Important flavor components in Louisiana brackish water clam (Rangia cuneata) were investigated in this dissertation research. Major achievements include: (1) positive identification of the major compound responsible for the objectionable musty/earthy off-flavor in the clam, (2) development of quantitative method to compare the levels of the off-flavor compound in clam samples before and after relaying, and (3) identification of desirable characteristic clam volatile flavor components. The identification of the musty/earthy off-flavor of rangia clam was accomplished by using dynamic headspace sampling (DHS) coupled with high resolution gas chromatography/mass spectrometry (HRGC/MS) in selective ion monitoring (SIM) mode and HRGC aroma perception. …

A new method, using Fourier transform infrared spectroscopy (FTIR), was developed for the simultaneous determination of nitrogen dioxide and nitric oxide in gas phase cigarette smoke and in model systems. The FTIR method also allows the simultaneous determination of other components. The method uses multivariate least-squares regression analysis, that allows simultaneous quantitation of several components even in the presence of overlapping peaks, and fast data acquisition for kinetic analysis. For the preparation of the gas mixtures an apparatus that uses mass flow controllers, check-valves, and ground glass joints was constructed. Nitrogen Dioxide is a toxic air pollutant that exists at less than 0.5 ppm in the atmosphere. This toxic compound is known to initiate autoxidation of unsaturated fatty acids both in vivo and in vitro. When autoxidation occurs in vivo, membrane damage that can lead to cell death can occur. Low concentrations of nitrogen dioxide were shown to react with the polyunsaturated fatty acid esters, methyl linoleate and methyl linolenate, by a H-abstraction mechanism. However, methyl oleate, an unsaturated fatty acid ester, was demonstrated to react with a low concentration of nitrogen dioxide by only an addition mechanism. Although methyl …

Various aspects of chromatographic peak quantitation and shape characterization are investigated in detail for single and overlapping chromatographic peaks. From the viewpoint of providing better quantitation of real chromatographic data while minimizing computational complexity, the results presented should be easily incorporated into existing routine chromatographic data analysis regimes. Three topics applicable to modern chromatographic data analysis are considered. First, progress in the application of the exponentially modified Gaussian (EMG) function to chromatography is reviewed. Two problems of current fiber optic chemical sensors (FOCS) were addressed in this research. First, an immobilized ligand was characterized as a possible reagent for detecting anions selectively. Second, a new approach to fiber optic chemical sensing based on polymer swelling was demonstrated.
The immobilized reagent explored for selective detection of anions is marketed as tris(carboxymethyl) ethylenediaminediacetic acid (TED). Characterization of this reagent showed that it is actually a mixture of ligands with ethylenediaminediacetic acid (EDDA) being the major species. Conclusive evidence was found in the EPR spectra. The ever-increasing power of modern infrared instrumentation, coupled with the decreasing number of experienced spectroscopists has created an imbalance between information generation and interpretation capabilities. At the same time, digital computers are being developed which continue to grow in storage and processing capabilities, and shrink in cost. Clearly, the computer may serve as a valuable tool to aid the analytical chemist in interpreting spectroscopic information. This dissertation deals with the development of new approaches to exploiting computer technology to interpret infrared spectroscopic data.

In chapter One, zero angle depolarized light scattering methods for particle size determinations are demonstrated by a study on aqueous colloidal suspensions of titanium dioxide. Previous results on a poly(tetrafluoroethylene) latex suspension are also briefly recounted and updated to include reanalysis by an additional independent Laplace inversion method, Provencher's CONTIN. Size distributions may be obtained by Laplace inversion of the correlation functions, with excellent agreement among the various Laplace inversion algorithms. Resolution is greatly enhanced, since rotational motions, which are more sensitive to size than translational motions, are detected at zero angle.
