Monitoring Immediate-Early Gene Expression Through Firefly Luciferase Imaging Of Hrs/J Hairless Mice, Michael E. Geusz, Anne M. Collaco

Background: Gene promoters fused to the firefly luciferase gene (luc) are useful for examining gene regulation in live transgenic mice and they provide unique views of functioning organs. The dynamics of gene expression in cells and tissues expressing luciferase can be observed by imaging this enzyme's bioluminescent oxidation of luciferin. Neural pathways involved in specific behaviors have been identified by localizing expression of immediate-early genes such as c-fos. A transgenic mouse line with luc controlled by the human c-fos promoter (fos::luc) has enabled gene expression imaging in brain slice cultures. GraphicsA new method for the synthesis of 5-arylethynyl-8-hydroxyquinoline ligands using Sonogashira-Hagihara coupling was developed. The electronic nature of arylethynyl substituents affects the emission color and quantum yield of the resulting Al(III) complex. Photophysical properties of the metallocomplexes correspond to the electron-withdrawing/-donating character of the arylethynyl substituents. Optical properties of such AI(Ill) complexes correlate with the Hammett constant values of the respective substituents. This strategy offers a powerful tool for the preparation of electroluminophores with predictable photophysical properties.

To address questions regarding the potential impact of elevated total phosphorus (TP) inputs (due to relaxed regulations of TP loading), a series of TP enrichment experiments were conducted at pelagic stations in the 3 hydrologically distinct basins of Lake Erie. Results of nutrient assimilation measurements and assays for nutrient bioavailability suggest that the chemical speciation, and not concentration, of nitrogenous compounds may influence phytoplankton community structure; this in turn may lead to the selective proliferation of cyanobacteria in the eastern basin of the lake. This paper deals with a pole assignment problem by single-input state feedback control arising from a one-dimensional vibrating system with aerodynamic effect. On the practical side, we derive explicit formulae for the required controlling force terms, which can reassign part of the spectrum to the desired values while leaving the remaining spectrum unchanged. On the mathematical side, unlike the classical Sturm–Liouville problem, our eigenvalue problem is associated with a cubic pencil with unbounded operators as coefficients and has many interesting new features, one of which is that a new controllability condition appears. Explicit solvent and counterion molecular dynamics simulations have been carried out for a total of > 80 ns on the bacterial and spinach chloroplast 5S rRNA Loop E motifs. The Loop E sequences form unique duplex architectures composed of seven consecutive non-Watson-Crick basepairs. The starting structure of spinach chloroplast Loop E was modeled using isostericity principles, and the simulations refined the geometries of the three non-Watson-Crick basepairs that differ from the consensus bacterial sequence. The deep groove of Loop E motifs provides unique sites for cation binding. Binding of Mg2+ rigidifies Loop E and stabilizes its major groove at an intermediate …

Three programs have been developed to aid in the classification and visualization of RNA structure. BPViewer provides a web interface for displaying three-dimensional (3D) coordinates of individual base pairs or base pair collections. A web server, RNAview, automatically identifies and classifies the types of base pairs that are formed in nucleic acid structures by various combinations of the three edges, Watson-Crick, Hoogsteen and the Sugar edge. RNAView produces two-dimensional (2D) diagrams of secondary and tertiary structure in either Postscript, VRML or RNAML formats. Crossing Offline And Online Media: A Comparison Of Online Advertising On Tv Web Sites And Online Portals, Louisa Ha

This paper presents findings of a study comparing the advertising on the web sites supported by offline media and on the dot.com media that only have online presence. The study analyzes the advertising strategies of leading U.S. TV networks' web sites and online portals, which respectively represent web sites with strong offline media support and web sites with no offline media counterparts respectively. TV networks' advertising strategies were identified based on a spectrum of brand extension and brand integration strategies. Peptide-Protein Interactions: Photoinduced Electron-Transfer Within The Preformed And Encounter Complexes Of A Designed Metallopeptide And Cytochrome C, Robin C. Lasey, Liu Liu, Ling Zang, Michael Y. Ogawa

Photoinduced electron-transfer (ET) occurs between a negatively charged metallopeptide, [Ru(bpy)2(phen-am)-Cys-(Glu)(5)-Gly](3-) = RuCE(5)G, and ferricytochrome c = Cyt c. In the presence of Cyt c, the triplet state lifetime of the ruthenium metallopeptide is shortened, and the emission decays via biexponential kinetics, which indicates the existence of two excited-state populations of ruthenium peptides. The faster decay component displays concentration-independent kinetics demonstrating the presence of a preformed peptide-protein complex that undergoes intra-complex electron-transfer. Values of K-b = (3.5 +/- 0.2) x 10(4) M-1 and k(ET)(obs) = (2.7 +/- 0.4) x 10(6) s(-1) were observed at ambient temperatures. The magnitude of k(ET)(obs) decreases …

Although democratization in Thailand in the 1990s is commonly characterized as a classic case of modernization theory in action, economic globalization provides a better explanation for Thailand's democratization process. Economic growth in the country has been based on foreign capital and has created a globalized economy sensitive to the confidence of world capital markets. Moreover, the Thai middle classes cannot be characterized as having coherent political preferences, and it is arguable that the 1992 middle class protests were more about suspicions of official corruption than about democracy.

Most community-based models in ecology assume that all individuals within a species respond similarly to environmental conditions and thereby exert identical effects as consumers or prey. Rather, individuals differ among systems, with important implications for population demographics and community interactions. For widely distributed assemblages made up of poikilotherms with high first-year mortality, species-specific differences in growth reaction norms as affected by both temperature and genotype will influence biotic interactions. For a broadly distributed fish assemblage, first-year growth does not vary with latitude for a planktivorous prey species, but declines with increasing latitude for a terminal piscivore. Severe summer monsoon flooding occurred along the Yangtze River Valley (YRV) in 1998. The heaviest precipitation fell during June 8-23 and July 20-29 and coincided with midtropospheric blocking over northeast Eurasia. An index based on the difference between Okhotsk High and west Pacific Low intensity is developed in this study to quantify blocking; analysis shows that blocking and equatorward displacement of the upper-tropospheric east Asian jet preceded (accompanied) the June (July) period. This suggests that heavy rainfall during each interval was caused by different atmospheric processes. An examination of joint variations in the index and YRV precipitation during 1958-1998 revealed …

Since the enactment of the Drug Free Schools and Communities Act Amendments of 1989, there have been several additional attempts by Congress to address the problem of alcohol abuse on college campuses. In 1 990, Congress passed the Student Right-to-Know and Campus Security Act containing a provision requiring institutions to notify students of the number of arrests for liquor law violations. In the 1998 Higher Education Amendments (HEA), Congress proposed a set of initiatives institutions should take "to change the culture of alcohol consumption on college campuses." In this paper, we discuss the history of the two organizations and the history of other attempts at organizational consolidation; explore the similarities and differences in the organizations’ missions and values, structures, and activities; and consider each organization’s unique characteristics. We conclude with a set of recommendations concerning organizational consolidation and will raise a number of pertinent questions we feel the profession must address as it considers consolidating ACPA and NASPA.

Throughout the United States, resident assistants (RAs) manage residence hall environments, develop communities, and contribute to the educational mission of student affairs. RA positions provide leadership experiences, opportunities for personal and professional development, and the potential to influence and assist students with whom they live and work. Some, however, ask whether the RA job is too demanding for students (Dodge, 1990) and if too much is expected of RAs (Bierman & Carpenter, 1994). Photoinduced Electron-Transfer Along Alpha-Helical And Coiled-Coil Metallopeptides, Anna Fedorova, Anita Chaudhari, Michael Y. Ogawa

A peptide-based electron-transfer system has been designed in which the specific positions of redox-active metal complexes appended to either an a-helix, or an a-helical coiled-coil, can be reversed to test the effect of the helix dipole in controlling photoinduced electron-transfer rates. Two 30-residue apopeptides were prepared having the following sequences: (1) Ac-K-(IEALEGK)(ICALEGK)(IEALEHK)-(IEALEGK)-G-amide, and (II) Ac-K-(IEALEGK)(IHALEGK)-(IEALECK)(IEALEGK)-G-amide. Each apopeptide was reacted first with [Ru(bpy)(2)(phen-ClAc)](2+), where bpy = 2,2'-bipyridine and phen-ClAc = 5-chloroacetamido-1,10-phenanthroline, to attach the ruthenium polypyridyl center to the cysteine side-chain of the polypeptide. The isolated products were then reacted with [Ru(NH3)(5)(H2O)](2+) to yield the binuclear electron-transfer metallopeptides ET-I and ET-II. …

Aggregation properties of meso-tetratolylporphyrins bearing cationic substituents of a lipophilic nature such as ammonium, pyridinium, phosphonium, sulfonium, and isothiouronium were studied by UV/vis and resonance light scattering spectroscopy. The exciton point-dipole approximation was used to predict the structural alignment of the porphyrin units within the aggregate. The contribution of various types of aggregates depends on the porphyrin substitution, ionic strength, and temperature of aqueous solution of the porphyrin. In general, the preferred structure of an aggregate formed by temperature-induced aggregation is the J-aggregate exhibiting the characteristically narrow, red-shifted Soret band indicating a parallel "side-by-side" arrangement of the porphyrin units.
