Panicum virgatum L., readily referred to as Switchgrass, is a perennial warm-season bunch grass, used as an alternative energy source for biofuel production. There is insufficient research on switchgrass pathogens, and is expected that an increase in disease pressure will result as more land is reserved for this perennial crop. Protoplasts are appropriate targets for genome editing, DNA functional screens and transgenesis. This project focuses on the comparison of inexpensive mesophyll protoplast isolation via the use of food-grade enzymes and transformation between diploid Panicum hallii Vasey (PAH) and polyploid cellulosic feedstock Panicum virgatum L (switchgrass), a relative of PAH. PAH has great potential as a C4 model species for crop and bioenergy research. Here an inexpensive switchgrass and PAH mesophyll protoplast isolation and transformation system was developed; the first protoplast system for PAH. Panicum virgatum (switchgrass), a perennial grass native to North America, is a leading biomass feedstock candidate for the manufacture of cellulosic ethanol. Switchgrass is considered a viable option for biofuel production due to its cheap production cost and ability to grow on marginal land. Biofuel derived from switchgrass has been shown to be very energy efficient, producing 540% more renewable energy versus nonrenewable energy expended. Switchgrass-derived biofuel is also estimated to have greenhouse gas emissions that are 94% lower than emissions from gasoline (Schmer et al 2008). Thousand Cankers Disease (TCD) is a disease complex involving the fungal pathogen Geosmithia morbida, an insect vector Pityophthorus juglandis, and the hosts, Juglans spp. and Pterocarya spp. Signs and symptoms of TCD include crown thinning due to branch dieback, yellowing and wilting of the leaves, appearance of epicormic shoots, numerous entrance/exit holes, gallery formation by P. juglandis, and the development of small, dark brown cankers underneath the bark. However, many techniques require invasive and complicated sample preparation procedures to detect and characterize engineered nanomaterials in complex matrices. In the first part of this thesis, we present a non-destructive and label-free approach based on surface enhanced Raman spectroscopic (SERS) mapping technique to qualitatively detect and characterize gold nanoparticles (AuNPs), on and in spinach leaves in situ. Iron is among the essential micronutrients for all living organisms and is a cofactor for many cellular redox reactions. Although iron is a highly abundant metal element found in the earth’s crust, it is also a limiting factor in plant development when it is present as insoluble ferric oxides. Plants have evolved two strategies to acquire soluble iron referred to as Strategy I and Strategy II. Our lab has focused on the Arabidopsis thaliana double mutant ysl1ysl3. Due to the constant concern with off-target contamination and application technology requirements associated with future herbicide-tolerant crops, the use of drift-reduction nozzle technology (DRT) may increase. The primary objective of this research was to evaluate the effects of coarse droplets generated by drift-reduction nozzles on foliar fungicide efficacy and residual in soybean infected by frogeye leaf spot caused by Cercospora sojina. No differences in disease control, soybean yield, spray retention, and residual when applying Quadris Top SB, a premix of azoxystrobin and difenoconazole, using nozzles that produce either a medium or ultra coarse droplet spectrum were determined.

Frogeye leaf spot (FLS) of soybean is caused by Cercospora sojina. In 2010, resistance to the quinone outside inhibitor (QoI) fungicides was reported. Since then, evaluating FLS for QoI-resistance has been of particular interest in Tennessee and other soybean-producing states. In order to determine alternative fungicide options, fungicides with solo and combination mode of action were tested in 2013-2014. The objectives were,1) to evaluate fungicide efficacy for disease control and yield protection, and 2) evaluate selection pressure for QoI-resistance. Due to the phase-out of methyl bromide, there is a need for alternative, nonchemical fumigation treatments in strawberry production. Anaerobic soil disinfestation and biofumigation are two non-chemical methods for controlling soilborne plant pathogens of strawberry. This study was designed to observe strawberry fruit nutrition and soil volatiles of a strawberry field being treated with biofumigation treatments, anaerobic soil disinfestation treatments, and a combination of the two alternative methods. Leaf nutrient concentrations are an example of a functional trait, a trait that can be used to understand community dynamics by observing how plants interact with their environment. I explored how leaf nutrient concentrations were affected over successional time by environmental changes. The majority of prior research on functional traits focuses on either woody or herbaceous species, but not both life forms simultaneously. To address this, I also explored the successional changes to leaf nutrient concentrations, separating the plant community into woody and herbaceous species.

The Southeastern United States is a biological hot-spot for carnivorous plants, with over half of the North American species occurring along the Gulf Coast. Pinguicula planifolia Chapman is one of six carnivorous perennial butterwort species found in the Florida Panhandle. Among these species, only P. planifolia expresses a distinct variation of red coloration on its leaves. The functional role of foliar anthocyanins may include stress response to drought and nutrient deficiency, herbivory defense, free radical scavenging, and photoprotection. Production of hazelnuts has historically been limited to regions with mild climates, with almost all hazelnut production in the United States occurring in Oregon. A recent boom in demand, along with an expanded selection of varieties with improved environmental tolerances, has pushed hazelnut production into new regions. Hazelnut production could prove profitable in areas of the Midwest that are less well-suited to the cultivation of maize and soybeans.
