Characterization Of Cell-Cell Interactions In Normal And Tw32 Homozygous Preimplantation Mouse Embryos, Phyllis M. Johnson

Immediate objectives of the following studies were to characterize and compare normal versus lethal mutant (tW32/tW32) cellular interactions during preimplantation mouse development. Principal experimental methods used included embryo culture and microscopy. Control and mutant embryos were analyzed in vitro to identify chronological events and phenotypic characteristics of tW32 homozygotes. Cell-cell associations of uncompacted and compacted 8-cell control (ICR) embryos were analyzed by quantitative electron microscopy. Lastly, light microscopic analyses were performed on control and mutant (tW32/tW32) morulae.

The objective of this experiment was to measure the effects of level of wintering nutrition and pasture treatment upon growth and reproduction of Hereford heifers from weaning to 4 1/2 years of age. Levels of certain blood constituents were determined at periodic intervals throughout the study as additional indicators of adequacy in intake for some major nutrients and for measures of general health of the animals. Purine-thiamine anabolism during cellular morphogenesis in Myxococcus xanthus was investigated. One of the approaches was to test vegetative and myxospore (2.5 h and 8 h) extracts of M. xanthus CW-2 for their ability to synthesize the second de novo intermediate, 5'-phosphoribosylglycinamide, from beginning precursors either by way of phosphoribosyl-pyrophosphate amidotransferase (EC 2.4.2.14) or ribose-5-phosphate aminotransferase. Results indicate the presence of both the amido- and aminotransferases in both types of extracts, and both enzymes appear to be present at about the same level (per milligram of protein) in vegetative cells, myxospores (2.5 h and 8 h), and in a bacterial prototype, …

Many specialty crops, especially vegetables, are shallow rooted and sensitive to periods of moisture stress. This makes irrigation of vegetables necessary to their production in South Dakota since summer rainfall is normally both inadequate and undependable. A study was initiated in conjunction with the South Dakota Agricultural Experiment Station and the Water Resources Institute to evaluate irrigation of specialty crops in eastern South Dakota. The treatment of wastewater to alleviate our freshwater needs and protect the environment at a reasonable cost is a major problem in the world today. The application of partially-treated wastewater to the land may be one method of solving this problem. Land application is neither a new concept, nor one that will solve all treatment problems, but it is likely to play an ever-increasing role in water pollution control planning. Haemophilus somnus was the tentative name given in 1969 to a fastidious gram negative pleomorphic coccobacillus which was consistently isolated from cattle affected with a clinical syndrome called thrombo embolic meningoencephalitis (TEME)(1). The organism was initially classidied as an Actinobacillus actinoides- like organism, but because of its demand for a blood base for primary isolationit was reclassified as Haemophilus-like. Bailie then designated the species name somnus, referring to the sleepiness commonly exhibited by TEME affected cattle(1). The ability of different fly ash samples to remove orthophosphate from standard phosphate solutions and from wastewater samples is reported. The results of studies to determine the temperature, the amount of fly ash, and the time necessary for maximum orthophosphate removal are included. The mode of action, whether by absorption onto the fly ash or by a precipitation reaction with one or more of the soluble components from the fly ash, is also investigated. The chemical compounds present in the fly ash, as determined by x-ray diffraction, are also included.

The need exists for a detection system which is specific in its diagnosis of viruses. The system presented in this investigation uses antiserum treated virus infected cell cultures to establish specificity. The need exists for an automated virus detection system. The system described in this thesis uses the Boulter Counter, a computer analysis, and a simplified procedure which makes the system suitable and automation. For more than thirty years, Mary McCarthy has served as the conscience of the liberal American intellectual. McCarthy’s method of writing is satire, and she treats both her sympathetic and unsympathetic characters satirically. The characters in her fiction are generally liberal intellectuals who follow one of two patterns. The sympathetic characters are those who, despite their flaws, endeavor to be conscientious, objective and truth seeking. They are plagued by self-doubt and constant awareness o their own limitations. In contrast, her unsympathetic characters are egotistical and self-deluding. The purpose of this work is to study the sensitivity of emulsions of various hypophosphite salts to irradiation by photons in the x-ray region of the electromagnetic spectrum. This hypophosphate system bears little resemblance to the more common silver halide currently used in emulsions. Consequently, little is known about the reaction conditions necessary to make fast, sensitive emulsions or how to develop resultant latent images. Detection of edges in imagery represented by the matrix of data and classification of the data surrounded by an edge are investigated. An edge detection algorithm is developed and used to locate edges in digitized imagery. An algorithm called the adjacency classifier is developed to classify all groups in adjacent data points that are surrounded by edges. These two algorithms incorporate three important characteristics of the gradient vector: high gradients are inherent at edges, low gradients are inherent at edges, low gradients are inherent within homogeneous objects and gradient vector directions are perpendicular to the edge direction. The vascular vegetation of Ordway Prairie, a preserve maintained by The Nature Conservancy as an example of virgin prairie in McPherson County, South Dakota, was studied to determine its present composition. The sample comprised representatives of each species recognized in each of the 13 square-mile sections within the prairie, collected in 1975 and 1 9 7 6 . Three hundred nine different species, representing 170 genera in 55 plant families were found. One thousand six hundred forty five permanent herbarium specimens, serving as vouchers for their distribution, are now in the herbarium of South Dakota State University .

The present study was undertaken to determine the kinds and numbers of obligate anaerobes possibly present in various frozen and refrigerated commercial convenience foods. Also, known numbers of pathogenic species of obligate anaerobes were added to various convenience foods and stored at refrigeration and freezing temperatures to determine their die-off or loss in viability. Forest, Miller and Raizen (1963) and Hutcheson and Olson (1967) have conducted two of the only investigations on soil algae in a prairie ecosystem. The present investigation dealt first with identification of genera of algae present in the soil and an evaluation of their numbers. Secondarily a relationship was established between the numbers of algae in the soil (by species) and their environment, by the correlation coefficients. The variation in the environment was then used to predict the occurrence of algae in the soil using stepwise additive multiple regression analysis. The Easter lily, L. longiflorum Thunb. Is known not only for its ornamental value, but also for its large chromosomes, making it ideal cytological material. Big bluestem, Andropogon gerardii Vitman, is a South Dakota native grass. It possesses many desirable characteristics and unique adaptations to this region. This research has primarily dealt with the development of methods for cloning the species L. longiflorum and A. gerardii by tissue culture of leaf and/or floral parts. The pattern of plantlet differentiation from the cultures was also histologically studied.

Sixty Brookings, South Dakota students in three schools participated in a group interview, week-long nutrition study designed to determine the pattern and quantity of their dietary intakes. The nutritional adequacies of the diets were determined. Comparisons were made between the student’s dietaries in the different schools and days of the week, family incomes, and levels of mother’s education. Special attention was given to lunch patterns and consumption of vitamins A and C. Fanger of Denmark was the first scientist to generalize the physiological basis for predicting human thermal comfort based on activity level, clothing type, air temperature, air velocity, mean radiant temperature, and air humidity. Thus any activity level and clothing type, it is possible to determine combinations of air temperature, mean radiant temperature, air humidity and relative air velocity which would produce optimum thermal comfort. The fish populations of a 193 km (120 mi) section of the James River between Tacoma Park and Redfield, South Dakota that is scheduled for stream modification under the Oahe Irrigation project were evaluated from October 74 to July 76. Twenty-two species of fish were captured in the James River between Tacoma Park and Redfield, South Dakota. The fishes use the area primarily as a spawning ground and nursery for young-of-the-year. Evidence of spawning was found for 19 species of fish. Only a residual population of forage fishes, young-of-the-year of various species, and a few adult black bullheads attempt to …
