Identification And Characterization Of Isoflavone Reductase Family Members In Soybean, Negin Azizkhani
Identification And Characterization Of Isoflavone Reductase Family Members In Soybean, Negin Azizkhani
Electronic Thesis and Dissertation Repository
Soybean’s yield is threatened by Phytophthora sojae, a pathogen responsible for stem and root rot disease. Glyceollins, unique antimicrobial agents specific to soybeans in partially preventing P. sojae infection, are derived from the isoflavonoid branch of the general phenylpropanoid pathway. One pivotal enzyme exclusively involved in glyceollin synthesis in soybean is the isoflavone reductase (GmIFR), which catalyzes the 2'-hydroxydaidzein conversion to 2'-hydroxy-2,3-dihydrodaidzein as a precursor for glyceollin biosynthesis. To comprehensively identify all members of the GmIFR gene family within the soybean genome, keyword and blast protein searches were conducted, identifying 98 putative GmIFRs. Among these candidates, …

Evaluation Of Cultivar Susceptibility And In-Furrow Fungicide Efficacy For Management Of Southern Blight Of Soybean, Adam Joseph Connor
Theses and Dissertations
Southern Blight (SB), caused by the fungus Athelia rolfsii, has increasingly impacted Mississippi soybean production with estimated economic losses in recent years rising from $181,616 in 2016 to $9,508,412 in 2021. Currently, there are no recommended fungicides and no known commercially available cultivar resistance. Eleven cultivars were evaluated for their response to the presence of Athelia rolfsii in the 2021 field trial. An in vitro fungicide assay was conducted to evaluate the efficacy of five fungicides to determine the respective EC50. These fungicides were then evaluated in combination with three cultivars, one mild, one moderate, and one severe in the …
A Robust Agroinfiltration Method, Bryce N. Trull
A Robust Agroinfiltration Method, Bryce N. Trull
Masters Theses
Stable transformation of soybean (Glycine max) is a markedly slow and laborious process. Thus, a tool that enables rapid evaluation of genetic elements in planta is critical to advance complex research and genetic engineering in soybean. To that end, a substantially robust agroinfiltration method was innovated in this work. Agroinfiltration is a technique that leverages Agrobacterium-mediated transient transformation to deliver genetic elements to the cells of whole plant tissues, usually leaves. Several factors were found to be relevant to successful soybean leaf agroinfiltration, including genotype, surfactant, developmental stage, and Agrobacterium culture medium. The research represents not only …

Evaluation Of Weed Control Efficacy And Crop Tolerance To Florpyrauxifen-Benzyl Coated Onto Urea, Bodie Lyn Cotter
Graduate Theses and Dissertations
Frequent off-target movement occurrences to soybean [Glycine max (L.) Merr.], rice (Oryza sativa L.) injury, and a lack of barnyardgrass [Echinochloa crus-galli (L.) P. Beauv.] control followed the commercial launch of florpyrauxifen-benzyl in 2018. Florpyrauxifen-benzyl is a synthetic auxin, Herbicide Resistance Action Committee (HRAC)/Weed Science Society of America (WSSA) Group 4 herbicide used for postemergence weed control in rice and labeled for application at 30 g ae ha-1. Previous research documented that soybean was susceptible to injury following spray applications of florpyrauxifen-benzyl at less-than-labeled rates, and certain varieties of rice are less tolerant to spray-applied florpyrauxifen-benzyl at 30 g ae …

Impact Of Fermented And Non-Fermented Plant-Based Foods Supplementation On Gut Microbiota And Metabolites In C57bl/6j Mice, Priya Darshan Gandhi
Masters Theses
Plant-based proteins have gained popularity because of their high nutritional value and more sustainable alternative to animal-based proteins. Soybean and chickpea are two widely consumed plant-based proteins, whereas tempeh is a popular plant-based fermented whole food product that is rich in protein. With the increase in the development of plant-based food products, there is little research into how plant proteins affect gut microbiota characteristics and metabolites. Therefore, there is a need to understand the underlying mechanisms surrounding the consumption of these foods. The purpose of this study was to investigate the health benefits of soybean, chickpea, and their tempeh counterparts’ …
Survey And Prevalence Of Palmer Amaranth Herbicide Resistance In South Carolina, Mitchell Williams
Survey And Prevalence Of Palmer Amaranth Herbicide Resistance In South Carolina, Mitchell Williams
All Theses
Palmer amaranth is a troublesome weed for growers to control, not only due to its aggressive growth characteristics that limit row-crop production, but because of its resistance to different herbicide modes of action. The first case of herbicide resistance in Palmer amaranth was detected in 1989 and has since grown to nine different herbicide classes throughout the United States. New herbicide modes of action have not been developed since the 1980s, so proper stewardship of the remaining modes of action is important for effective control of Palmer amaranth. Increased herbicide resistance from states bordering South Carolina have been reported; therefore, …

Theses and Dissertations
HearNPV was applied to a soybean field and evaluated for residual efficacy at different time intervals (0, 12, 24, 36, 48, and 72 hours) after application in laboratory bioassays with treated leaves. Larval mortality was rated at five, seven and twelve days after larval infestation. Larval mortality increased as the number of days after infestation increased. To evaluate the feeding habits of Helicoverpa zea at the 2nd and 3rd instar, HearNPV exposed larvae and untreated larvae were weighed at zero (before diet exposure), four, and seven days. At four days after exposure, larval weight gain of …
Planting Date, Seed Size, And Maturity Effects On Cotton And Soybean Growth And Yield In South Carolina, Sarah Holladay
Planting Date, Seed Size, And Maturity Effects On Cotton And Soybean Growth And Yield In South Carolina, Sarah Holladay
All Dissertations
Seed size, seeding rates, pre-emergence (PRE) herbicides, planting dates, and maturity groups are all important factors that growers must consider before planting cotton or soybean. Previous research has suggested that seed size and seedling vigor are positively correlated and having greater seeding vigor may compensate for adverse planting conditions, pest and disease pressure, and potentially injury from PRE herbicides. Seeding rates and planting dates for cotton are dependent on the variety, environmental conditions, and grower goals. Planting date and maturity group selection are also important considerations for soybean growers in SC as in many cases soybean is planted in rotation …

Effect Od Different Sources And Application Rates Of Sulfur On Corn And Soybean Production Systems In Louisiana, Diego Mayorga Valladares
LSU Master's Theses
Sulfur (S) is a structural component of amino acids such as cysteine and methionine and is involved in important functions within the plant like photosynthesis, carbon and nitrogen metabolism, protein synthesis of oils, and detoxification mechanisms. Sulfur deficiency in crops has intensified around the world. Some of the reasons are improvement in controlling SO2 emissions from industries, growing usage of high analysis S-free fertilizers and augmented cropping intensity. Sulfur fertilization has become an important factor in crop production systems and fertilization guidelines for S need to be up-to-date to improve use efficiency and compensate for rising prices of fertilizers. …
Metagenomic Analysis Of Root-Associated Microbiome Of Healthy And Taproot Decline-Affected Soybeans And Identification Of Healthy Soybean Root Endophytes With Protective Activity Against The Causal Agent, Xylaria Necrophora, Uyen Wesser
Theses and Dissertations
Plant roots are associated with a diverse microbial community of bacteria and fungi. The root microbiome communities associate with the root surface or penetrate the superficial layers of root tissues. It is of high agronomic interest to characterize root microbiomes and identify microbes with beneficial effects on plant defense against pathogens. Soybean taproot decline (TRD) is an emerging soil-borne disease caused by a fungus classified as Xylaria necrophora, challenging to control and with devastating effects on yield. Our group has initiated a study of the soybean root microbiome with three primary experiments: (I) perform a comparative study of root …

Doctoral Dissertations
Proteinase inhibitors (PIs) from legumes have the potential for use as protectants in response to pests and pathogens. Soybean (Glycine max) contains two trypsin inhibitors (TIs): Kunitz trypsin inhibitor (KTI) and Bowman-Birk inhibitor (BBI). In this study, the possible role of soybean TIs in plant defense against insects and nematodes was investigated. In addition to the three known TIs (KTI1, KTI2 and KTI3), novel inhibitors KTI5, KTI7, and BBI5 were identified in soybean. Their functional role was further examined by overexpression in soybean and Arabidopsis. In vitro enzyme inhibitory assays showed significant increase in trypsin and chymotrypsin inhibitory …
Additional Sulfur Does Not Alleviate Cadmium Toxicity In Soybean, Spencer Matt
Additional Sulfur Does Not Alleviate Cadmium Toxicity In Soybean, Spencer Matt
Electronic Thesis and Dissertation Repository
Cadmium (Cd) is non-essential and toxic. Sulfur (S) addition to contaminated soils reduces Cd toxicity in rice and corn. I aimed to determine the underlying mechanisms by which S reduces Cd toxicity in hydroponically- and soil-grown soybean. In the presence of Cd, plant biomass was reduced by ~20%, Cd accumulated up to 45 μg/g in roots and 15 μg/g shoots, and concentrations of Cd chelators increased by more than 10-fold. Addition of S to Cd- treated plants had no effect on plant biomass, concentrations of Cd in roots and shoots, or vacuolar Cd sequestration in the root cortex. While additional …

Determining Residual Control And Systemic Activity Of Commonly Used Insecticides In Soybean And Cotton, Jacob H. Smith
Theses and Dissertations
Chemical control is a critical component of integrated pest management in cotton and soybean production. Residual efficacy of foliar insecticides can be highly variable and difficult to quantify due to several factors. The purpose of this research was to determine residual control and concentrations in flowering structures of commonly used insecticides. This research focused on the concentrations of active ingredient within the plant as well as efficacy over time. Previous research suggested chlorantraniliprole had a long residual and was highly lethal on corn earworm up to 28 days after treatment (DAT) in soybean; however, the results found in cotton were …
Evaluating The Ecological Relationships Among Beauveria Bassiana, Kudzu Bug And Kudzu, Kassie L. Hollabaugh
Evaluating The Ecological Relationships Among Beauveria Bassiana, Kudzu Bug And Kudzu, Kassie L. Hollabaugh
Masters Theses
When kudzu bug, Megacopta cribraria, was identified in the United States in 2009 as an invasive species, populations began to establish, increase, and spread throughout the southeastern region at intense rates. Kudzu bug invades urban structures, causing unpleasant scenes for homeowners, and reduces crop yield, such as soybean. Kudzu bug caused about $2.8 million in soybean yield loss and treatment in Tennessee in 2020. Although initial spread was rapid throughout the southeastern U.S., spatial modeling has shown populations of kudzu bug are declining and reducing risk of crop damage to growers. These local and regional declines in populations appear …

Soybean [Glycine max (L) Merr.], a legume species native to East Asia in the Fabaceae family, ranks among the most important food crops in the world. It is widely grown and known for its high protein and oil concentration. Soybean is valuable because its seeds have multiple applications in food, feed, pharmaceutical, and industrial enterprises. Even though seed yield is the most important trait, breeders have recently given a significant attention to quality traits, such as high protein or modified oil concentration. Soybean seed protein inheritance has been extensively studied; however, genetics of high-protein ‘BARC-7’ soybean are still unknown. On the …
Root Phenotyping Of Peptide-Treated Glycine Max, Salem Jackson
Root Phenotyping Of Peptide-Treated Glycine Max, Salem Jackson
Biological Sciences Undergraduate Honors Theses
Plant elicitor peptides (Peps) – endogenous chains of amino acids involved in natural plant defense – have been shown to decrease damage from herbivores and pathogens by inducing an immune response, increasing the emission of volatile organic compounds (VOCS), transcripts, and metabolites. Exogenous treatment of soybean seeds with plant elicitor peptide GmPep3 has been shown to induce these broad-spectrum defenses and offers a new method for increasing crop yield. However, the effects of GmPep3 on indicators of soybean health – root characteristics, growth stages, etc. – have not been fully realized.
Using the root-phenotyping platform RhizoVision Explorer, several root traits …

Understanding The Cellular And Physiological Mechanisms Of Fertilization And Early-Stage Seed Development, Mohammad Foteh Ali
Theses and Dissertations--Plant and Soil Sciences
Flowering plants have evolved a unique double-fertilization process. Two sperm cells fuse with two female gametophytic cells, the egg and central cells within the ovule, giving rise to the embryo and endosperm, respectively. Sperm cells in flowering plants are nonmotile and delivered in close proximity to the egg and central cells by the pollen tube. Flowering plants have established filamentous actin (F-actin) based sperm nuclear migration system for successful fertilization. Prior to fertilization, the female gamete forms a mesh-like structure of F-actin that shows constant inward movement from the plasma membrane periphery to the center of the cell where the …

Soybean (Glycine Max) Response To Multiple, Sublethal Exposures Of 2,4-D And Dicamba From Vegetative Through Reproductive Growth, Graham Robert Oakley
Theses and Dissertations
This study was conducted to determine whether soybean productivity is affected by multiple, sublethal herbicide exposures. The effects of dicamba and 2,4-D on soybean (Glycine max) productivity was investigated at 17 site-years. Relative to a single exposure of dicamba at R1, an additional exposure at either V3 or R3 reduced yield up to 23%. Three or more applications did not further decrease yields relative to an R1&R3 exposure. For 2,4-D, a single application to V3, R1, R3, or R5 soybean did not affect grain yield. However, two exposures of 2,4-D occurring from V3 through R3 reduced yield 5 to 7%. …

The Effects Of Silver Nanoparticles On Soybean (Glycine Max) Growth And Nodulation, Paul J. Boersma
Electronic Thesis and Dissertation Repository
Due to their antimicrobial properties, silver nanoparticles (AgNPs) have become more popular in consumer and industrial products, leading to increasing agricultural and environmental concentrations. Exposure to AgNPs could be detrimental to plants, microbes, and their symbiotic relationships. When subjected to 10 µg/mL AgNPs in a 96-well plate, growth of Bradyrhizobium japonicum USDA 110 was halted. In hydroponic culture with 2.5 µg/mL AgNPs, biomass of inoculated Glycine max (L.) Merr. was 50% of control. Axenic plants were unaffected by this dose, but growth was inhibited at higher doses, indicating that AgNPs inhibit both nodulation and growth. Nodules treated with 2.5 µg/mL …
