Belize is a food insecure, developing country. Although Belize has plenty of nutritious food in-country, the exportation of that food to generate income contributed to a distribution issue of that nutritious food. More impoverished areas of Belize do not have access to nutritious food because of the lack of affordability and high export rates. Diets with a lack of food, or lack of balanced, nutritious food can negatively impact growth and cognitive development, especially in children. In 2019, around 40% of school-aged children in Arkansas were classified as overweight (~17%) and obese (~23%), according to BMI data collected. Children with obesity have an increased risk of developing type 2 diabetes, hypertension, and dyslipidemia. Unhealthy eating habits, such as skipping breakfast, have been strongly associated with overeating, weight gain and obesity. Breakfast is a key component of a healthy diet and can positively impact children’s health and well-being. However, there has been a steady decline in breakfast consumption in US children over the past 40 years. Soybean [Glycine Max (L.) Merrill] is an important economic crop because of its high content of seed protein and oil. Seed oil and protein content are quantitative inherited traits. The genetics of seed protein and oil levels have been extensively studied, with 367 QTL reported for protein and 475 QTL reported for oil to date. Validation of such QTLs, and identification of easily-automatable molecular markers around these QTL will aid the progression of breeding for such traits. The shape and charge of a protein play significant roles in protein dynamics in the biological system of humans and animals. Characterizing and quantifying the shape and charge of a protein at the single-molecule level remains a challenge. Solid-state nanopores made of silicon nitride (SiNx) have emerged as novel platforms for biosensing such as diagnostics for single-molecule detection and DNA sequencing. SSN detection is based on measuring the variations in ionic conductance as charged biomolecules translocate through nanometer-sized channels driven by an external voltage applied across the membrane. Single Molecule Fluorescence Studies Of Protein Structure And Dynamics Underlying The Chloroplast Signal Recognition Particle Targeting Pathway, Dustin R. Baucom

The work presented in this dissertation explores the structural dynamics in the chloroplast signal recognition particle pathway. Findings include cpSRP shows scanning functionality similar to that in the cytosolic SRP with the ribosome. The intrinsically disordered C-terminal tail of the Albino3 protein has some transient secondary structure. Upon binding to cpSRP43 in solution, separate secondary structure formation was identified in the C-terminal tail of Albino3. Finally, to increase efficiency of analyzing fluorescence time traces for this work, a modular software was produced.

Soy protein isolate (SPI) and probiotics can serve as a functional ingredient and beneficial supplement, respectively, to food formulations, but few studies focus on the compatibility of these two ingredients. The objective of this study was to prepare soy protein isolate (SPI) from defatted soybean flour and identify an optimal hydrolysis protocol to create soy protein hydrolysates (SPH) with improved solubility and functional properties and achieve an optimum encapsulation technique for probiotics and to determine the in vitro viability of encapsulated probiotic cells with SPH in a gastrointestinal environment. Introduction: Blood pressure is a critical indicator for a number of health risks and conditions, such as hypertension (high blood pressure), heart failure, and stroke. Decreasing blood pressure occurs by vasodilation (relaxation) of blood vessels. The consumption of high amounts of protein is hypothesized to have a correlation with the vasodilation of vessels, thus creating a decrease in blood pressure. PURPOSE: The purpose of this study was to investigate the effects of 12 weeks of protein supplementation and Autoregulatory Progressive Resistance Exercise (APRE) training on measures of body composition, muscular performance, and free testosterone in middle-aged men. METHODS: Untrained males (n = 35; 43.3 ± 6.6 years) participated in this study. Lean mass (LM) and fat mass (FM) were measured via Dual Energy X-ray Absorptiometry (DEXA), while strength was measured utilizing one repetition maximum (1RM), endurance measured using maximum repetitions completed (REPS) at 75% 1RM for the bench and leg press, and free testosterone was measured using changes in serum …

Biotherapeutic drugs, derived from biological molecules such as proteins and DNA, are becoming an integral and exceptionally critical aspect of modern medicine. Compared to common pharmaceutical drugs, biotherapeutics are much larger in size and have greater target specificity, allowing them to treat many chronic diseases ranging from cancer to rheumatoid arthritis. The major issue with protein based therapeutics is that they readily undergo proteolysis, or enzymatic degradation, when administered through subcutaneous injections. Protein targeting is a fundamental cellular process that directs proteins from their site of synthesis to the site where they function. The signal recognition particle (SRP) dependent targeting pathway is conserved in both eukaryotes and prokaryotes where it co-translationally targets polypeptide chains emerging from ribosomes to the endoplasmic reticulum (eukaryotes) or cytoplasmic membrane (prokaryotes). A structurally unique form of SRP is found in chloroplasts where it functions to post-translationally bind and target a subset of integral thylakoid membrane proteins, the light harvesting chlorophyll binding proteins (LHCPs). The Ras superfamily of G-proteins are of great research interest for structure-function relationships among proteins as they act as molecular switches in the regulation of various biochemical reactions in the cell. They are regulated by protein-protein interactions targeted to the highly flexible switch regions. Protein and oil are the major chemical constituents of soybean seed that affect the quality of soyfood, feed, and oil products. Therefore, soybean cultivars with high protein and/or high oil are desirable for the soyfood and feed markets. Rising feed costs, environmental pollution concerns targeted at animal agriculture, a worldwide focus on sustainability and the never ending battle to improve production and efficiency within the poultry industry challenges nutritionists to focus on products to improve nutrient digestibility and decrease feed costs while at the same time maximizing performance. The research presented in this dissertation focuses on improving nutrient utilization of protein sources with protein liberating enzymes currently available commercially. The research also conducted addresses the addition of crystalline amino acids valine and isoleucine and their effect on improving efficiency of broilers.
