African Americans are disproportionately impacted by HIV compared to other racial groups. Stigma surrounding HIV has created challenges for effective HIV prevention and stigma reduction programs. The African American church has been criticized for its slow and stigmatizing response to the HIV epidemic. Despite this, African American faith leaders play significant roles in guiding the perspective of the community. Numerous studies examine African American pastors’, faith leaders’, and ministers’ perspective about HIV and other HIV-related topics. There are roughly 340,000 adults and children living with HIV. Despite the progress in the global response and the lowing of infections, HIV rates in Ghana remain stable. Cultural and societal norms that exist at a given time and place influence masculine socialization and behavior. These norms promote heterosexual men’s engagement in high-risk sexual behaviors which result in HIV infections. Factors such as HIV knowledge, risk perception, male condom use, HIV testing, and counseling which minimize the risk of HIV, are also influenced by these norms. This dissertation examined and provided basis for addressing factors related to HIV pre-exposure prophylaxis (PrEP) engagement, outreach, and uptake among high-risk African American youth groups in Louisville, KY. This qualitative dissertation study was designed through the lens of an interpretive framework of social constructivism which holds that multiple realities and interpretations exist and are socially constructed through the lived experiences of individuals and their interactions with others. Human immunodeficiency virus-1 (HIV-1) and herpes simplex virus 2 (HSV-2) affect hundreds of millions of people worldwide, with women disproportionately impacted by these infections. Currently, only oral pre-exposure prophylaxis (PrEP) is approved specifically for the prevention of HIV-1, but is challenged with adverse side effects associated with long-term use. Topical delivery platforms, such as gels and films, deliver agents directly to the female reproductive tract, but are limited in providing transient-release. The technology of polymeric electrospun fibers may serve as alternative topical delivery platform to the female reproductive tract. This thesis describes the development of a novel lectin-Fc fusion protein and its antiviral and anti-cancer activity. The molecule, Avaren-Fc (AvFc), is a fusion of a variant of the actinomycete lectin actinohivin (Avaren) and the Fc region of human IgG1, and is selective for the terminal α1,2-mannose residues found at the ends of high-mannose-type glycans that can be found on the surface of certain heavily glycosylated viruses and cancer cells. Here, AvFc was found to be able to neutralize simian immunodeficiency virus as well as Hepatitis C virus with nanomolar IC50 values. Furthermore, AvFc recognizes a number of cell …

Sources warn the global HIV/AIDS epidemic will not be halted without significant reductions in Intimate partner violence (IPV). South African women have been hardest hit by the HIV-IPV link, suffering deleterious health outcomes as sequelae of significant barriers to HIV care. Extant literature focuses on HIV prevention, and there is a paucity of effective methods for mitigating IPV-related barriers among women living with HIV. Over the past thirty years, HIV has become a worldwide epidemic. Given the surge in the rate of sexually transmitted diseases including HIV, this sheds light as a new worry for sexually active older adults, raising questions about risk behaviors among this group. Griffithsin (GRFT) is a protein derived from the red alga Griffithsia sp. off the coast of New Zealand. It binds and inactivates number of enveloped viruses including HIV-1. For a product to be developed as a microbicide, it must be safe and effective after repeated use. For initial in vivo safety testing we exposed mice repeatedly to various vaginal products and found that increased concentration of mouse serum albumin (MSA) in the vaginal lumen is predictive of epithelial damage. Repeated intravaginal administration of GRFT did not result in increased amounts of MSA, but the known epithelial disrupters nonoxynol-9 and benzylkonium …

Griffithsin (GRFT) is a carbohydrate binding protein derived from the red alga Griffithisia sp. that has been shown to effectively interfere with the cellular attachment and infectivity of a number of viruses, including HIV-1. However, it is currently unknown what impact systemic GRFT administration may have upon a biological system. For initial in vivo testing, we introduced GRFT into guinea pigs (Cavia Parcel/us) via subcutaneous injection. Based on histological, serological, and biochemical data derived from these experiments, GRFT is well tolerated and maintained at physiologically relevant concentrations while retaining its potent antiviral activity.
