Purification And Properties Of A Hela Cell Enzyme Able To Remove The 5'-Terminal Protein From Poliovirus Rna, Victor Ambros, David Baltimore

Using a rapid phenol extraction assay, an enzyme was purified from uninfected HeLa cells that can cleave the 5'-terminal protein (VPg) from poliovirus RNA. Both cytoplasmic and nuclear extracts had enzymes with similar behavior. A polypeptide of molecular weight 27,000 was the major one present in the purified preparation. Assuming that this protein is the enzyme, a very low turnover number was calculated for it. The purified enzyme would cleave the tyrosine-phosphate bond linking VPg to poliovirus RNA with minimal degradation of the RNA or of VPg. The kinetics of rod outer segment production and turnover were studied in Irish setter dogs with a recessively inherited photoreceptor degeneration (rod-cone dysplasia). Affected and littermate control (heterozygote) dogs, 16–50 days of age, were injected intravitreally with [3H]leucine after electroretinographic studies. Normal age-matched mongrel dogs were also used as controls. Between 6 hr and 8 days following intravitreal injection, eyes were removed and processed for microscopic and radioautographic studies.
In affected dogs, the rod components of the electroretinogram were not recordable. Cone components were recordable but failed to develop normally. Morphologically, rod inner segments were diminutive and the …

Murine spleen cell surface IgD is found in two forms upon SDS-polyacrylamide gel electrophoresis under nonreducing conditions. Both IgDI and IgDII are present on the surface of intact cells and appear to be native structures. Neither form could be accounted for by proteolytic degradation or disulfide bond rearrangement. IgDII has an apparent m.w. of 96,000, suggesting an HL structure. IgDI has an apparent m.w. of 150,000, approximately 33,000 daltons lower than that expected for an H2L2 structure. Significant variation in the relative amounts of IgDI and IgDII was found when mice of different strains were examined.

There are many methods and techniques available to environmental designers to obtain and use information on spatial behavior, attitudes, preferences, opinions, and so on. Among them are rating-scale techniques. This paper discusses these from a particular theoretical orientation and covers the utility, concept, and an evaluation of rating scales. Three case studies are described to illustrate the application and usefulness of the graphic rating-scale technique to environmental designers.
