseudohomothallic species in which ascus development results, as in Neurospora tetrasperma but through a different process, in the formation of four large ascospores containing nuclei of both mating types.

We report the construction of a Destination Vector, called pJHAM007, for the targeted integration of DNA sequences at the histidine-3 (his-3) locus of Neurospora crassa. pJHAM007 has all the necessary features required to perform a simple, rapid and efficient GATEWAY™ recombinational cloning with an Entry Clone to yield a his-3-gene replacement Destination Vector.

The Neurospora crassa osmotic-1 locus (os-1) encodes a protein with homology to two component histidine kinase sensors. We formed forced heterokaryons between each of ten os-1 alleles in all pair-wise combinations and found, in contrast to a previous report, no evidence for intra-allelic complementation.

Communicating Biotechnology: Relationships Between Tone, Issues, And Terminology In U.S. Print Media Coverage, Jefferson D. Miller, Mamane Annou, Eric J. Wailes

As part of a nationwide study on the public acceptability of agricultural biotechnology, researchers performed a content analysis on two years of print news coverage of biotechnology (2000-2002). Qualitative analysis methods included examining text from selected national newspapers, regional newspapers, and trade publications for common issues, tone, and terminology. A team of three coders, trained to an acceptable level of agreement (Cohen’s K = .80), examined and coded the articles. The relationship between the tone of the article and the terminology used in reference to biotechnology (e.g., “genetically engineered,” “genetically modified,” or “biotechnology”) has practical applications for both public relations …

How Newspapers In Kansas Are Using News Releases From K-State Research And Extension, Pat Melgares, Tracy Rutherford, Lori Alexander

This study looks at how newspaper editors in Kansas use news releases from Kansas State University Research and Extension (KSRE), which distributes more than 700 news releases annually to newspapers across the state. An analysis of clippings over 10 weeks showed that editors rarely change the original text or headline in KSRE news releases, and more than half of the releases were published within 10 days of release. KSRE news releases were published in all regions of Kansas. Using a formula to determine the advertising equivalency value of news clips, the data indicates a value of $33,286.94 for the sample …
