Adipose tissue can be characterized as either being a white (energy storing) depot or a brown (energy expending) depot and both have been found to contain dense networks of neural innervation. This adipose nerve supply regulates numerous metabolic functions and likely plays an important role in the function of adipose blood vessels. This study examines locomotor activity in samples of Frontinella pyramitela collected over its active season (April-October) to investigate whether seasonality and/or age may help explain the exceptional variability typically found in spider clock systems. Despite its noteworthy variability (%CV= 7.7), we have found that Frontinella has a mean free-running period of 28.4±2.18 hours that does not significantly vary over time. There is no correlation between day length and free-running period, indicating that varying length of FRP is not a function of photoperiod length. As lifespan is increasing globally, there is a critical need to identify strategies to extend healthspan and prevent chronic diseases into older age. The long-term goal of my research is to identify novel strategies to ameliorate aging-induced decline in hematopoietic stem cell (HSC) function. HSCs give rise to all mature blood and immune cells. With age, HSCs undergo defects in their differentiation ability which correlates with a decline in immune function. Comprehensive knowledge of gene regulatory and epigenetic mechanisms underlying this defect is a barrier to developing therapies to ameliorate aging-associated decline in HSC function. Knowing the age of an organism is essential in understanding the dynamics and management of wild populations. Age determination has been an especially long-standing challenge in the study of crustaceans, since they posed a unique challenge, shedding all calcified structures with each molt. The Jonah crab (Cancer borealis), is one of many commercially harvested crustaceans for which no absolute aging method has yet been established. Volunteering among older adults has been associated with numerous benefits for older adults and their communities. As the U.S. population continues to age, new and continued opportunities for engagement emerge not just within the formal volunteering arena, but also within paid employment, caregiving, and informal volunteering. Grounded in role theory, this study examined the extent to which current volunteers experience role conflict and role enhancement between their volunteer role and other social roles that they occupy. Inflammaging is the chronic low-grade inflammation that occurs with age that contributes to the pathology of age-related diseases. Monocytes are innate immune cells that become dysregulated with age and which can contribute to inflammaging. Metabolism plays a key role in determining immune cell functions, with anti-inflammatory cells primarily relying on fatty acid oxidation and pro-inflammatory cells primarily relying on glycolysis. It was recently shown that lipopolysaccharide (LPS)-stimulated monocytes can compensate for a lack of glucose by utilizing fatty acid oxidation. Deficits in Reelin expression and signaling play a pathogenic role in Alzheimer’s disease (AD). Thus, strategies aimed at correcting Reelin deficits may provide a novel therapeutic approach to treating AD. The cysteine-rich, whey protein supplement, Immunocal®, has recently been shown to rescue Reelin expression in a mouse model of Schizophrenia. Given that Reelin-expressing neurons of the entorhinal cortex region are a highly vulnerable population of cells that are lost early in AD, we examined the effects of Immunocal® in the hippocampal-entorhinal cortex formation in a mouse model of AD. Parental age can affect offspring fitness across taxa and through various mechanisms. However, the effect(s) of advanced maternal age on offspring, particularly in insects, has not been comprehensively reviewed making it difficult to draw conclusions about the effects of advanced maternal age on offspring in insects. In my first chapter, I reviewed maternal age literature and found overall negative effects of advanced maternal age on offspring fitness. Skeletal muscle is a highly adaptive tissue that possesses the ability to regenerate following damage. Regenerative capacity of skeletal muscle declines as age advances, leading to restricted mobility and poor quality of life. The impairment of the regeneration process in aged muscle is partly due to the accumulation of structural proteins in the extracellular matrix (ECM), termed fibrosis. In this study, two important proteins regulating ECM remodeling, plasminogen activator inhibitor-1 (PAI-1) and matrix metalloproteinase-9 (MMP-9), were investigated to determine their role in modulating changes in the ECM during aged muscle regeneration.
