Laser Speckle Contrast Imaging (LSCI) is a non-scanning wide field-of-view optical imaging technique specifically developed for cerebral blood flow (CBF) monitoring. In this project, a versatile Laser speckle contrast imaging system has been designed and developed to monitor CBF changes and examine the physical properties of cerebral vasculature during functional brain activation experiments.

The hardware of the system consists of a high speed CMOS camera, a coherent light source, a trinocular microscope, and a PC that does camera controlling and data storage. The simplicity of the system’s hardware makes it suitable for biological experiments.

Fluorescent sensors have been widely used as microscopic tools to image Zn2+ on a cellular level. Recently, it has been established that the sensors TSQ and Zinquin form adducts with Zn-proteins and image fractions of the Zn-proteome.1 Since TSQ and Zinquin bind specifically to many Zn-proteins, it is hypothesized that other metal binding ligands, both synthetic and natural, may also bind to the Zn-proteome. This study considers a remanufactured electrical product under a tiered warranty policy. Warranty is key in ensuring a good manufacturer—consumer relationship. Manufacturers hope to minimize warranty costs while consumers believe that good warranty promises better product quality and reliability. This Thesis presents an optimal warranty period from the perspective of a manufacturer to maximize the total expected profits, while ensuring sustained consumer relation. This study investigates the interactions of various combinations of frequency and duration that result in the same duty cycle at a given applied grip force (measured in % maximum voluntary contraction (MVC)) to determine their relative effect on perceived exertion. Eight female subjects (median age 20.5 years) performed 27 randomized trials containing combinations of dynamic grips, performing each combination twice. Each session contained three 25 minute trials, with a minimum of 12 minutes of rest between trials. Interest in the development of cancer therapies with improved selectivity and reduced host toxicity has been growing. In this thesis, we designed and synthesized a series of novel non-toxic arylboronic ester and biarylboronic ester derivatives that can be activated by hydrogen peroxide (H2O2) to induce DNA interstrand cross-link formation. The mechanism of DNA cross-linking induced by these arylboronates involves generation of phenol intermediates 1 followed by departure of leaving group (L) leading to quinone methides (QMs) 2, which directly cross-link DNA via alkylation. Discharge of stormwater runoff to receiving waters is a known source of human pathogens; however the primary mechanisms by which these pathogens enter the stormwater system have yet to be quantified. This study builds upon and utilizes prior research findings in an attempt to explain the influence of the age of the pipes within stormwater and sanitary conveyance systems, rainfall and hydrogeological characteristics, and select infrastructure variables that contribute to the observed contamination of an urban stormwater basin in Southeastern Wisconsin.

Beach closures and public health protection are confounded by analytical procedures that result in delays in notification of adverse water quality conditions and the lack of affordable analytical methods to identify pollutant sources. Attempts have been made to develop predictive frameworks using ancillary hydrometeorological data to statistically anticipate deteriorated water quality. Many urban coastal beaches are impacted by river runoff. There are several occupations that require prolonged kneeling as part of their daily practice. Employees who kneel for a prolonged period of time have been reported to have high rates of knee and low back musculoskeletal disorders (Coggon et al. 2000, Halmstrom et al. 1992, Harkness et al. 2003, Manninen et al. 2002). These disorders result in excessive worker’s compensation costs. Gallagher el al. (2009) looked at the low seam mining industry and found that companies spent the most on knee related injuries with $4.2 million and $2.7 million for low back injuries.

Until recently, auditory fear extinction was not thought to modify substrates involved in the storage of the original auditory fear memory. Evidence now suggests that extinction results in the reversal of the fear conditioning-induced potentiation of thalamic inputs to the lateral amygdala. However, little is known about the molecular mechanisms that support this depotentiation of synaptic strength. The following qualitative study used semi-structured qualitative interviews to investigate the educational trajectories of five undocumented Latinos who were either in the process of transitioning into a college program or had recently enrolled in a college program. An initial interview was conducted with each of the participants followed by a follow-up interview about 3-months after the initial interview was conducted. Iodine-131 is a short-lived (half-life=8.0233 days), gamma emitting, radiopharmaceutical that, when excreted by patients, enters aquatic systems via sewage effluent discharged from water reclamation facilities (WRFs). Here, I report on 131I activities in the nearshore of southwest Lake Michigan in the vicinity of Milwaukee, Wisconsin. This is the first report on 131I activity in any of the Great Lakes of North America.

In my first essay I consider Orcutt’s (1950) hypothesis in which trade flows respond to changes in exchange rate more quickly than they do to changes in prices. There are several studies, which test the Orcutt’s hypothesis by imposing lag structure on both relative prices and exchange rate. I employ generalized impulse response analysis as an alternative approach to test Orcutt’s hypothesis using the sample of developed and developing countries. Sewage contamination from failing infrastructure and sewer overflows is a major environmental and human health concern in waterways, especially in urban communities bordering the Great Lakes such as Milwaukee, Wisconsin. Culture-based fecal indicator bacteria, such as Escherichia coli, enterococci, and fecal coliforms are traditionally used to indicate the presence of a human health risk due to fecal contamination.
