Antibacterial Activity And Chemical Characterization Of Resin From Sciadopitys Verticillata (Thunb.) Siebold And Zuccarini, David Ira Yates

Sciadopitys verticillata produces white viscous resin that is unique among the conifers. This research investigated effects of resin on bacteria from different ecological niches and the chemical composition of the resin. Each bacterial species was evaluated separately for response to winter- and summer-collected resins. Exposure to winter-collected resin reduced numbers of colonies of Bacillus cereus, Erwinia amylovora, Agrobacterium tumefaciens, and Escherichia coli and increased numbers of Xanthomonascampestris, Pseudomonas fluorescens, and Pseudomonas syringae. Summer-collected resin affectedpopulation growth of two bacterial species; population counts of E. amylovora decreased and those of P. fluorescens increased. Selected strains of …

Large patch (Rhizoctonia solani AG 2-2LP) epidemics cause significant damage to Japanese lawngrass (JLG; Zoysia japonica) in the transition zone. Large patch primarily affects the stems and sheaths of JLG and is difficult to control using traditional fungicide sprays. Field and growth chamber experiments were conducted during 2015-2016 in TN and GA to evaluate methods to enhance fungicidal control of large patch in JLG landscapes. Panicum virgatum (switchgrass), a perennial grass native to North America, is a leading biomass feedstock candidate for the manufacture of cellulosic ethanol. Switchgrass is considered a viable option for biofuel production due to its cheap production cost and ability to grow on marginal land. Biofuel derived from switchgrass has been shown to be very energy efficient, producing 540% more renewable energy versus nonrenewable energy expended. Switchgrass-derived biofuel is also estimated to have greenhouse gas emissions that are 94% lower than emissions from gasoline (Schmer et al 2008). Protoplasts are appropriate targets for genome editing, DNA functional screens and transgenesis. This project focuses on the comparison of inexpensive mesophyll protoplast isolation via the use of food-grade enzymes and transformation between diploid Panicum hallii Vasey (PAH) and polyploid cellulosic feedstock Panicum virgatum L (switchgrass), a relative of PAH. PAH has great potential as a C4 model species for crop and bioenergy research. Here an inexpensive switchgrass and PAH mesophyll protoplast isolation and transformation system was developed; the first protoplast system for PAH. Thousand Cankers Disease (TCD) is a disease complex involving the fungal pathogen Geosmithia morbida, an insect vector Pityophthorus juglandis, and the hosts, Juglans spp. and Pterocarya spp. Signs and symptoms of TCD include crown thinning due to branch dieback, yellowing and wilting of the leaves, appearance of epicormic shoots, numerous entrance/exit holes, gallery formation by P. juglandis, and the development of small, dark brown cankers underneath the bark. Translation of mRNA into protein is a critical step in gene expression, but the principles guiding its regulation at the genome level are not completely understood. Translation can be quantified at a genome scale by measuring the ribosome loading of mRNA—the extent to which mRNA is associated with ribosomes. In this dissertation, I present investigations into how genome-wide ribosome loading is controlled in Arabidopsis thaliana. The natural recalcitrance of plant cell walls is a major commercial hurdle for plant biomass to be converted into a viable energy source as alternative to fossil fuels. To circumvent this hurdle manipulation of carbohydrate enzymes active in the cellulose and hemicellulose portions of the plant cell wall can be utilized to improve feedstocks. Production of cellulolytic enzymes by plants have been evaluated for reducing the cost associated with lignocellulosic biofuels. Eukaryotic organisms are defined by their compartmentalization and various organelles. The membranes that define these organelles require complex nanomachines (known as translocons) to selectively mediate the import of proteins from the cytosol where they are synthesized into the organelle. The plastid, (specifically the chloroplast) which is characteristic of plant cells, possibly represents the most complex system of protein sorting, requiring many different translocons located in the three membranes found in this organelle. Despite having a small genome, the vast majority of plastid-localized proteins are nuclear-encoded and must be post-translationally imported from the cytosol. Anaerobic soil disinfestation (ASD) is an environmentally friendly and cost effective pre-plant soil treatment technique that allows effective control of soilborne pests by creating anaerobic conditions, particularly for specialty and organic crop production under diverse environmental conditions. In spite of being a proven technique, ASD has to be optimized to fit into local production systems with specific pathogen pressure using locally available amendments for successful implementation on a commercial scale. In plants, ethylene functions as a hormone regulating many growth and developmental processes. Ethylene receptors in plants resemble bacterial two-component signaling systems. Because of this it, ethylene receptors are thought to have been acquired by gene transfer from the cyanobacterial endosymbiont that lead to the development of the chloroplast. However, prior to this work, functional ethylene receptors were thought to only be found in green plants. Here, we show that the cyanobacterium Synechocystis sp. Due to the constant concern with off-target contamination and application technology requirements associated with future herbicide-tolerant crops, the use of drift-reduction nozzle technology (DRT) may increase. The primary objective of this research was to evaluate the effects of coarse droplets generated by drift-reduction nozzles on foliar fungicide efficacy and residual in soybean infected by frogeye leaf spot caused by Cercospora sojina. No differences in disease control, soybean yield, spray retention, and residual when applying Quadris Top SB, a premix of azoxystrobin and difenoconazole, using nozzles that produce either a medium or ultra coarse droplet spectrum were determined.

Frogeye leaf spot (FLS) of soybean is caused by Cercospora sojina. In 2010, resistance to the quinone outside inhibitor (QoI) fungicides was reported. Since then, evaluating FLS for QoI-resistance has been of particular interest in Tennessee and other soybean-producing states. In order to determine alternative fungicide options, fungicides with solo and combination mode of action were tested in 2013-2014. The objectives were,1) to evaluate fungicide efficacy for disease control and yield protection, and 2) evaluate selection pressure for QoI-resistance. Due to the phase-out of methyl bromide, there is a need for alternative, nonchemical fumigation treatments in strawberry production. Anaerobic soil disinfestation and biofumigation are two non-chemical methods for controlling soilborne plant pathogens of strawberry. This study was designed to observe strawberry fruit nutrition and soil volatiles of a strawberry field being treated with biofumigation treatments, anaerobic soil disinfestation treatments, and a combination of the two alternative methods.
