Transcription factor (TF) footprinting uncovers putative protein-DNA binding via combined analyses of chromatin accessibility patterns and their underlying TF sequence motifs. TF footprints are frequently used to identify TFs that regulate activities of cell/condition-specific genomic regions (target loci) in comparison to control regions (background loci) using standard enrichment tests. However, there is a strong association between the chromatin accessibility level and the GC content of a locus and the number and types of TF footprints that can be detected at this site. Traditional enrichment tests (e.g. hypergeometric) do not account for this bias and inflate false positive associations ...

BACKGROUND: In the United States, worksite wellness programs are more often offered by larger employers. The Massachusetts Working on Wellness (WoW) program is an innovative, statewide capacity-building model designed to increase the number of smaller employers (200 or fewer workers) adopting health promotion initiatives. This article describes the WoW program design and approaches to recruitment, implementation, and evaluation.

METHODS/DESIGN: WoW provides employer training, technical assistance and seed funding, utilizing a Wellness Program Development framework based on recognized good practices. BACKGROUND: Glioblastoma is a malignant brain tumor characterized by rapid growth, diffuse invasion and therapeutic resistance. We recently used microRNA expression profiles to subclassify glioblastoma into five genetically and clinically distinct subclasses, and showed that microRNAs both define and contribute to the phenotypes of these subclasses. Here we show that miR-29a activates a multi-faceted growth and invasion program that promotes glioblastoma aggressiveness.

METHODS: microRNA expression profiles from 197 glioblastomas were analyzed to identify the candidate miRNAs that are correlated to glioblastoma aggressiveness. The candidate miRNA, miR-29a, was further studied in vitro and in vivo.

Amorphous SiOC films were synthesized by magnetron sputtering at room temperature with/without radio frequency (RF) bias and further improved in terms of mechanical properties by ion irradiation. As-deposited SiOC films without RF bias exhibit catastrophic failure at a low stress and strain, which is ascribed to microstructural heterogeneities associated with the formation of voids during deposition, as evidenced by transmission electron microscopy. Ion irradiation unifies microstructure accompanied with eliminating the voids, resulting in a simultaneously increase in strength and plasticity (ultimate strength of 5–7 GPa and the strain to shear instability of over 20%). Quantifying the genetic correlation between cancers can provide important insights into the mechanisms driving cancer etiology. Using genome-wide association study summary statistics across six cancer types based on a total of 296,215 cases and 301,319 controls of European ancestry, here we estimate the pair-wise genetic correlations between breast, colorectal, head/neck, lung, ovary and prostate cancer, and between cancers and 38 other diseases. Fire suppression and exclusion, the historically dominant paradigm of fire management, has resulted in major modifications of fire-dependent ecosystems worldwide. These changes are partially credited with a recent increase in wildfire number and extent, as well as more extreme fire behavior. Fire and herbivory historically interacted, and research has shown that the interaction creates a unique mosaic of vegetation heterogeneity that each disturbance alone does not create.
