Characterization Of Genes Encoding Poly(A) Polymerases In Plants: Evidence For Duplication And Functional Specialization, Lisa R. Meeks, Bala Subrahmanyam Addepalli, Arthur G. Hunt

Background: Poly(A) polymerase is a key enzyme in the machinery that mediates mRNA 3′ end formation in eukaryotes. In plants, poly(A) polymerases are encoded by modest gene families. To better understand this multiplicity of genes, poly(A) polymerase-encoding genes from several other plants, as well as from Selaginella, Physcomitrella, and Chlamydomonas, were studied.

Methodology/Principal Findings: Using bioinformatics tools, poly(A) polymerase-encoding genes were identified in the genomes of eight species in the plant lineage. Whereas Chlamydomonas reinhardtii was found to possess a single poly(A) polymerase gene, other species possessed between two and six possible poly(A) polymerase genes. With the exception …

BACKGROUND: In soybean somatic embryo transformation, the standard selection agent currently used is hygromycin. It may be preferable to avoid use of antibiotic resistance genes in foods. The objective of these experiments was to develop a selection system for producing transgenic soybean somatic embryos without the use of antibiotics such as hygromycin.

RESULTS: When tested against different alternate selection agents our studies show that 0.16 microg/mL glufosinate, 40 mg/L isopropylamine-glyphosate, 0.5 mg/mL (S-(2 aminoethyl)-L-cysteine) (AEC) and the acetolactate synthase (ALS) inhibitors Exceed and Synchrony both at 150 microg/mL inhibited soybean somatic embryo growth. BACKGROUND: The Arabidopsis ortholog of the 30 kD subunit of the mammalian Cleavage and Polyadenylation Specificity Factor (AtCPSF30) is an RNA-binding endonuclease that is associated with other Arabidopsis CPSF subunits (orthologs of the 160, 100, and 73 kD subunits of CPSF). In order to further explore the functions of AtCPSF30, the subcellular distribution of the protein was examined by over-expressing fusion proteins containing fluorescent reporters linked to different CPSF subunits.

RESULTS: It was found that AtCPSF30 by itself localizes, not to the nucleus, but to the cytoplasm. AtCPSF30 could be found in the nucleus when co-expressed with AtCPSF160 or AtCPSF73(I), …

It is important to establish the various effects of legume cover crops on soil physicochemical properties because they have been considered for use as improved fallows (with shorter rest periods) to enhance development and maintenance of soil productivity. Our objectives were to assess: (i) aboveground dry matter yields of legume cover crops; and (ii) cover crop effects on weed infestation and soil physicochemical properties in a minimum tillage management system. Multimeric Hemicellulases Facilitate Biomass Conversion, Zhanmin Fan, Kurt Wagschal, Wei Chen, Michael D. Montross, Charles C. Lee, Ling Yuan

Two highly active trifunctional hemicellulases were constructed by linking the catalytic portion of a xylanase with an arabinofuranosidase and a xylosidase, using either flexible peptide linkers or linkers containing a cellulose-binding domain. The multifunctional enzymes retain the parental enzyme properties and exhibit synergistic effects in hydrolysis of natural xylans and corn stover.

Glandular trichomes of many plant species synthesize and secrete a variety of phytochemicals that play an important protective role against pathogens and pests. Acyl sugars, in particular, are powerful natural insecticides. We demonstrated the existence of the α-ketoacid elongation (αKAE) pathway, an alternative to the fatty acid synthase (FAS) pathway, for the extension of carbon chains of acyl groups in sugar esters (SE). The α-KAE pathway utilizes one carbon elongation per cycle in contrast to two carbons per cycle in FAS, and it was found to function in a variety of tobaccos and petunia.
