Physiological And Genetic Mechanism(S) Of Cold Acclimation In Rhododendron, Chon Chong Lim Dec 1999

Physiological And Genetic Mechanism(S) Of Cold Acclimation In Rhododendron, Chon Chong Lim

Graduate Theses, Dissertations, and Problem Reports

An in vitro laboratory protocol for assessing cold hardiness (CH) in rhododendrons based on electrolyte-leakage was developed. The raw leakage data were transformed into percent-adjusted injury, which showed high correlation with injury data based on visual observations. The percent-adjusted injury data---fitted to the Gompertz function coupled with the Jackknife method---allowed for reliable and statistically sound estimations of Rhododendron leaf freezing-tolerance (LFT). Utilizing this method, Tmax values for Rhododendron cultivars, species, and populations were determined and ranked relatively. Our observation of a continuous distribution of LFT in F2 and backcross (BC) populations derived from a R. catawbiense x R. fortunei cross …

Occurrence, Etiology And Management Of Ringwomb In Ewes, Nancy Jean Kerr Aug 1999

Occurrence, Etiology And Management Of Ringwomb In Ewes, Nancy Jean Kerr

Graduate Theses, Dissertations, and Problem Reports

Pedigrees of ewes with Ringwomb were studied to determine the etiology, incidence and treatment of Ringwomb. Purebred Suffolk and crossbred Hampshire and Shropshire ewes aged from one to seven years were observed over a period of five lambing seasons. The study was conducted on 24 clinical cases of ringwomb occurring in one flock. Ringwomb or incomplete cervical dilation accounted for an incidence of 20.5% (24/117) of all ewes lambing. The ewes affected were all daughters, granddaughters, or great-granddaughters of one particular sire used on the farm as a service sire. Because ringwomb does not typically affect ewes in two consecutive …

Regulation Of The Synthesis And Protein Stability Of The Alternative Sigma Factor Rpos In Salmonella Typhimurium, Christofer Lee Cunning May 1999

Regulation Of The Synthesis And Protein Stability Of The Alternative Sigma Factor Rpos In Salmonella Typhimurium, Christofer Lee Cunning

Graduate Theses, Dissertations, and Problem Reports

RpoS is an alternative sigma factor which is required for the expression of genes that help bacteria cope with environmental stress. The regulation of RpoS is multifactorial and occurs at the levels of synthesis and protein turnover. Our lab has previously identified a rpoS translation enhancing factor in Salmonella typhimurium, the RNA-binding protein HF-I. An antisense element consisting of rpoS mRNA secondary structure is predicted to sequester the rpoS ribosome binding site and inhibit translation. HF-I may interact with this element to increase translation. Using rpoS genetic fusions, we have shown that native rpoS promoters can be substituted with tac …

Differences In Lipoxygenase Activity As Related To Cultivar Differences In Superficial Scald Of Apples, Judith Lynne Hercules Reynolds May 1999

Differences In Lipoxygenase Activity As Related To Cultivar Differences In Superficial Scald Of Apples, Judith Lynne Hercules Reynolds

Graduate Theses, Dissertations, and Problem Reports

The relationship between lipoxygenase activity (LOX) and superficial scald was studied in the four apple cultivars (Malus domesticus Borkh.), Granny Smith (GS), Red Delicious (RD), Golden Delicious (GD) and York (YK) to determine if LOX is the genetic source of superficial scald or a means of predicting % scalded fruit. For 1993, three harvests (five trees each) for GS and four harvests (five trees each) for RD, GD and YK of one bushel box each were picked at one week intervals. For 1994, there were three harvests (five trees each) for GS, RD and GD and four harvests (three trees …

Regulation Of Heme Biosynthesis Targets The Key Enzyme Hema By A Mechanism Of Protein Stabilization In Salmonella Typhimurium, Liying Wang May 1999

Regulation Of Heme Biosynthesis Targets The Key Enzyme Hema By A Mechanism Of Protein Stabilization In Salmonella Typhimurium, Liying Wang

Graduate Theses, Dissertations, and Problem Reports

Heme serves as a cofactor of cytochromes and catalases. It is essential for energy generation and in defense against toxic hydrogen peroxide in nearly all cells including Salmonella typhimurium and Escherichia coli. Indirect evidence has suggested that heme synthesis is a regulated process. Little is known about how heme synthesis is regulated in enteric bacteria even though the heme synthetic pathway is genetically well-defined. This research represents the first report that heme synthetic regulation affects the first committed heme pathway enzyme, glutamyl-tRNA reductase (HemA), by an unusual mechanism.;HemA, encoded by hemA gene, catalyzes the rate-limiting step of heme biosynthesis. This …
