Investigating The Effects Of Subchronic Dietary Zinc Supplementation On Diet-Induced Metabolic Dysfunction-Associated Steatotic Liver Disease., Oluwanifemi Esther Bolatimi Dec 2023

Investigating The Effects Of Subchronic Dietary Zinc Supplementation On Diet-Induced Metabolic Dysfunction-Associated Steatotic Liver Disease., Oluwanifemi Esther Bolatimi

Electronic Theses and Dissertations

Zinc deficiency is associated with metabolic dysfunction-associated steatotic liver disease (MASLD). Previous studies show zinc supplementation improves steatosis, but its therapeutic effects in established MASLD remain unclear. We developed a model to characterize the effects of zinc supplementation on high-fat diet (HFD) induced MASLD and hypothesized established MASLD would be attenuated. Mice were fed control diet or HFD for 12 weeks and then grouped into normal or zinc-supplemented diets for 8 additional weeks. At euthanasia, plasma and liver tissues were collected for phenotypic analysis. Twelve weeks of HFD altered glucose clearance and body composition. Eight weeks of subsequent zinc supplementation …

Synergistic Strategies In Sinter-Based Material Extrusion (Mex) 3d Printing Of Copper: Process Development, Product Design, Predictive Maps And Models., Kameswara Pavan Kumar Ajjarapu Dec 2023

Synergistic Strategies In Sinter-Based Material Extrusion (Mex) 3d Printing Of Copper: Process Development, Product Design, Predictive Maps And Models., Kameswara Pavan Kumar Ajjarapu

Electronic Theses and Dissertations

3D printing pure copper with high electrical conductivity and exceptional density has long been challenging. While laser-based additive manufacturing technologies suffered due to copper's highly reflective nature towards laser beams, parts printed via binder-assisted technologies failed to reach over 90% IACS (International Annealed Copper Standard), electrical conductivity. Although promising techniques such as binder jetting, filament, and pellet-based 3D printing that can print copper exist, they however still face difficulties in achieving both high sintered densities and electrical conductivity values. This is due to a lack of comprehensive understanding of property evolution from green to sintered states and the strategies that …

Communicating In Crisis: Rhetorical (De)Stabilization During The Covid-19 Pandemic., Brittany Nicole Smart Dec 2023

Communicating In Crisis: Rhetorical (De)Stabilization During The Covid-19 Pandemic., Brittany Nicole Smart

Electronic Theses and Dissertations

This project explores the role of rhetoric in crisis—how rhetoric can contribute to both the stabilization and destabilization of a worldwide health emergency. Specifically, I utilize the COVID-19 pandemic as a case study to investigate how institutional rhetorics exacerbated the ongoing burnout epidemic amongst healthcare workers. Through a feminist, materialist take on institutional ethnography (Fullagar & Pavlidis, 2021; Griffith & Smith, 2014), I show how, while institutions like the CDC were under pressure to contain the spread of the virus, in the chaos of communicating safety regulations to healthcare professionals, they inadvertently subverted clinician autonomy and expertise by “coordinating” (LaFrance, …

Quantification Of Tumor Biophysical Heterogeneity Through Mechanical And Ultrastructural Analysis., Bradley James Mahaffey Dec 2023

Quantification Of Tumor Biophysical Heterogeneity Through Mechanical And Ultrastructural Analysis., Bradley James Mahaffey

Electronic Theses and Dissertations

Glioblastoma (GBM) is a highly invasive, aggressive brain cancer that carries a median survival of 15 months. This poor prognosis is due, in part, to its resistance to standard therapeutic intervention. Recent studies have demonstrated that tumor heterogeneity plays a critical role in facilitation therapy resistance by mediating tumor adaptation through microenvironmental cues. Efforts to describe these microenvironmental differences may aid in the development of strategies to combat resistance. GBM can be separated into two distinct regions – a core and a rim, which are thought to drive specific aspects of tumor evolution. The core is proliferative as evidenced by …

Working as part of an emerging category of soft robotics called Soft, Curved, Reconfigurable, Anisotropic Mechanisms, or SCRAMs, this research also introduces the Thermally-Activated SCRAM Limb (TASL) technology, which employs shape-memory alloy (SMA) wire embedded in curved sheets for surface actuation and served as the initial inspiration for OptiGap. In addition, the EneGate system is presented as a complementary …

Causal Inference For The Effect Of Continuous Treatment On Time-To-Event Outcomes And Mediation Analysis On Health Disparities In Observational Studies., Triparna Poddar Dec 2023

Causal Inference For The Effect Of Continuous Treatment On Time-To-Event Outcomes And Mediation Analysis On Health Disparities In Observational Studies., Triparna Poddar

Electronic Theses and Dissertations

The dissertation comprises two projects related to causal inference based on observational data. In healthcare research, where abundant observational data such as claims data and electronic records are available, researchers often aim to study the treatment effect and the pathway of that effect. However, estimating treatment effects in observational data presents challenges due to confounding factors. The first project focuses on estimating continuous treatment effects for survival outcomes, while the second concentrates on mediation analysis, allowing the exploration of the pathway of the causal effect. Both projects involve addressing confounding variables. In the first project, I investigate estimation of the …

Quantifying Impacts Of Climate Change On Headwater Streamflow Regime In Robinson Forest: Insights From 35-Years Of Data Collection., Lauren Brown Dec 2023

Quantifying Impacts Of Climate Change On Headwater Streamflow Regime In Robinson Forest: Insights From 35-Years Of Data Collection., Lauren Brown

Electronic Theses and Dissertations

Climate change may shift patterns of streamflow permanence in headwater systems by altering the frequency, magnitude, duration, timing, and rate of change of surface streamflow, impacting both local ecosystems as well as regional water budgets and availability. While much uncertainty surrounds modeling-based methods to quantify the impacts of climate change on water budgets, long-term hydrologic data collected from headwaters in experimental research forests serve as critical evidence to reduce such uncertainty. The objective of this study is to quantify shifts in frequency, magnitude, duration, timing, and rate of change of streamflow in two headwater catchments with relatively little recent disturbance …

Ethanol Dysregulates Sirt1-Mediated Lipogenic Signaling Pathways Through Α4*-Nicotinic Acetylcholine Receptors In Hepatocytes., Caitlin Christina Wilkerson Dec 2023

Ethanol Dysregulates Sirt1-Mediated Lipogenic Signaling Pathways Through Α4*-Nicotinic Acetylcholine Receptors In Hepatocytes., Caitlin Christina Wilkerson

Electronic Theses and Dissertations

Alcohol-associated liver disease (ALD) is a broad spectrum of diseases ranging from steatosis to severe hepatic cirrhosis, none of which have any FDA approved therapies. Downregulation of hepatic Sirtuin 1 (Sirt1) activity by ethanol has been shown to lead to an upregulation of the pro-lipogenic gene targets under its jurisdiction, resulting in increased lipogenesis and triglyceride accumulation. It was hypothesized that ethanol acts on α4*-nicotinic acetylcholine receptors (α4*-nAChRs) in hepatocytes to decrease Sirt1 expression, which ultimately dysregulates lipid metabolic homeostasis. Ethanol’s effects were investigated in cultured AML12 hepatocytes and in WT primary mouse hepatocytes acutely exposed to ethanol. Consistent with …

Genetically Engineering Gingipains For High Yield Large Scale Purification., Andrew E. Fuchs Dec 2023

Genetically Engineering Gingipains For High Yield Large Scale Purification., Andrew E. Fuchs

Effects Of A Protection Zone In A Reaction-Diffusion Model With Strong Allee Effect., Isaac Johnson Dec 2023

Effects Of A Protection Zone In A Reaction-Diffusion Model With Strong Allee Effect., Isaac Johnson

Electronic Theses and Dissertations

A protection zone model represents a patchy environment with positive growth over the protection zone and strong Allee effect growth outside the protection zone. Generally, these models are considered through the corresponding eigenvalue problem, but that has certain limitations. In this thesis, a general protection zone model is considered. This model makes no assumption on the direction of the traveling wave solution over the Strong Allee effect patch. We use phase portrait analysis of this protection zone model to draw conclusions about the existence of equilibrium solutions. We establish the existence of three types of equilibrium solutions and the necessary …

Understanding how innocuous organisms can evolve to be pathogenic to humans is of increasing global concern. Further, understanding how existing pathogens may evolved to be more virulent is also vital to our ability to provide healthcare to people afflicted with diseases that promote chronic bacterial infections, such as cystic fibrosis. With the rise of antibiotic resistance in both bacteria and fungi it is paramount that new therapeutics are identified. Understanding what mutations occur that result in increased virulence in microbes can potentially provide new targets for antimicrobial drugs to combat antibiotic resistance. The Coincidental Evolution Hypothesis is a fundamental hypothesis …

Environmental Exposures And Aging., Daniel Chris Gomes Dec 2023

Environmental Exposures And Aging., Daniel Chris Gomes

Electronic Theses and Dissertations

In recent years, research into air pollution has shown that exposure to certain components in air pollution, primarily PM2.5 can accelerate biological aging and thereby lead to increased susceptibility to multiple diseases. We hypothesize that prolonged exposure to air pollutants can result in premature aging leading to extensive tissue dysfunction and susceptibility to diseases. To examine this, we exposed mice to PM2.5 for 9, 15, and 21 days, then measured the telomere lengths, cellular senescence, and histone methylation patterns of multiple cell types. We found consistently increased telomere attrition, cellular senescence and advanced age-consistent histone methylation patterns in groups exposed …

Perovskite solar cells (PSCs) are a promising alternative to silicon-based photovoltaics. However, PSCs face several challenges due to shortcomings in their stability, module efficiency, and scaled production. Although PSCs is still a young field of research, significant attention has been given to demonstrating power conversion efficiencies that are on par with traditional silicon. With that target reached, converting the laboratory demonstration into practical materials to increase access and abundance of solar energy are among the next large targets for the field. This comes with material challenges for perovskite and their companion charge transport layers (CTLs). Among the charge transport materials …

From "Smart Talk" To "Living Well": Commonplaces And Their Role In Narratives Of Rare Disease., Caitlin E. Ray Dec 2023

From "Smart Talk" To "Living Well": Commonplaces And Their Role In Narratives Of Rare Disease., Caitlin E. Ray

Electronic Theses and Dissertations

As healthcare becomes more complex, automated, and bureaucratic, patients often suffer from a lack of resources, agency, and visibility when seeking medical care. Rhetoric and Composition, specifically the subfield of Rhetoric of Health and Medicine (RHM), is interested in studying and intervening into such issues. One way to challenge our current understanding of healthcare is to consider how the rare disease patient experience reveals the gaps, limitations, and assumptions of illness and health. I argue here that through rare diseases, rhetoricians of health and medicine can better understand the representation, advocacy, and patient experience within healthcare, and potentially lead to …

Using A Toxic Aging Coin To Assess Hexavalent Chromium-Induced Neurotoxicity., Samuel Thomas Vielee Dec 2023

Using A Toxic Aging Coin To Assess Hexavalent Chromium-Induced Neurotoxicity., Samuel Thomas Vielee

Electronic Theses and Dissertations

We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. We address this crisis using a ‘toxic aging coin’ approach; heads examines how age impacts toxicity, tails examines how chemicals accelerate aging. This thesis applies the heads side for Cr(VI)-induced neurotoxicity across ages. We …

Connections Between Polyvictimization, Daily Stress, Emotion Regulation, And Mental Health Symptoms: An Ecological Momentary Assessment Study., Zoe L. Bridges-Curry Dec 2023

Connections Between Polyvictimization, Daily Stress, Emotion Regulation, And Mental Health Symptoms: An Ecological Momentary Assessment Study., Zoe L. Bridges-Curry

Electronic Theses and Dissertations

Background and Objectives: Individuals who experience polyvictimization, or victimization across multiple life contexts, evidence particularly severe outcomes across a range of mental health diagnoses. Preliminary evidence suggests that emotion regulation difficulties and stress sensitization effects may help explain observed links between trauma exposure and mental health symptoms. However, the use of between-subjects designs to address within-subjects processes – a common approach across this literature – can result in erroneous interpretation of findings. Moreover, widespread reliance on retrospective trait measures of emotion regulation, daily stressors, and mental health symptoms have limited insight into the ways that these processes unfold in daily …
