Apolipoprotein E ɛ4 allele is a common susceptibility gene for late-onset Alzheimer's disease. Brain vascular and metabolic deficits can occur in cognitively normal apolipoprotein E ɛ4 carriers decades before the onset of Alzheimer's disease. The goal of this study was to determine whether early intervention using rapamycin could restore neurovascular and neurometabolic functions, and thus impede pathological progression of Alzheimer's disease-like symptoms in pre-symptomatic Apolipoprotein E ɛ4 transgenic mice. Using in vivo, multimodal neuroimaging, we found that apolipoprotein E ɛ4 mice treated with rapamycin had restored cerebral blood flow, blood–brain barrier integrity and glucose metabolism, compared …

The presently-disclosed subject matter includes thermophilic glucan phosphatase polypeptides. In some embodiments the polypeptide includes non-native laforin polypeptides, or fragments and/or variants thereof, and in some instances the polypeptide can alter the biophysical properties of starch in vitro or in planta. The presently-disclosed subject matter also includes isolated polynucleotides encoding the present polypeptides, methods for processing starch by exposing starch to the present polypeptides, and methods for making the present polypeptides.

Cellular mechanisms that achieve protein diversity in eukaryotes are multifaceted, including transcriptional components such as RNA splicing. Through alternative splicing, a single protein-coding gene can generate multiple mRNA transcripts and protein isoforms, some of which are tissue-specific. We have conducted qualitative and quantitative analyses of the Bodymap 2.0 messenger RNA-sequencing data from 16 human tissue samples and identified 209,363 splice junctions. Of these, 22,231 (10.6%) were not previously annotated and 21,650 (10.3%) were expressed in a tissue-restricted pattern. Tissue-restricted alternative splicing was found to be widespread, with approximately 65% of expressed multi-exon genes containing at least one tissue-specific splice junction. …

Consequences Of Shifts In Abundance And Distribution Of American Chestnut For Restoration Of A Foundation Forest Tree, Harmony J. Dalgleish, Charles Dana Nelson, John A. Scrivani, Douglass F. Jacobs

Restoration of foundation species, such as the American chestnut (Castanea dentata) that was devastated by an introduced fungus, can restore ecosystem function. Understanding both the current distribution as well as biogeographic patterns is important for restoration planning. We used United States Department of Agriculture Forest Service Forest Inventory and Analysis data to quantify the current density and distribution of C. dentata. We then review the literature concerning biogeographic patterns in C. dentata. Currently, 431 ± 30.2 million stems remain. The vast majority (360 ± 22 million) are sprouts < 2.5 cm dbh. Although this number is approximately 10% of the estimated pre-blight population, blight has caused a major shift in the size structure. The current-day population has a larger range, particularly west and north, likely due to human translocation. While climate change could facilitate northward expansion, limited seed reproduction makes this unlikely without assisted migration. Previous research demonstrates that the current, smaller population contains slightly higher genetic diversity than expected, although little information exists on biogeographic patterns in the genetics of adaptive traits. Previously, we reported the effects of fescue toxicosis on developing Angus-cross steer growth, carcass, hepatic mRNA, and protein expression profiles of selected serum proteins, and blood clinical and chemical profiles, after summer-long grazing (85 days) of high endophyte (HE)- vs. low endophyte (LE)-infected fescue pastures. We now report the temporal development of acute, intermediate, and chronic responses of biochemical and clinical blood analytes determined at specified time intervals (period 1, day 0–36; period 2, day 37–58; and period 3, day 59–85). We present an analysis of the nuclear infrared (IR, 1.6–18 μm) emission of the ultraluminous IR galaxy UGC 5101 to derive the properties of its active galactic nucleus (AGN) and its obscuring material. We use new mid-IR high angular resolution (0.3–0.5 arcsec) imaging using the Si-2 filter (λC = 8.7 μm) and 7.5–13 μm spectroscopy taken with CanariCam (CC) on the 10.4 m Gran Telescopio CANARIAS. We also use archival Hubble Space Telescope/NICMOS and Subaru/COMICS imaging and Spitzer/IRS spectroscopy. We present the second extensive study of the coronal line variability in an active galaxy. Our data set for the well-studied Seyfert galaxy NGC 5548 consists of five epochs of quasi-simultaneous optical and near-infrared spectroscopy spanning a period of about five years and three epochs of X-ray spectroscopy overlapping in time with it. Whereas the broad emission lines and hot dust emission varied only moderately, the coronal lines varied strongly. However, the observed high variability is mainly due to a flux decrease. Mapping Nearby Galaxies at Apache Point Observatory (MaNGA), one of three core programs in the Sloan Digital Sky Survey-IV, is an integral-field spectroscopic survey of roughly 10,000 nearby galaxies. It employs dithered observations using 17 hexagonal bundles of 2'' fibers to obtain resolved spectroscopy over a wide wavelength range of 3600–10300 Å. To map the internal variations within each galaxy, we need to perform accurate spectral surface photometry, which is to calibrate the specific intensity at every spatial location sampled by each individual aperture element of the integral field unit. An electronic, resistance-based sensor array and data acquisition system was developed to measure spray deposition from hydraulic nozzles. The sensor surface consisted of several parallel tin plated copper traces of varying widths with varying gap widths. The system contained an embedded microprocessor to monitor output voltage corresponding to spray deposition every second. In addition, a wireless module was used to transmit the voltage values to a remote laptop. Tests were conducted in two stages to evaluate the performance of the sensor array in an attempt to quantify the spray deposition. Mycobacterium marinum infection has been historically associated with exposure to aquariums, swimming pools, fish, or other marine fauna. We present a case of M marinum left wrist tenosynovitis and elbow bursitis associated with a puncture injury and exposure to coal mine water in Illinois.

The STAR Collaborationpresents for the first time two-dimensional di-hadron correlations with identified leading hadrons in 200 GeV central Au+Au and minimum-bias d+Au collisions to explore hadronization mechanisms in the quark gluon plasma. The enhancement of the jet-like yield for leading pions in Au + Au data with respect to the d + Au reference and the absence of such an enhancement for leading non-pions (protons and kaons) are discussed within the context of a quark recombination scenario. The correlated yield at large angles, specifically in the ridge region, is found to be significantly higher for leading non-pions than pions. …

Removal of terminal buds (topping) and control of the formation of axillary shoots (suckers) are common agronomic practices that significantly impact the yield and quality of various crop plants. Application of chemicals (suckercides) to plants following topping is an effective method for sucker control. However, our current knowledge of the influence of topping, and subsequent suckercide applications, to gene expression is limited. We analyzed the differential gene expression using RNA-sequencing in tobacco (Nicotiana tabacum) that are topped, or treated after topping by two different suckercides, the contact-localized-systemic, Flupro® (FP), and contact, Off-Shoot-T®. Background: The Affordable Care Act created new incentives for hospitals, insurers, public health agencies, and others to contribute to disease prevention and health promotion activities, potentially changing inter-organizational relationships and expanding implementation of strategies that improve population health. This study uses data from the 1998-2014 National Longitudinal Survey of Public Health Systems to examine: (1) the extent and nature of change in inter-organizational contributions to public health activities; (2) whether network changes attenuate or exacerbate disparities in public health implementation across communities; and (3) how network changes affect preventable mortality and resource use.

We use cosmological adaptive mesh refinement code enzo zoom-in simulations to study the long-term evolution of the collapsing gas within dark matter haloes at z. This direct collapse process is a leading candidate for rapid formation of supermassive black hole (SMBH) seeds. To circumvent the Courant condition at small radii, we apply the sink particle method, focusing on evolution on scales ∼0.01–10 pc. The collapse proceeds in two stages, with the secondary runaway happening within the central 10 pc. The sink particles form when the collapsing gas requires additional refinement of the grid size at the highest refinement level. …

Sr2IrO4 is a magnetic insulator assisted by strong spin-orbit coupling (SOC) whereas Sr2RuO4 is a p-wave superconductor. The contrasting ground states have been shown to result from the critical role of the strong SOC in the iridate. Our investigation of structural, transport, and magnetic properties reveals that substituting 4dRu4+(4d4) ions for 5dIr4+(5d5) ions in Sr2IrO4 directly adds holes to the t2g bands, reduces the SOC, and thus rebalances the competing energies in single-crystal Sr …

The cold boroncarbide free radical (BC X 4Σ−) has been produced in a pulsed discharge free jet expansion using a precursor mixture of trimethylborane in high pressure argon. High resolution laser induced fluorescencespectra have been obtained for the B 4Σ−–X 4Σ− and E 4Π–X 4Σ− band systems of both 11BC and 10BC. The perception of a stimulus can be influenced by previous perceptual experience, a phenomenon known as perceptual priming. However, there has been limited investigation on perceptual priming of shape perception of three-dimensional object structures defined by moving dots. Here we examined the perceptual priming of a 3D object shape defined purely by motion-in-depth cues (i.e., Shape-From-Motion, SFM) using a classic prime-target paradigm. The results from the first two experiments revealed a significant increase in accuracy when a "cloudy" SFM stimulus (whose object structure was difficult to recognize due to the presence of strong noise) was preceded by an unambiguous SFM …

Silicon is capable of delivering a high theoretical specific capacity of 3579 mAh g−1 which is about 10 times higher than that of the state-of-the-art graphite based negative electrodes for lithium-ion batteries. However, the poor cycle life of silicon electrodes, caused by the large volumetric strain during cycling, limits the commercialization of silicon electrodes. As one of the essential components, the polymeric binder is critical to the performance and durability of lithium-ion batteries as it keeps the integrity of electrodes, maintains conductive path and must be stable in the electrolyte. Renal failure after left ventricular assist device (LVAD) implantation occurs either due to worsening chronic kidney disease which is common in end-stage heart failure, or due to acute kidney injury in the peri-implantation period, and is associated with high morbidity and short-term mortality. The increased utilization of LVAD in refractory heart failure either as a bridge to transplantation or destination therapy will eventually create a population of patients with LVADs who are dialysis-dependent. The Auca Mahuida volcano (2.03–0.88 Ma) located east of the Andean thrust front in the Neuquén basin (Argentina) hosts an oil system of thermogenic origin and is affected by the NW–SE striking-faults. Intrusive bodies and the underlying Jurassic sediments constitute the reservoir rocks. Aeromagnetic data collected in the Auca Mahuida area detected multiple dipolar magnetic anomalies, many of which have reverse polarity. Palaeomagnetic measurements on rock samples collected in the field together with available age determinations indicate that the reversely magnetized sources were mainly emplaced during the Matuyama reverse polarity chron while the normal polarity sources were emplaced during the …

We report the observation of transverse polarization-dependent azimuthal correlations in charged pion pair production with the STAR experiment in p↑+p collisions at RHIC. These correlations directly probe quark transversity distributions. We measure signals in excess of 5 standard deviations at high transverse momenta, at high pseudorapidities η>0.5, and for pair masses around the mass of the ρ meson. This is the first direct transversity measurement in p+p collisions.

Reverse transcriptase-quantitative polymerase chain reaction (RT-qPCR) is a reliable technique for quantifying gene expression across various biological processes, of which requires a set of suited reference genes to normalize the expression data. Coleomegilla maculata (Coleoptera: Coccinellidae), is one of the most extensively used biological control agents in the field to manage arthropod pest species. In this study, expression profiles of 16 housekeeping genes selected from C. maculata were cloned and investigated. For a formation 𝔉, a subgroup U of a finite group G is said to be 𝔉-pronormal in G if for each g ∈ G, there exists x ∈ ⟨U, Ug⟩ 𝔉 such that Ux = Ug. If 𝔉 contains 𝔑, the formation of nilpotent groups, then every 𝔉-pronormal subgroup is pronormal and, in fact, 𝔑-pronormality is just classical pronormality. The main aim of this paper is to study classes of finite soluble groups in which pronormality and 𝔉-pronormality coincide.

H1 linker histones are highly abundant proteins that compact nucleosomes and chromatin to regulate DNA accessibility and transcription. However, the mechanisms that target H1 regulation to specific regions of eukaryotic genomes are unknown. Here we report fluorescence measurements of human H1 regulation of nucleosome dynamics and transcription factor (TF) binding within nucleosomes. H1 does not block TF binding, instead it suppresses nucleosome unwrapping to reduce DNA accessibility within H1-bound nucleosomes. We then investigated H1 regulation by H3K56 and H3K122 acetylation, two transcriptional activating histone post translational modifications (PTMs). Background: United States (US) epidemiological studies suggest that for every 5-8 who start drinking alcoholic beverages, at least one drinker will develop an alcohol dependence (AD) syndrome within the first 10 years after onset of drinking (Lopez-Quintero et al., 2011; Wagner & Anthony, 2002). Recently, we described a multiparametric functional analysis approach for new research to estimate these transition probabilities with a one-dimensional function (1D; Vsevolozhskaya & Anthony, 2015). Here, we demonstrate extension of this analysis to two-dimensional (2D) functions that combine information about number of recent drinking days and number of drinks on the typical drinking day.

A novel computational method and generation of mutant butyrylcholinesterase for cocaine hydrolysis is provided. The method includes molecular modeling a possible BChE mutant and conducting molecular dynamics simulations and hybrid quantum mechanical/molecular mechanical calculations thereby providing a screening method of possible BChE mutants by predicting which mutant will lead to a more stable transition state for a rate determining step. Site-directed mutagenesis, protein expression, and protein activity is conducted for mutants determined computationally as being good candidates for possible BChE mutants, i.e., ones predicted to have higher catalytic efficiency as compared with wild-type BChE. Both epidemiological and experimental studies suggest that ethanol may enhance aggressiveness of breast cancer. We have previously demonstrated that short term exposure to ethanol (12–48 hours) increased migration/invasion in breast cancer cells overexpressing ErbB2, but not in breast cancer cells with low expression of ErbB2, such as MCF7, BT20 and T47D breast cancer cells. In this study, we showed that chronic ethanol exposure transformed breast cancer cells that were not responsive to short term ethanol treatment to a more aggressive phenotype. BACKGROUND: Leishmania major infection induces robust interleukin-12 (IL12) production in human dendritic cells (hDC), ultimately resulting in Th1-mediated immunity and clinical resolution. The surface of Leishmania parasites is covered in a dense glycocalyx consisting of primarily lipophosphoglycan (LPG) and other phosphoglycan-containing molecules (PGs), making these glycoconjugates the likely pathogen-associated molecular patterns (PAMPS) responsible for IL12 induction.

A mixed ionic–electronic conductor based on nanofibrillated cellulose composited with poly(3,4-ethylene-dioxythiophene):poly(styrene-sulfonate) along with high boiling point solvents is demonstrated in bulky electrochemical devices. The high electronic and ionic conductivities of the resulting nanopaper are exploited in devices which exhibit record values for the charge storage capacitance (1F) in supercapacitors and transconductance (1S) in electrochemical transistors.
