Modulation Of Antibacterial Activity And Cytotoxicity In Amphipathic Cyclic Peptide [R4w4] Using Histidine Substitution, Ryan Stueber
Pharmaceutical Sciences (MS) Theses
Antibiotics have been the gold standard frontline defense against bacterial infections for decades. At the same time, these infecting bacteria have continued to evolve to resist developed antibiotics in an almost endless cycle. As such, we aim to take an alternative approach utilizing antimicrobial peptides (AMPs) as a means to end this cycle. [R4W4] is among known AMPs that demonstrated antimicrobial activity against methicillin-resistant staphylococcus aureus (MRSA) with a minimum inhibitory concentration (MIC) of 2.67 μg/mL. This peptide had notably effective antimicrobial activity, especially relative to linear (R4W4). However, it displayed a concerning level of cytotoxicity; eliciting a human embryonic …

The use of insects in traditional medicine and unveiling the chemical structure of the bright pigments in butterfly wings led to the discovery of bioactive phenolic compounds in the insect bodies. These metabolites have been found not only due to the insect absorption and metabolisation of the plant-derived phenolic present in their diet, but also from the ability of insects to synthesise phenolic compounds de novo through the sclerotisation process. Plant phenolics are secondary metabolites involved in the protection of tissues against UV radiation, herbivores, and pathogens, as well as pigmentation of fruits and flowers. These bioactive compounds exhibit antioxidant, …

Terpenes are naturally occurring compounds produced by plants that are of great commercial interest in the food, agricultural, cosmetic, and pharmaceutical industries due to their broad spectra of antibacterial, antifungal, anthelmintic, membrane permeation enhancement, and antioxidant biological activities. Applications of terpenes are often limited by their volatility and the need for surfactants or alcohols to produce stable, soluble (non-precipitated) products. Yeast particles (YPs) are hollow, porous microspheres that have been used for the encapsulation of terpenes (YP terpenes) by passive diffusion of terpenes through the porous YP cell walls. We here report the development of a second generation YP encapsulated …

Nanostructured Self-Assembled Thin Films Of Cationic Bottlebrush Block Copolymers, Hathaithep Senkum
Electronic Theses and Dissertations
Biofouling which is an accumulation of small species on submerged surfaces that causes detrimental impacts on economic and environmental factors for aquaculture activities and human health. To suppress the fouling, polymeric coatings from amphiphilic block copolymers provide nanostructured surfaces and carry multiple functional groups in a molecular chain. Polymers with quaternary ammonium functional groups enable material coatings to inhibit microbial adhesion by killing bacteria, consequently prolonging the material efficiency of, for example, medical implanted devices. Herein, well-defined architectures with full-arm density of quaternary ammonium bottlebrush polymers were generated from grafting-through ring opening metathesis polymerization (ROMP). Factors such as the halide …

Antimicrobial Activity Of Silver(I) Cyanoximates Against Streptococcus Mutans Ua159 Planktonic Growth And Biofilm Formation, Sarah Hameer
Honors Program Theses
Streptococcus mutans is identified as the causative agent of human dental caries, with biofilm formation serving as its most important virulence factor. Biofilms are typically more challenging to treat than planktonic or free-floating bacteria, adding to the already challenging issue of antibiotic resistance. This has led to an increased need for the discovery of alternative antibacterial compounds other than antibiotics. Among these compounds, silver(I) cyanoximates have been demonstrated to have antibacterial activity against a variety of microorganisms, specifically those that colonize in-dwelling medical devices. To assess the antibacterial properties of two silver(I) cyanoximate compounds, silver(I) nitrosodicyanomethanide (Ag(ACO)) and silver(I)α-oximido-(2-benzoyl)acetonitrile (Ag(BCO)), …
