Transient gene expression in plant protoplasts has been widely utilized for rapid functional screening of promoters. This project describes a novel soybean cell suspension culture and protoplast system for rapid and predictive analysis of promoter activity. Since recovery of transgenic lines is low in soybean stable transformation, high-throughput studies of promoter activity have intrinsic limitations. First, I describe a novel cell suspension culture system from leaf-derived callus. Once the cell culture is established, it is easy to maintain, sterile, and produces fast-growing cells amenable for protoplast production. Thiamethoxam, an insecticide used in soybean seed treatments, effectively suppresses soybean aphids (Aphis glycines) Matsumura (Hemiptera: Aphididae) for a short time after planting. However, exactly when and how quickly soybean aphid populations could increase is unknown. Likewise, we lack data on virulent soybean aphid biotypes (that can overcome soybean resistance) when fed on seed-treated soybean. The narrow genetic pool of soybean (Glycine max L. Merr.) in North America can limit its future yield gains. Among the worldwide germplasm collection of 45,000 unique landraces, only 80 contribute 99% to the collective parentage of North American soybean cultivars. Among these 80 landraces, just 17 contribute to 86% of the collective parentage of the modern cultivars. The Soybean Nested Association Mapping population (SoyNAM) was therefore developed with the objective of diversifying the soybean gene pool. Flood tolerance in soybean (Glycine max) is not a well-characterized trait, yet flooding damage is second only to drought stress in terms of yield reduction. The objectives of this study were to determine genetic variation for flooding tolerance in two populations of soybean recombinant inbred lines (RILs) and to identify and confirm flood tolerant QTL. Merr.], fourteen injury criteria observed following dicamba at 0.6 to 280 g ae ha-1 (1/1000 to ½ of 560 g ha-1 use rate) were rated using a scale of 0= no injury, 1= slight, 2= slight to moderate, 3= moderate, 4= moderate to severe, and 5= severe. A declining market for soybean oil production caused by added restrictions on oil hydrogenation practices, has fueled improvements in oil industry standards. Antinutritional carbohydrates such as raffinose and stachyose lead to irritation to the gut for monogastric livestock as well as unhealthy weight gain. Loss of pod dehiscence is a key step during soybean [Glycine max (L.) Merr.] domestication. Genome-wide association analysis for soybean shattering identified loci harboring Pdh1, NST1A and SHAT1-5. Pairwise epistatic interactions were observed, and the dehiscent Pdh1 overcomes the resistance conferred by NST1A or SHAT1-5 locus, indicating that Pdh1 predominates pod dehiscence expression. Further candidate gene association analysis identified a nonsense mutation in NST1A associated with pod dehiscence. Allele composition and population differential analyses unraveled that Pdh1 and NST1A, but not SHAT1-5, underwent domestication and modern breeding selections. Field trials are commonly used to estimate the effects of different factors on crop yields. In the present study, we followed an alternative approach to identify factors that explain field-to-field yield variation, which consisted of farmer survey data, a spatial framework, and multiple statistical procedures. Arkansas is the leading soybean-producing state in the mid-southern United States. Arkansas ranked 11th in soybean production in 2016 when compared to the other soybean-producing states in the U.S. The state represents 3.4% of the total U.S. soybean production and 3.8% of the total acres planted to soybean in 2016. The 2016 state soybean average was 47 bushels per acre, 2.5 bushels per acre less than the state record soybean yield set in 2014. The compatible interaction of SCN with susceptible soybean plants is mediated by differential expression of thousands of genes in the infected root cells, leading to the formation of a functional feeding site, the syncytium. Introduction of a new dicamba-resistant (Xtend) trait in soybean and cotton will increase dicamba herbicide use. Consequently, concern for injury to sensitive crops from off-target movement and tank contamination will likely increase. Soybean sudden death syndrome (SDS) is one of the most important soybean diseases in the Midwestern United States, leading to average losses of $396 million per year from 1996 to 2015. The causal agent of SDS, soilborne fungus Fusarium virguliforme (Fv), causes root rot symptoms and releases phytotoxins taken up by the plant to produce chlorosis and necrosis in the leaves. Background: Ubiquitination is a major post-translational protein modification that regulates essentially all cellular and physiological pathways in eukaryotes. The ubiquitination process typically involves three distinct classes of enzymes, ubiquitin-activating enzyme (E1), ubiquitin-conjugating enzyme (E2) and ubiquitin ligase (E3). To date, a comprehensive identification and analysis of core components comprising of the whole soybean (Glycine max) ubiquitin system (UBS) has not been reported.

Glyphosate (N-(phosphonomethyl)glycine) herbicide has been extensively used on several herbicide tolerant crops in the United States since the introduction of the first commercially available genetically modified glyphosate-tolerant soybean [Glycine max (L.) Merr.] seed in 1996. The launch of commercially available glyphosate-tolerant corn (Zea mays L.) seed in 1998 provided farmers in Iowa and other states, where corn and soybean are primarily grown, the ability to apply glyphosate to every acre of these crops annually.

The agricultural industry had a rapid increase in commodity prices from late in 2010 until the fall of 2014. With those higher prices, there was a niche market for added-value products brought to the market, many of which promised the local grower a five-bushel yield increase per acre on soybeans. I wanted to focus on a select group of these products, plant growth regulators (PGRs). I specifically worked with a commercial product that contained three phytohormones, cytokinin, a gibberellic acid, and an auxin.
