Heterodera glycines, the soybean cyst nematode, penetrates soybean roots and migrates to the vascular cylinder where it forms a feeding site called the syncytium. MiRNA396 (miR396) targets growth-regulating factor (GRF) genes, and the miR396–GRF1/3 module is a master regulator of syncytium development in model cyst nematode H. schachtii infection of Arabidopsis. Loss of pod dehiscence is a key step during soybean [Glycine max (L.) Merr.] domestication. Genome-wide association analysis for soybean shattering identified loci harboring Pdh1, NST1A and SHAT1-5. Pairwise epistatic interactions were observed, and the dehiscent Pdh1 overcomes the resistance conferred by NST1A or SHAT1-5 locus, indicating that Pdh1 predominates pod dehiscence expression. Further candidate gene association analysis identified a nonsense mutation in NST1A associated with pod dehiscence. Allele composition and population differential analyses unraveled that Pdh1 and NST1A, but not SHAT1-5, underwent domestication and modern breeding selections. Premise of research. A male-sterile, female-sterile mutant was discovered in a w4-m mutable line of Glycine max L. The mechanism of its sterility was not well understood. Therefore, different cytological and microscopic techniques were undertaken to better understand the process of mutant phenotype development. Aconitase catalyzes the reversible isomerization of two tricarboxylic acids, citrate and isocitrate, during the Krebs cycle. Five aconitase genes, namely, Aco1, Aco2, Aco3, Aco4, and Aco5, have been identified in soybean. Previously, Aco4 was mapped on chromosome 11. The purpose of this investigation was to isolate and sequence the candidate gene for Aco4. We mapped the Aco4 gene to a 148 kb region on chromosome 11 that contained 19 predicted genes. We have identified a viable-yellow and a lethal-yellow chlorophyll-deficient mutant in soybean. Segregation patterns suggested single-gene recessive inheritance for each mutant. The viable- and lethal-yellow plants showed significant reduction of chlorophyll aand b. Photochemical energy conversion efficiency and photochemical reflectance index were reduced in the viable-yellow plants relative to the wildtype, whereas the lethal-yellow plants showed no electron transport activity. The viable-yellow plants displayed reduced thylakoid stacking, while the lethal-yellow plants exhibited failure of proplastid differentiation into normal chloroplasts with grana. The basis of VIGS is that a viral genome is engineered so that it can carry fragments of plant genes, typically in the 200 to 300 base pair size range. Intra-cultivar variation reported in pure lines of soybean has been hypothesized to result from genetic mechanisms contributing to de novo genetic variation. In this study we have detected allele switching by following segregation pattern of Aconitase-4 isozyme in sexual crosses and pure lines. In sexual crosses, one F2 plant showed switch at the Aconitase- 4 (Aco4) locus from the expected heterozygous genotype Aco4-ac to Aco4-ab. Chlorophyll-deficient mutants have been studied persistently to understand genetic mechanisms controlling metabolic pathways. A spontaneous chlorophyll-deficient lethal mutant was observed in self-pollinated progeny of a soybean cultivar “BSR 101”. Observed segregation patterns indicated single-gene recessive inheritance for this lethal-yellow mutant. The objectives of this investigation were to develop a genetic linkage map of the region containing the lethal-yellow (YL_PR350) gene and identify putative candidate genes for this locus. The YL_PR350 gene was mapped to chromosome 15 and is flanked by BARCSOYSSR_15_1591 and BARCSOYSSR_15_1597. Male and female reproductive structures play an important role in seed development in plants. Abnormalities in male or female reproductive structures can lead to sterility. In soybean, Glycine max (L.) Merr., about 75 sterility mutants have been identified and most of them have been mapped to chromosomes. Mapping results have shown that some chromosomal regions are hotspots for fertility genes. Fine mapping of some of the male-sterile, female-fertile mutants and male-sterile, femalesterile mutants resulted in identification of candidate genes for fertility. Cell-free Fsg culture filtrates containing phytotoxins have been shown to develop foliar sudden death syndrome (SDS) in soybean. We have investigated the changes in protein profiles of diseased leaves caused by cell-free Fsg culture filtrates prepared from Fsg isolates. Two-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis (PAGE) was conducted to investigate the protein profiles of diseased and healthy leaves. We previously isolated a partial soybean cDNA clone whose transcript abundance is increased upon infection by the sedentary, endoparasitic soybean cyst nematode Heterodera glycines. We now isolated the corresponding full-length cDNA and determined that the predicted gene product was similar to the group of cofactor-dependent phosphoglycerate mutase/bisphosphoglycerate mutase enzymes (PGM/bPGM; EC 5.4.2.1/5.4.2.4). Floral nectaries of annual cultivated Glycine max develop between the bases of the central gynoecium and lateral stamen ring. Each discoid nectary forms immediately before flower opening and degenerates within 24 h. Three stages of nectary development are identified: preactive, active, and postactive. Preactive and active nectaries are composed of a single-layered epidermis that contains many open stomata, with guard cells having thickened walls, starch-engorged plastids, and other organelles. Cytoplasmic male sterility has been found independently in soybean three times since 1995, but no microscopic investigation has been published. The purpose of this microscopic study was to establish the developmental sequence leading to sterility in a cytoplasmic male-sterile soybean line that has been found to be stable under all environmental conditions tested and to demarcate the temporal and spatial parameters that result in degeneration of the microspores and pollen grains. Young developing soybean seeds contain relatively large amounts of calcium oxalate (CaOx) monohydrate crystals. A test for Ca and CaOx indicated that Ca deposits and crystals initially occurred in the funiculus, where a single vascular bundle enters the seed. Crystals formed in the integuments until the embryo enlarged enough to crush the inner portion of the inner integument. Crystals then appeared in the developing cotyledon tissues and embryo axis. Consumption of soybeans and food products made from them is increasing because of their desirable nutritional value. However, the oxalate content of seeds from 11 cultivars of soybean showed relatively high levels of total oxalate from 0.67 to 3.5 g/100 g of dry weight. Oxalate primarily was found as calcium oxalate crystals. Thirteen tested commercial soyfoods contained between 16 and 638 mg of total oxalate per serving. The hatching of Heterodera glycines eggs in soybean root exudates collected after postemergence application of three herbicides, and the hatching potential of H. glycines eggs from females feeding on herbicide-treated plants, were measured in vitro. Hatching in all root exudate solutions (RES) was greater than in deionized water but less than in 0.003 M ZnSO[sub4] solution. Filtering RES with a 0.22-[mu]m-filter increased H. glycines hatching in RES. Developing soybean seeds accumulate very large amounts of both soluble oxalate and insoluble crystalline calcium (Ca) oxalate. Use of two methods of detection for the determination of total, soluble, and insoluble oxalate revealed that at +16 d postfertilization, the seeds were 24% dry mass of oxalate, and three-fourths of this oxalate (18%) was bound Ca oxalate. During later seed development, the dry mass of oxalate decreased. Crystals were isolated from the seeds, and X-ray diffraction and polarizing microscopy identified them as Ca oxalate monohydrate.
