From gametogenesis until death an organism’s genome is under constant bombardment from endogenous and exogenous sources of DNA damage. To maintain genomic integrity amid this damage, cells have evolved responses which allow them to either preserve viability for recovery or initiate self-destructive pathways depending on the severity of DNA damage. One protein involved in initiating and carrying out these responses is the protein kinase ataxia telangiectasia and Rad3-related (ATR). ATR is known primarily for its regulatory role in initiating the checkpoint-signaling cascade following DNA damage and replicative stress. Carbon dots (CDs) have attracted much attention as an excellent gene/drug delivery and biological imaging agent for early cancer theranostics. In this study, we prepared two series of nanoparticles (NPs), which are composed of (CDs) with a targeting agent, folic acid (FA), and a chemotherapeutic agent Doxorubicin (Dox). All the NPs and their intermediates were characterized using ultraviolet-visible spectroscopy (UV-vis), fluorescence spectroscopy, and Fourier transform-infrared spectroscopy (FT-IR). The drug loading capacity (DLC) and drug loading efficiency (DLE) of two series of FA-CDs-Dox were assessed using UV-vis absorption spectroscopy at the wavelength of 485 nm. This dissertation describes my research into the involvement of phosphoserine aminotransferase 1 (PSAT1) in breast cancer progression; specifically, in triple negative breast cancer (TNBC) metastasis and endocrine resistance in estrogen receptor positive breast cancer (ER+BC). Breast cancer is the most common tumor diagnosis among women. While the overall 5-year survival for breast cancer is reaching 90%, the 5-year survival for metastatic disease is only 22%. Metastasis and endocrine resistance combined can affect over 50% of patients. Bis-thiosemicarbazones (BTSC) and their metal chelates have properties that are useful in several different scientific fields. These systems have already received attention in major fields of biology and engineering. Hydrogen evolution reaction (HER) catalysts need to be cheap and operate under minimal overpotentials with a long lifetime. The treatment of cancer requires, novel agents that have potent cytotoxic activity against cancer cells while displaying minimal side effects. Non-small cell lung carcinoma (NSCLC) comprises 85% of lung cancer diagnoses and is plagued by drug resistance. Thus, elucidating the underlying mechanisms of NSCLC is paramount to expand future treatment options. Paraoxonase 2 (PON2), an intracellular enzyme with arylesterase and lactonase functions, has well-established anti-atherosclerotic activity. Recent studies show PON2 is overexpressed in a variety of tumors and confers drug resistance, although these interactions have not been thoroughly examined in NSCLC. The purpose of this dissertation was to evaluate the impact of physical and functional limitations among older cancer survivors and its effect on healthcare utilization and mortality among them. The Medicare Current Beneficiary Survey (MCBS) is nationally representative longitudinal study of older Medicare beneficiaries. Physical Limitations (PL), Activities of Daily Living (ADL), and Instrumental Activities of Daily Living (IADL) were measured on a five-point scale. MCBS data from 2005 to 2013 were used in this study. Propensity score weighting was developed using logistic regressions. This Dissertation consists of three essays on health, labor, and public economics. The first essay investigates how gender-specific cancers affect the labor market outcomes of cancer survivors. Using a theoretical construct of labor supply and health capital, this essay exploits a nationally representative dataset, the 20082015 Medical Expenditure Panel Survey (MEPS), for estimating the correlated random effects (CRE) and over-dispersion empirical models to capture the job market effects for cancer survivors. Mammary development is a continuous and cyclic process that is under tight regulatory control from hormones and cell cycle regulators to mediate transition from the various proliferative, differential and apoptotic steps. Puberty is a time-point of high proliferation during development that has higher susceptibility to breast cancer. Spy1 is a cyclin-like protein known to regulate mammary development and increase proliferation with previous work also showing Spy1 increases tumor susceptibility and pubertal lateral side branching. ¬In this work we demonstrate that elevated levels of Spy1 in puberty significantly increase the number of lateral branches and total epithelial content in mice. Many conventional chemotherapies have indicated side effects due to a lack of treatment specificity and are thus not suitable for long-term usage. Natural health products (NHPs) are well-tolerated and safe for consumption, and some have pharmaceutical uses particularly for their anti-cancer effects.
