N-Succinyl-L,L-2,6-Diaminopimelic Acid Desuccinylase (DapE) is a bacterial enzyme located in the lysine biosynthetic pathway of all Gram-negative and most Gram-positive species of bacteria including the notorious ESKAPE pathogens, or pathogenic strains of bacteria that have developed significant levels of resistance to currently available antibiotics. As a means to combat this challenge of growing antimicrobial resistance, we have identified the bacterial enzyme DapE as a conserved, novel enzymate target within these resistant bacterial strains,as DapE is responsible for the production of m-DAP and lysine which are ultimately employed in bacterial cell wall synthesis. The COMPASS-like family of lysine methyltransferases, MLR/MLX complexes, are epigenetic regulators that are essential for normal development through the methylation of the fourth lysine residue on histone 3 (H3K4), a universal epigenetic mark associated with active transcription. This family of complexes is highly conserved from yeast to mammals and the genes encoding the human MLR complexes have been associated with various developmental diseases and cancers (Dingwall and Fagan, 2019). Throughout my life as a dancer, I often wondered if body image issues were purely found in Western concert dance (such as ballet and modern dance) and if folk dance instilled a sense of ownership over one’s body. Parents play an important role guiding children’s learning of science, technology, engineering, and mathematics (STEM) in informal educational contexts. In this project, we considered the ways in which parents use autonomy supportive or controlling language to encourage or discourage children's independence in problem solving, as well as their feedback to children’s ideas and behaviors. We looked at the association between autonomy support and children’s behavioral, STEM, story, and emotional engagement during an at-home tinkering and storytelling activity. The TANK-binding kinase 1 (TBK1) is a cytoplasmic, serine/threonine kinase and a critical activator of the innate immune system. Reports have implicated TBK1 as a promoter of multiple cancer types. Acute myeloid leukemia (AML) is a cancer arising from the hematopoietic stem/progenitor cells (HSPCs). Although ~70% of AML patients achieve complete remission with chemotherapy, at least half will relapse due to the failure to eradicate leukemia stem cells (LSCs). As our previous data indicate the TLR-TAK1/TICAM-1 axes regulate LSCs, we suspect TBK1 also regulates LSCs. Repetitive mild traumatic brain injury (rmTBI) increases an individual’s risk for developing persistent deficits in recognition memory with symptoms experienced by up to 30% of rmTBI patients. Cellular senescence has been suggested as an underlying mechanism of persistent rmTBI symptoms; however, the subacute progression of cellular senescence following rmTBI has not been evaluated. Furthermore, the use of subacute testosterone administration to treat persistent recognition memory deficits and the cellular and molecular underpinnings has yet to be well characterized. SELENOF is an understudied selenium-containing protein that has previously been postulated to behave as a tumor suppressor in the breast. Examination of patient databases showed that SELENOF levels were lowest in tumors from patients with aggressive late-stage breast cancers. Whether loss of SELENOF drives breast tumorigenesis remains to be determined. To address this question, we used juvenile female wild type or systemic Selenof knockout mice and exposed them to 7,12-Dimethylbenz[a]anthracene (DMBA), a carcinogen that replicates the multistep process of breast tumorigenesis. This research explores the intersection of abortion, Medieval Christianity, and the Christian far-right, with a focus on the harms of rhetoric and the misconstruing of history. The study argues that a selective reading of history is being used by the Christian far-right to justify violence against reproductive rights activists, including police brutality and mass shootings, as well as to promote forced sterilization and other forms of reproductive control. Indigenous boarding and residential schools were in use for over 100 years in both Canada and the United States. Grown from numerous policies aimed at assimilation, dispossession, and genocide of the Indigenous peoples of North America, these residential schools had a particular aim: to wipe away all cultural and familial ties of every individual and to replace them with beliefs and values of white settlers. Though the schools are no longer in operation, their legacy of erasure and pain continues to affect survivors, their descendants, and Indigenous communities across both nations. Previous literature suggests that autistic individuals engage in physical activity less frequently compared to typically developing peers (Bandini et al., 2013; Jones et al., 2017). This is noteworthy because exercise interventions for autistic individuals have found that engaging in physical activity can improve motor, social, and behavioral skills (Anderson-Hanley et al., 2011; Duffy et al., 2017; Nicholson et al., 2011; Oriel et al., 2011). Previous research on spatial assimilation has described ethnic enclaves as places withmany recently arrived immigrants and fewer socioeconomic resources. As immigrants become more assimilated, they move to more affluent neighborhoods in proximity to Anglos. However, recent studies on resurgent ethnicity challenge the idea of the spatial assimilation by suggesting that Asian immigrants and subsequent generations continue to live near co-ethnics, despite gaining socioeconomic status. The transition from traditional ethnic enclaves to resurgent ethnic communities or ‘ethnoburbs’ indicate shifting understandings of what ethnic communities mean to Asian Americans. In confined spaces, migrating cells can undergo mesenchymal-to-amoeboid transitions by altering their cortical dynamics and adhesion with the environment. Septins frequently associate with cortical actin and non-muscle myosin (NMII), but the functional nature of these interactions remains unclear. Upon non-adhesive confinement and NMII enrichment, fibroblasts can switch to a fast, leader bleb-based mode of motility, characterized by the absence of adhesions and stress fibers and formation of a single, elongated leader bleb. Spina bifida (SB) is associated with neurologic impairments that increase individuals’ risk for neuropsychological deficits, particularly inattention and executive dysfunction. While extant literature has yet to examine the development of inattention in youth with SB, some research suggests that these youth may not experience the age-related improvements in executive dysfunction seen in the general population. Like many industries in contemporary society, the fitness sector is heavily gendered, andthus needs to be examined as a sector that both creates and reinforces gendered bodies. Building on previous single-method studies, I utilized a multi-methods approach of ethnography and interviews to analyze organizational and individual experiences with gender and fitness. Using West and Zimmerman’s concept of “doing gender,” I analyzed fitness organizations as a setting where gender and inequality are actively reproduced. Transcriptome prediction models built with data from European-descent individuals are less accurate when applied to different populations because of differences in linkage disequilibrium patterns and allele frequencies. We hypothesized multivariate adaptive shrinkage may improve cross-population transcriptome prediction, as it leverages effect size estimates across different conditions - in this case, different populations. To test this hypothesis, we made transcriptome prediction models for use in transcriptome-wide association studies (TWAS) using different methods (Elastic Net, Matrix eQTL and Multivariate Adaptive Shrinkage in R (MASHR)) and tested their out-of-sample transcriptome prediction accuracy in population-matched and cross-population scenarios. Bcl2-associated athanogene-3 (BAG3) is a pro-autophagy co-chaperone that we havepreviously shown localizes to the cardiac sarcomere and is critical for proteostasis and maintenance of normal sarcomeric function. Indeed, BAG3 loss in heart failure (HF) results in accumulation of ubiquitinated sarcomeric proteins, and depressed maximum force generating capacity (Fmax). However, how BAG3 is regulated in the cell is not well understood, with uncertainty about its structure and proteoforms. The incredible diversity within A. lentiginosus, particularly in respect to its fruit morphology, makes it an exceptional model to study adaptation and evolution. This study aimed to describe the relation between experiences of intrafamilial racism (i.e., racism perpetrated by family members against other family members), caregiver support, and identity conflict in biracial individuals. Identity conflict is defined as perceived incompatibility with two or more aspects of one’s identity (Sarno et. al., 2015) and is an experience likely to be common among biracial individuals. Pseudomonas aeruginosa is a gram-negative environmental bacterium that uses its intrinsic and acquired antibiotic resistance to infect patients with compromised epithelium, such as cystic fibrosis and burn wounds, and corneal keratitis. In order to cause disease, most strains of P. aeruginosa use the type three secretion system (T3SS) to disrupt cellular signaling and integrity. However, P. aeruginosa may also secrete exotoxins after invading host cells, which prolongs host cell death and maintains an intracellular niche.
