Zebrafish Electroretinogram Responses, Brooke Campbell Jan 2024

Zebrafish Electroretinogram Responses, Brooke Campbell

Williams Honors College, Honors Research Projects

The goal of this project is to be able to streamline a protocol for conducting ERGs on zebrafish from mice ERG protocol already present in Dr. Renna’s lab. The objective is to create a protocol specifically for zebrafish and to eliminate any issues that occur when transiting from a mice ERG protocol to a zebrafish ERG protocol and to obtain a light response from zebrafish retinae in differing light intensities. Dr. Renna’s lab has designed an Ex Vivo ERG system with less electrical interference creating defined electrical responses from mouse retina. The setup allows for continual perfusion of the retinal …

Tropical Fish Study In Tahiti, French Polynesia, Miranda Brainard, Caitlyn Swango, Paityn Houglan, Richard Londraville

Williams Honors College, Honors Research Projects

In May of 2023, I embarked on an exciting research journey to Moorea, French Polynesia, alongside fellow students and faculty members from the University of Akron and Syracuse University. This expedition was part of the university-sponsored Tropical Vertebrate Biology course, where we delved into the exploration of various tropical species inhabiting the island, including sea urchins, geckos, and my primary focus, the blackspotted rockskipper.

My research team, composed of my co-authors and me, was particularly intrigued by the unique refuge-seeking behavior displayed by blackspotted rockskippers. These amphibious fish are renowned for their remarkable ability to inhabit tide pools and rocky …

Perceptions Regarding Drinking Water Quality And Its Effects On Human And Animal Health Among Plain-Sect Community Members, Anil Kumar Chaudhary, Parmveer Singh, Sharmistha Basak Dec 2023

Perceptions Regarding Drinking Water Quality And Its Effects On Human And Animal Health Among Plain-Sect Community Members, Anil Kumar Chaudhary, Parmveer Singh, Sharmistha Basak

Journal of Amish and Plain Anabaptist Studies

The quality of drinking water can affect human and animal health, and plain-sect populations may be more susceptible than other rural populations due to their use of traditional farm management practices and their reliance on well water. Therefore, an interdisciplinary team conducted a pilot study to understand the status of existing drinking water quality, community perceptions regarding causes of water deterioration, its associated effect on human and animal health, and solutions to address such challenges. The study included water testing and a focus group discussion with plain-sect community members. The findings revealed that participants perceived the drinking water quality as …

A Short History Of The Holmes County, Ohio, Amish Directory And Genetics Research, Harold Cross Jun 2023

A Short History Of The Holmes County, Ohio, Amish Directory And Genetics Research, Harold Cross

Journal of Amish and Plain Anabaptist Studies

In this first-hand account, Harold “Hal” Cross, a researcher instrumental in the dawning of Amish genetic/demographic studies, recounts his memories of early projects and the development of the first Amish directories. Interest in Amish genetics emerged in the 1960s, as Cross, then a graduate student at Johns Hopkins University School of Medicine, informed Victor A McKusick, Professor of Medicine, about the potential for studying genetic disease among the Amish. Cross, working under McKusick’s direction, then collaborated with Amishman Ervin Gingerich of Holmes County, OH, in completing the first full-size household-level Amish directory in 1965. The effort was financed in part …

Effect Of An Oncogenic Her2 Mutant On Egfr Activity, Paige Kalik Jan 2023

Effect Of An Oncogenic Her2 Mutant On Egfr Activity, Paige Kalik

Williams Honors College, Honors Research Projects

Studies have shown that HERs are involved in the progression of human cancers. Researchers have found that HER2 mutations in colorectal cancer are similar to mutations found in breast cancer. Recombinant antibodies are effective therapeutic treatments for cancers. Antibodies can also be used to determine how proteins function under normal and pathophysiological conditions while also identifying interacting partners. In this Honors Project, my aim is to investigate the effect of an oncogenic mutant of HER2. Considering the information above, the question researched in this project was: how does the S310F mutation affect the organization and function of HER2? The specific …

The Accuracy Of Recreational Athletes In Self Detecting Ventilation Threshold During A Maximal Exercise Test, Andrew Biegner Jan 2023

The Accuracy Of Recreational Athletes In Self Detecting Ventilation Threshold During A Maximal Exercise Test, Andrew Biegner

Williams Honors College, Honors Research Projects

INTRODUCTION: Ventilatory threshold (VT) is where minute ventilation (VE) increases nonlinearly with increasing exercise intensity. Prior studies have shown that subjects can recognize VT by changes in breathing during exercise. Teaching recreational athletes to use VT as a training method could result in better quality training. PURPOSE: Determine if recreational athletes can accurately perceive changes in ventilation associated with VT during maximal exercise testing. METHODS: Subjects were recruited (n=20, age= 20.5 ± 1.7 yrs.) to participate in the study. Subjects performed a modified maximal treadmill protocol with gas analysis. Subjects indicated when they noticed a considerable change …

The Effect Of Alpha Lipoic Acid On Cardiac Autonomic Neuropathy In Danio Rerio, Sophia Scott Jan 2023

The Effect Of Alpha Lipoic Acid On Cardiac Autonomic Neuropathy In Danio Rerio, Sophia Scott

Williams Honors College, Honors Research Projects

Even though poor glycemic control puts diabetics at risk for a variety of disease complications, there are many barriers to achieving adequate blood glucose regulation. Diabetic cardiac autonomic neuropathy (CAN) is a progressive autonomic denervation of the cardiovascular system resulting from chronic hyperglycemia and characterized by orthostatic hypertension, sinus tachycardia, low heart rate variability, and fatal arrhythmias. Available CAN therapeutics are limited to symptom alleviation, and do not prevent or reverse cardiac nerve damage. Limited evidence from clinical trials suggests the antioxidant Alpha Lipoid Acid (ALA) prevents CAN progression (Lee et al., 2017; Serhiyenko et al., 2020; Tankova, Koev & …

The Affect Of Dha Concentration On Lipid Membrane Fluidity, Elise Rezabeck Jan 2023

The Affect Of Dha Concentration On Lipid Membrane Fluidity, Elise Rezabeck

Williams Honors College, Honors Research Projects

My proposed project will be creating large unilaminar vesicles with varying concentrations of DHA in the vesicles. The diffusion in and out of these vesicles will be tracked by using singular lipid molecules. These singular lipid molecules will be tagged with fluorescence and viewed using an internal reflection fluorescence microscope. The data collected over the course of this project will then be analyzed using Matlab. The result of the project will be to find the specific lipid diffusion constant of these unilaminar vesicles. The relationship between the concentration of DHA and lipid diffusion and membrane fluidity will be found as …

Nicotine And E-Cigarette Effect On Maximal Metabolic Rate (Mmr) In Procambarus Clarkii, Nathan Cuttica Jan 2023

Nicotine And E-Cigarette Effect On Maximal Metabolic Rate (Mmr) In Procambarus Clarkii, Nathan Cuttica

Williams Honors College, Honors Research Projects

The use of electronic cigarettes, commonly referred to as e-cigarettes or vapes, has increased exponentially in recent years despite the lack of research into potentially negative physiological effects. The rise in e-cigarette use is likely due to the overwhelming evidence regarding the addictive properties of nicotine and the damage caused by traditional tobacco cigarettes. Most e-cigarettes, however, still contain nicotine and are associated with a rise in nicotine-related intoxications (Belkoniene et al., 2019). There are nicotine-free e-cigarette options, which are gaining popularity, but it is unknown if they carry similar or other negative physiological effects. Studies involving e-cigarettes containing nicotine …

The Effects Of Sex On Zebrafish Bone Metabolism, Simon Bagatto Jan 2023

The Effects Of Sex On Zebrafish Bone Metabolism, Simon Bagatto

Williams Honors College, Honors Research Projects

I investigated the effects of diet and sex on zebrafish bone metabolism. Zebrafish were subjected to either a high-calorie or low-calorie diet over a five-week period. After this diet, zebrafish scales were used to measure alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) enzyme activity with a fluorescent substrate. The enzyme activities were markers of the osteoblasts (ALP) and the osteoclasts (TRAP). These data were compared among sex and diet of the zebrafish using a repeated measures ANOVA statistical test. I also measured the number of resorption lacunae per scale (another indicator of osteoclast activity). The results showed higher TRAP …

Buoyancy And Emergence In Elymus Species, Sandor Jakab Jan 2023

Buoyancy And Emergence In Elymus Species, Sandor Jakab

Williams Honors College, Honors Research Projects

It is important that native plants are able to successfully emerge in wetlands because they play an important role in maintaining diversity. The relationship between seed buoyancy and seedling emergence was analyzed for three wetland species of the same genus: Elymus canadensis (FACU), Elymus macgregorii (FACW), and Elymus virginicus (FACW). E. macgregorii seemed to be the most buoyant, and E. virginicus and E. canadensis demonstrated very similar, less buoyant levels. Based on the idea that more buoyant seeds will be deposited at a shallow depth, and less buoyant seeds will be deposited at a deeper soil level, a relationship between …

Amphisbaenian Head Movement And Burrowing Forces In Damp Granular Media, Jacob Newell Jan 2023

Amphisbaenian Head Movement And Burrowing Forces In Damp Granular Media, Jacob Newell

Williams Honors College, Honors Research Projects

Damp granular media is a difficult environment to study because it is both practically complex and it lacks equations which fully describe its behavior. In this study, an oscillatory lateral head movement and its effects while penetrating damp granular media were tested using a robophysical model. This experimental research was inspired by the burrowing behavior of the clade Amphisbaenia, a group of usually limbless squamates that employ a variety of different burrowing behaviors, but it can apply to a wide range of burrowers. This research could help with both human burrowing technologies and the further investigation of animal behaviors.

Knockout Of Endospanin 1 Via Crispr In Zebrafish, Danio Rerio, Jared Kittinger Jan 2023

Knockout Of Endospanin 1 Via Crispr In Zebrafish, Danio Rerio, Jared Kittinger

Williams Honors College, Honors Research Projects

I made endospanin 1 knockout (KO) zebrafish to examine its effects on lipid and bone metabolism. Endospanin 1, or leptin receptor overlapping transcript (leprot), is a cytosolic protein linked to the protein hormone leptin that influences the trafficking of leptin receptors to the plasma membrane of cells. Genes for endospanin and tyrosinase (a pigmentation enzyme) were targeted via a microinjection of guide RNAs and CRISPR Cas9 into zebrafish embryos at 2-4 cell stages. I was able to disrupt the endospanin 1 gene (based upon the disruption of tyrosinase), but very few mutant zebrafish fully developed into adults. Only low KO …

Sustainability And Biomimicry, Tabitha Cooper Jan 2023

Sustainability And Biomimicry, Tabitha Cooper

Williams Honors College, Honors Research Projects

Standardization of business management systems is a vital part of the economic climate, as it allows for fair play between all competitors and encourages growth. The International Organization of Standardization (ISO) independently provides hundreds of standards ranging through dozens of different fields, providing standards applying to topics from quality management to material composition. Not much is readily available on the topics of ISO 9001, which outlines quality management systems (QMS) ISO 14001, which details environmental management systems (EMS), and their relation to ISO 18457, which explains what composes a biomimetic material or product. A literature review was conducted to gather …

Bite Force And Stress Content Of Varying Tooth Morphology And Jaw Alignment, Emily Newenhisen Jan 2023

Bite Force And Stress Content Of Varying Tooth Morphology And Jaw Alignment, Emily Newenhisen

Williams Honors College, Honors Research Projects

Biting is one of the primary physical actions against a food substrate involved in the process of mastication and consumption. Different shapes of solidified tissue (e.g., teeth, mandibles) are developed based on dietary preferences of the organism to improve mechanical function. Models of various geometric shapes were used to loosely replicate several common biological tooth structures. As there is little research conducted on tooth penetration force in relation to tooth shape, collection of such data was the main focus of the research. Molariform teeth found in herbivores were represented by the ball model, carnivorous needle-like teeth by cone model, cutting …

The Use Of Dynamic Light Scattering To Determine Mineral Precipitation In Bacteria., Isabella Manning Jan 2023

The Use Of Dynamic Light Scattering To Determine Mineral Precipitation In Bacteria., Isabella Manning

Williams Honors College, Honors Research Projects

Particulate calcium carbonate (PCC) is an important filler in industrial products, including paints, which are obtained through mining and sintering (heating to 1,100°C), which is responsible for up to 2% of global emissions. A potential solution to reduce these emissions is the use of calcium carbonate produced by bacteria from atmospheric , through a process called microbially induced carbonate precipitation (MICP). One of the primary uses of PCCs is in paint, requiring them to be a specific size (~700nm). When we have been collecting samples of PCCs produced by bacteria, which demonstrate clumping, making them too large for use in …

How Akron Affects The Water Quality Of The Cuyahoga River, Allyson Darst Jan 2023

How Akron Affects The Water Quality Of The Cuyahoga River, Allyson Darst

Williams Honors College, Honors Research Projects

The purpose of this study was to analyze whether the city of Akron had negative effects on the Cuyahoga river, and determine if the Cuyahoga River was naturally able to remediate these effects downstream. The pollutants measured in this study include hardness, bromine, residual chlorine, iron, copper, lead, nitrate, nitrite, ammonium chloride, total chlorine, fluoride, carbonate, pH, total alkalinity and cyanuric acid. This study was done using water testing strips (JNW Direct) to test the water at four different locations along the Cuyahoga River downstream (North) of Akron. This study found that the city of Akron does have a negative …

Effects Of Electronic Cigarettes On The Standard Metabolic Rate Of Crayfish, Frank Shaffer Jan 2023

Effects Of Electronic Cigarettes On The Standard Metabolic Rate Of Crayfish, Frank Shaffer

Williams Honors College, Honors Research Projects

This study aims to examine the relationship between e-cigarettes (both with nicotine and without) and the effects on the standard metabolic rates of crayfish. In this experiment, 42 crayfish were divided into three treatment groups and exposed to a designated solution of either a control solution or a solution containing e-cigarette (nicotine or non-nicotine) vapor. Their standard metabolic rates were then measured by using oxygen uptake as a proxy. The results of this experiment indicate that there is no statistically significant relationship between the use of electronic cigarettes and the standard metabolic rate of crayfish. However, this study can be …

Finding An Accurate Method To Measure Pollinator Visitation Rates, Claire Struhsaker Jan 2023

Finding An Accurate Method To Measure Pollinator Visitation Rates, Claire Struhsaker

Williams Honors College, Honors Research Projects

Pollinator visitation rates are a helpful way to monitor the health of ecosystems; however, there lacks a standardized method for obtaining these rates. The pollinator visitation rates for five plant species were collected and the relationship between the standard error of these rates and the time interval was determined. Monarda, Echinacea purpurea, Pycnanthemum muticum, and Baptisia alba all exhibit more accurate pollinator visitation rates as the time interval increased. Trifolium repens exhibited less accurate pollinator visitation rates as the time interval increased.

Comparison Of Bone Robustness Between Fore- And Hindlimb In Neonatal And Infant Galago, Jeremy Busken Jan 2023

Comparison Of Bone Robustness Between Fore- And Hindlimb In Neonatal And Infant Galago, Jeremy Busken

Williams Honors College, Honors Research Projects

According to Wolff’s law, bone is constantly subject to change based on loading patterns (Ruff et al., 2006). Those subjected to greater stress become more robust by increasing bone mineral density and often changing geometric properties. Previous studies of primate anatomy have found significant differences in bone robustness between the fore- and hindlimbs in adult leaping primates. Because leaping primates, such as Galago senegalensis and Galago moholi, subject their hindlimbs to much greater stress, this difference in robustness may come as a result of bone functional adaptation/Wolff’s law. This study sought to investigate this possibility by testing neonatal and infant …

Physiological Responses Between Collegiate Female Athletes Versus Collegiate Female Non-Athletes To Body Image, Nutrition, And Mental Health Inquiries., Molly Mcchesney, Jenna Holloway Jan 2023

Physiological Responses Between Collegiate Female Athletes Versus Collegiate Female Non-Athletes To Body Image, Nutrition, And Mental Health Inquiries., Molly Mcchesney, Jenna Holloway

Williams Honors College, Honors Research Projects

Participation in college athletics poses numerous demands on young adults. Not only are there academic demands to remain eligible for competition, but there are physical and mental demands that often go unnoticed. With all these demands, it is understandable how one's nutrition, body image, and mental health may go on the back burner, resulting in insufficiencies in all three categories. The results from this study suggest that athletes displayed stronger physiological responses with respect to heart rate and blood pressure when responding to the surveys surrounding the topics of mental health, body image, and nutrition as compared to non-athletes. The …

Effects Of Nicotine Vs Non-Nicotine E-Cigarettes On The Aggression Behavior Of Crayfish, Alex Kimble

Williams Honors College, Honors Research Projects

This study looks at the effects of e-cigarette vapor with and without nicotine on the aggression behavior of crayfish (Procambarus clarkii). The treatment groups were control, nicotine, and non-nicotine. The mass, sex, and condition of their appendages were recorded before random assignment. For the purpose of this study only male crayfish were used as there were not enough female crayfish. The crayfish were then randomly assigned to each group and contained ten crayfish that were subjected to the e-cigarette vapor infused water environment for a twenty-four hour period. The control group did not contain e-cigarette vapor and was exposed to …

Dimerization Of Dopamine (D2) And Adenosine (A2a) Receptors, Elena Vukovic, Jackson M. Macatol Jan 2023

Dimerization Of Dopamine (D2) And Adenosine (A2a) Receptors, Elena Vukovic, Jackson M. Macatol

Williams Honors College, Honors Research Projects

This experiment will aim to explore the heterodimerization of Dopamine (D2) and Adenosine (A2A) receptors in cells when bound to different ligands. Borroto-Escuela et al. (2018) investigated the dimerization of the A2AR-D2R in rats when administered with cocaine, and they found that disrupting the A2AR-D2R complex restores the cocaine reward system in rats. This was a strong influence as to why we chose to investigate A2AR-D2R dimerization in Chinese hamster ovary (CHO) cells.

One goal that we have for this experiment is to combine our current laboratory skills with new laboratory techniques. There are constantly new techniques in labs, and …

Attitudes Toward Transgenic Corn Usage Among Amish & Conservative Mennonite Farmers In Ohio, Scot Long Aug 2022

Attitudes Toward Transgenic Corn Usage Among Amish & Conservative Mennonite Farmers In Ohio, Scot Long

Journal of Amish and Plain Anabaptist Studies

Mass adoption and planting of genetically modified corn are part of the larger industrialized agricultural production system in the United States. Amish and conservative Mennonite farmers in the Holmes County settlement region offer an alternative production system often characterized by lower usage of chemical inputs, greater implementation of crop rotation, and significantly higher usage of hybrid versus GMO field corn. Moreover, the rationale among Amish/Mennonite farmers toward adoption of GMO (based on “convenience”) or rejection of GMO (based on “too many unknowns”) stems both from cultural diffusion of neighboring farms as well as variable need for nonfarm income. This article …

Potential Effects Of Amynthas Agrestis Invasion On Woody Understory Flora In The Cvnp, Christian Mammana Jan 2022

Potential Effects Of Amynthas Agrestis Invasion On Woody Understory Flora In The Cvnp, Christian Mammana

Williams Honors College, Honors Research Projects

Ohio forests are threatened by the invasive ecosystem engineer A. agrestis. A. agrestis invasion typically co-occurs with the ecosystem engineer, Odocoileus virginianus, where their impacts may synergize. To determine the direct effects of A. agrestis invasion, fenced plots across the Cuyahoga Valley National Park that excluded deer were utilized. The species richness, Shannon diversity and evenness of woody understory flora was measured in each plot. Mustard extraction was used to determine earthworm abundance. Correlations between abundance and measured variables were used to highlight potential invasion effects. Abundance and species richness was found to have a significant, positive correlation (p = …

This study investigated the viability of bioremediating rubber cryogrind using enriched indigenous bacteria. To begin the experiment, soils from three highway roadside locations in Kansas, KS 96 and West, KS 400 and 143rd, and 199th, were collected and transported to the lab to be studied. An initial soil characterization was run on the soil samples using distilled (DI) water mixture and 0.01 M CaCl2 to assess conductivity. The soils were tested to gather a baseline of the relationship between pH and conductivity and the impact of its distance from the roadside. Bacteria were isolated from …

Functionalization Of Chitosan Based Microparticles For In Vitro 3d Culture Of Human Liver Cells, William Imes Jan 2022

Functionalization Of Chitosan Based Microparticles For In Vitro 3d Culture Of Human Liver Cells, William Imes

Williams Honors College, Honors Research Projects

Previous work involving 3D culture of human liver cells with fluorinated chitosan based microparticles has shown that their incorporation provides needed structural cues that the culture of cells alone does not provide, such as increased gas transport. However, using bare microparticles to grow large 3D cellular structures is not practical as they tend to collapse before any meaningful research can be done on them. It is proposed that this is because of the lack of extracellular matrix (ECM)components within pure cell culture in vitro, which in their absence cannot adequately facilitate biochemical communication and adhesion between cells. It is …

How Will The Expanding Agriculture Business Impact Wetland Graminoids?, Travis Darago Jan 2022

How Will The Expanding Agriculture Business Impact Wetland Graminoids?, Travis Darago

Williams Honors College, Honors Research Projects

As human population increases in the years to come so too will the agriculture business. We already know that the use of chemicals such as fertilizers and other agriculture chemicals aid to promote accelerated crop growth so that we can harvest the most food for the most people. There is less research on the effects that the runoff of these chemicals have on wetland environments that are downstream from farmland. I would like to explore how some of these agricultural chemicals, like ammonium, will affect the growth of different types of wetland graminoids. I will be looking at Poa polustris, …

Immunohistochemistry Of The Zebrafish Retina, Alison Ruf Jan 2022

Immunohistochemistry Of The Zebrafish Retina, Alison Ruf

Williams Honors College, Honors Research Projects

The retina is made up of many different essential parts that work together to allow for vision of many different species. Some of these crucial cells and proteins include ganglion cells, bipolar cells, photoreceptors, synaptic markers, such as VGLUT, and receptors that bind glutamate. The basis of this project is to use immunohistochemistry to identify these important aspects of the retina in zebrafish via monoclonal and polyclonal antibodies. The goal being to compare specific cells in a control group retina to an experimental group of zebrafish.
