Evidence That Talin Alternative Splice Variants From Ciona Intestinalis Have Different Roles In Cell Adhesion, Richard H. Singiser, Richard O. Mccann
Molecular and Cellular Biochemistry Faculty Publications
BACKGROUND: Talins are large, modular cytoskeletal proteins found in animals and amoebozoans such as Dictyostelium discoideum. Since the identification of a second talin gene in vertebrates, it has become increasingly clear that vertebrate Talin1 and Talin2 have non-redundant roles as essential links between integrins and the actin cytoskeleton in distinct plasma membrane-associated adhesion complexes. The conserved C-terminal I/LWEQ module is important for talin function. This structural element mediates the interaction of talins with F-actin. The I/LWEQ module also targets mammalian Talin1 to focal adhesion complexes, which are dynamic multicomponent assemblies required for cell adhesion and cell motility. Although Talin1 is …

Lipid droplet proteins of the PAT (perilipin, adipophilin, and TIP47) family regulate cellular neutral lipid stores. We have studied a new member of this family, PAT-1, and found that it is expressed in highly oxidative tissues. We refer to this protein as "OXPAT." Physiologic lipid loading of mouse liver by fasting enriches OXPAT in the lipid droplet tissue fraction. OXPAT resides on lipid droplets with the PAT protein adipophilin in primary cardiomyocytes. Ectopic expression of OXPAT promotes fatty acid-induced triacylglycerol accumulation, long-chain fatty acid oxidation, and mRNAs associated with oxidative metabolism. Consistent with these observations, OXPAT is induced in mouse …

The Pulmonary Effects Of Intravenous Adenosine In Asthmatic Subjects, Nausherwan K. Burki, Mahmud Alam, Lu-Yuan Lee
Physiology Faculty Publications
BACKGROUND: We have shown that intravenous adenosine in normal subjects does not cause bronchospasm, but causes dyspnea, most likely by an effect on vagal C fibers in the lungs [Burki et al. J Appl Physiol 2005; 98:180-5]. Since airways inflammation and bronchial hyperreactivity are features of asthma, it is possible that intravenous adenosine may be associated with an increased intensity of dyspnea, and may cause bronchospasm, as noted anecdotally in previous reports.
METHODS: We compared the effects of placebo and 10 mg intravenous adenosine, in 6 normal and 6 asthmatic subjects.
RESULTS: Placebo injection had no significant (p > 0.05) effect …

Gene Expression Patterns That Predict Sensitivity To Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitors In Lung Cancer Cell Lines And Human Lung Tumors, Justin M. Balko, Anil Potti, Christopher Saunders, Arnold J. Stromberg, Eric B. Haura, Esther P. Black
Statistics Faculty Publications
BACKGROUND: Increased focus surrounds identifying patients with advanced non-small cell lung cancer (NSCLC) who will benefit from treatment with epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKI). EGFR mutation, gene copy number, coexpression of ErbB proteins and ligands, and epithelial to mesenchymal transition markers all correlate with EGFR TKI sensitivity, and while prediction of sensitivity using any one of the markers does identify responders, individual markers do not encompass all potential responders due to high levels of inter-patient and inter-tumor variability. We hypothesized that a multivariate predictor of EGFR TKI sensitivity based on gene expression data would offer a …

Hydrologic Properties Of Pervious Concrete, Joe D. Luck, Stephen R. Workman, Stephen F. Higgins, Mark S. Coyne
Plant and Soil Sciences Faculty Publications
Pervious concrete is concrete made by eliminating most or all of the fine aggregate (sand) in the concrete mix, which allows interconnected void spaces to be formed in the hardened product. These interconnected void spaces allow the concrete to transmit water at relatively high rates. The main objective of this project was to conduct research on the potential application of pervious concrete in agricultural settings, specifically for use in animal feed lots, manure storage pads, animal manure and bedding compost facilities, or floor systems in animal buildings. Laboratory tests were conducted on replicated samples of pervious concrete formed from two …

Best Management Practices Used By Kytc For On-Site Erosion Control, David L. Allen, Sudhir Palle
Kentucky Transportation Center Research Report
New erosion control procedures (ECP) have recently been implemented by the Kentucky Transportation Cabinet (KYTC). Previously, consultants and designers developed the ECP as a part of the design plans. In many cases these did not work in the field. The new procedures allow construction personnel to develop and implement their own ECPs on site.
This study conducted a survey of construction personnel in the KYTC to determine their level of familiarity with the new ECPs and to determine any issues that may have developed with the new procedures. The results of the survey concluded that most KYTC construction personnel were …

Acyl-coenzyme A:diacylglycerol transferase (DGAT), fatty acid synthetase (FAS), and LPL are three enzymes important in adipose tissue triglyceride accumulation. To study the relationship of DGAT1, FAS, and LPL with insulin, we examined adipose mRNA expression of these genes in subjects with a wide range of insulin sensitivity (SI). DGAT1 and FAS (but not LPL) expression were strongly correlated with SI. In addition, the expression of DGAT1 and FAS (but not LPL) were higher in normal glucose-tolerant subjects compared with subjects with impaired glucose tolerance (IGT) (P < 0.005). To study the effects of insulin sensitizers, subjects with IGT were treated with pioglitazone or metformin for 10 weeks, and lipogenic enzymes were measured in adipose tissue. After pioglitazone treatment, DGAT1 expression was increased by 33 ± 10% (P < 0.05) and FAS expression increased by 63 ± 8% (P < 0.05); however, LPL expression was not altered. DGAT1, FAS, and LPL mRNA expression were not significantly changed after metformin treatment. The treatment of mice with rosiglitazone also resulted in an increase in adipose expression of DGAT1 by 2- to 3-fold, as did the treatment of 3T3 F442A adipocytes in vitro with thiazolidinediones. These data support a more global concept suggesting that adipose lipid storage functions to prevent peripheral lipotoxicity.

Cross-linking of the B cell receptor (BCR) on the immature B lymphoma cell line BKS-2 induces growth inhibition and apoptosis accompanied by rapid down-regulation of the immediate-early gene egr-1. In these lymphoma cells, egr-1 is expressed constitutively and has a prosurvival role, as Egr-1-specific antisense oligonucleotides or expression of a dominant-negative inhibitor of Egr-1 also prevented the growth of BKS-2 cells. Moreover, enhancement of Egr-1 protein with phorbol 12-myristate 13-acetate or an egr-1 expression vector rescued BKS-2 cells from BCR signal-induced growth inhibition. Nuclear run-on and mRNA stability assays indicated that BCR-derived signals act at the transcriptional level to …

More than a decade has passed since apolipoprotein E4 (APOE-epsilon4) was identified as a primary risk factor for Alzheimer 's disease (AD), yet researchers are even now struggling to understand how the apolipoprotein system integrates into the puzzle of AD etiology. The specific pathological actions of apoE4, methods of modulating apolipoprotein E4-associated risk, and possible roles of apoE in normal synaptic function are still being debated. These critical questions will never be fully answered without a complete understanding of the life cycle of the apolipoprotein receptors that mediate the uptake, signaling, and degradation of apoE. The present review will focus …

Navigating Adolescence With A Chronic Health Condition: A Perspective On The Psychological Effects Of Hair-An Syndrome On Adolescent Girls, Kimberly K. Mcclanahan, Hatim A. Omar
Pediatrics Faculty Publications
HAIR-AN syndrome is a subphenotype of polycystic ovary syndrome and is characterized by acne, obesity, hirsutism, and acanthosis nigricans. It usually manifests in early adolescence, a time of significant developmental change in females across physical, cognitive, social, and emotional domains. We contend that adolescent development for females is difficult, even in the best of circumstances, and having a chronic health condition, like HAIR-AN syndrome, will likely impact the afflicted individual's development and psychological well-being. While many researchers have discussed the long-term health effects of HAIR-AN and similar disorders, little has been written about the potential psychological sequelae of HAIR-AN on …

A Missense Mutation In Pmel17 Is Associated With The Silver Coat Color In The Horse, Emma Brunberg, Leif Andersson, Gus Cothran, Kaj Sandberg, Sofia Mikko, Gabriella Lindgren
Veterinary Science Faculty Publications
BACKGROUND: The Silver coat color, also called Silver dapple, in the horse is characterized by dilution of the black pigment in the hair. This phenotype shows an autosomal dominant inheritance. The effect of the mutation is most visible in the long hairs of the mane and tail, which are diluted to a mixture of white and gray hairs. Herein we describe the identification of the responsible gene and a missense mutation associated with the Silver phenotype.
RESULTS: Segregation data on the Silver locus (Z) were obtained within one half-sib family that consisted of a heterozygous Silver colored stallion with 34 …

Aluminum Bioavailability From The Approved Food Additive Leavening Agent Acidic Sodium Aluminum Phosphate, Incorporated Into A Baked Good, Is Lower Than From Water, Robert A. Yokel, Rebecca L. Florence
Pharmaceutical Sciences Faculty Publications
There are estimates of oral aluminum (Al) bioavailability from drinking water, but little information on Al bioavailability from foods. Foods contribute ∼95% and drinking water 1–2% of the typical human's daily Al intake. The objectives were to estimate oral Al bioavailability from a representative food containing the food additive acidic sodium aluminum phosphate (acidic SALP), a leavening agent in baked goods. Rats were acclimated to a special diet that resulted in no stomach contents 14 h after its withdrawal. They were trained to rapidly consume a biscuit containing 1.5% acidic SALP. Oral Al bioavailability was then determined from a biscuit …
Florida East Coast Railway And The Structure Of Administrative Law, Michael P. Healy
Florida East Coast Railway And The Structure Of Administrative Law, Michael P. Healy
Law Faculty Scholarly Articles
A typical Administrative Law course presents the Supreme Court's decision in United States v. Florida East Coast Railway Co. as establishing the rule that statutory text quite close to the magic words, "on the record after opportunity for an agency hearing," is needed to trigger the Administrative Procedure Act's (APA) formal hearing requirements for a rulemaking. Florida East Coast Railway is a prime example of an underrated case because, even though the case is well known, its renown is a consequence only of its black letter rule about rulemaking procedures. Many scholars and practitioners do not appreciate the case for …
Book Review | Dan Sarooshi, International Organizations And Their Exercise Of Sovereign Powers (2005) & Margaret P. Karns & Karen A. Mingst, International Organizations: The Politics And Processes Of Global Governance (2004), Christopher G. Bradley
Law Faculty Scholarly Articles
This book review considers two books on international organizations: (1) Margaret P. Karns & Karen A. Mingst, International Organizations: The Politics and Processes of Global Governance, and (2) Dan Sarooshi, International Organizations and Their Exercise of Sovereign Powers.
The review notes several features that set the Karns & Mingst book apart from other treatments of international organizations. First is a thoroughgoing commitment to an integrated view of international organizations. The book insists (and demonstrates) that knowledge of politics, theory, and history are all indispensable to a rich understanding of the problems and processes of global governance. Second, Karns …

The laboulbenialean fungi occur throughout the world and are closely associated with a range of arthropods, including many coleopteran hosts. Throughout the summer of 2004, coccinellids were collected from a Bluegrass savanna woodland ecosystem, dominated by blue ash Fraxinus quadrangulata and Chinkapin Oak Quercus muehlenbergii, and the adults were examined for the presence of Hesperomyces virescens using binocular and scanning electron microscopy. Over 80% of adult Harmonia axyridis, a species previously reported as having a persistent association with the fungus, were infected. No significant differences were observed in incidence on male and female hosts, however, the distribution of …
