The Southwestern US is predicted to become hotter and drier, as global climate change forces increasing temperatures and variability in timing and size of precipitation inputs. Drought stress has become more frequent in recent decades, and resulted in massive forest mortality in piñon-juniper woodlands. During recent severe droughts (2000-2003, 2009-2012), piñon pine (Pinus edulis Englem.) suffered disproportionately high mortality compared to co-occurring one-seed juniper (Juniperus monosperma [Engelm.] Sarg.). A large-scale precipitation manipulation experiment was established in a piñon-juniper woodland in central New Mexico to test hypotheses regarding tree survival and mortality with respect to altered water regimes. Pink rot of potato (Solanum tuberosum) is a widespread soilborne disease that causes significant losses in the field and storage. It is caused by Phytophthora erythroseptica (Pethybr.), an oomycete pathogen that produces sexual spores that can survive in soil for years. The management of pink rot mainly relies on chemical control. However, the most effective chemical in pink rot control, mefenoxam, is losing its efficacy owing to the development of mefenoxam resistance in P. erythroseptica. Salvinia molesta is an invasive aquatic fern. It is now the second worse aquatic invader in the world. Since the 1930s, it has invaded most tropical and some temperate countries. S. molesta plants grow vegetatively and can increase in size rapidly. S. molesta can form thick mats of up to 1-meter-thick. Sciadopitys verticillata produces white viscous resin that is unique among the conifers. This research investigated effects of resin on bacteria from different ecological niches and the chemical composition of the resin. Each bacterial species was evaluated separately for response to winter- and summer-collected resins. Exposure to winter-collected resin reduced numbers of colonies of Bacillus cereus, Erwinia amylovora, Agrobacterium tumefaciens, and Escherichia coli and increased numbers of Xanthomonascampestris, Pseudomonas fluorescens, and Pseudomonas syringae. Summer-collected resin affectedpopulation growth of two bacterial species; population counts of E. amylovora decreased and those of P. fluorescens increased. Selected strains of …

African leafy vegetables (ALVs) are a diverse set of crops grown across sub-Saharan Africa. They have been a staple of traditional diets and contain many critical micronutrients but their importance has been largely ignored by researchers and policymakers at the expense of imported crops. Availability, accessibility, and utilization of ALVs are limited by factors across the supply chain. This research investigated seed systems, drought tolerance, hydroponic production, market barriers, consumer preference, and the impact of gender as related to ALVs. Annual forage cropping systems are a vital aspect of livestock forage production. One area where this production system can be enhanced is the integration of novel annual forages into conventional cropping systems. Two separate projects were conducted to investigate alternative forage options in annual forage production. Black walnut (Juglans nigra L.) is a fine hardwood tree species native to the central hardwood region of the United States. High-quality black walnut timber is highly desirable. Traded in both regional and global markets, it has been used for veneer, and the manufacture of high-end products such as cabinets, furniture, and gunstocks. As a result of its high economic value, black walnut has been commercially cultivated for many years, and breeding programs have generated superior timber genotypes with improved marketable traits. Turfgrass lawns require supplemental nitrogen (N) to maintain green color and seasonal shoot density. Improper lawn fertilization with excess N or phosphorus has the potential to contaminate both surface and groundwater. Phytophthora blight is a serious threat to the Midwest vegetable industry, because the oomycete pathogen responsible for this disease, Phytophthora capsici, has a wide host range, can spread quickly in fields, and produces resilient oospores that can survive in soil for years. Phytophthora capsici has become resistant to commonly used fungicides and resistant crop varieties are rare. Amending soil with complex organic substrates has potential to improve soil quality and suppress soil-borne pathogens including P. capsici. Incremental gains in grain yield of maize hybrids over the decades are the consequence of genotype, environment and management interactions. Historically, genetic improvements in newer hybrids have included longer active grain filling periods (achieved by advancing silking and extending functional stay green in maize leaves); stronger source and sink during grain filling; enhanced tolerance to higher density; and canopy architecture changes. Palmer amaranth is a pernicious summer annual weed that has evolved resistance to six herbicide sites of action in the U.S. and threatens agriculture production. In 2011, Palmer amaranth was identified in flood plains in two southern Indiana counties (Posey and Vanderburgh). DNA methylation is an important epigenetic mark present in many eukaryotes, and is involved in many crucial biological processes, such as gene imprinting, regulation of gene expression, and genome stability. Proper genomic DNA methylation patterns are achieved through the concerted action of DNA methylation and demethylation pathways. In the model plant species Arabidopsis thaliana, ROS1 (REPRESSOR OF SILENCING 1) is one of the DNA demethylases and the key component in the demethylation pathway. Pro-Vitamin A (proVA) carotenoids, which are converted into retinol (Vitamin A) in the human body, have been the subject of human nutrition studies and are a target for biofortification of staple crops. Historically, β-carotene has been the principal target for enhancing levels of proVA, yet there is recent interest in enhancing the proVA carotenoid β-cryptoxanthin. This thesis examines the effects of shade on ‘Prime-Ark 45’ blackberries (Rubus spp.) in greenhouse (GH) and field (FD) experiments aimed at improving fruit production in the southern United States Ozark Plateau region. Primocane blackberry production in the southern United States is limited in acreage of production as well as low yields by high temperatures during the bloom and early fruiting period, resulting in poor fruit set and poor fruit quality. Mixed Linkage Glucan (MLG, or (1,3;1,4)-ß-D glucan) is a component of cell walls for major cereal crops and is significant to food and beverage industries. To better understand genetic factors affecting MLG content in oats, this study investigates the presence of glucosidases likely to participate in MLG production. A glucosidase showing co-expression with CslF6—the primary gene responsible for MLG synthesis—could indicate a hand in MLG production by association. Greater sage-grouse (Centrocercus urophasianus; hereafter, sage-grouse) are a species of conservation concern in the rangelands of western North America due to their dramatic decline over the last half century. Effective conservation and management of sensitive species requires an understanding of how species respond to management actions. We examined two aspects of the reproductive phases of sage-grouse: nest predation, and habitat selection by female sage-grouse with chicks. The purpose of this study was to use soil physical and chemical analyses to better understand the ancient agricultural landscape around the ancient Maya cities of Rio Amarillo and Piedras Negras, two tributary sites to Copan, Honduras. Our primary objective was to determine whether a mass erosion event around 800 A.D. occurred which could have caused crop failure and famine or if stable soil conditions persisted during the collapse of these city-states. Large patch (Rhizoctonia solani AG 2-2LP) epidemics cause significant damage to Japanese lawngrass (JLG; Zoysia japonica) in the transition zone. Large patch primarily affects the stems and sheaths of JLG and is difficult to control using traditional fungicide sprays. Field and growth chamber experiments were conducted during 2015-2016 in TN and GA to evaluate methods to enhance fungicidal control of large patch in JLG landscapes. The apple rootstock ‘Geneva® 41’ (‘G.41’), is desirable for its resistance to fire blight, Erwinia amylovora, while producing high yields. However, ‘G.41’ and other Geneva® selections tend to form weak graft unions in the nursery that are susceptible to breaking in the wind, at harvest, or during shipment. Panicum virgatum (switchgrass), a perennial grass native to North America, is a leading biomass feedstock candidate for the manufacture of cellulosic ethanol. Switchgrass is considered a viable option for biofuel production due to its cheap production cost and ability to grow on marginal land. Biofuel derived from switchgrass has been shown to be very energy efficient, producing 540% more renewable energy versus nonrenewable energy expended. Switchgrass-derived biofuel is also estimated to have greenhouse gas emissions that are 94% lower than emissions from gasoline (Schmer et al 2008). The Arabidopsis thaliana meiotic mutant 6491 has been identified as displaying temperature sensitive male reduced- fertility. It has been determined that callose wall formation is defective, both in temporal and structural areas. There is irregular rippling in the plasma membrane and aberrant formation of the exine portion of the pollen wall. A developmental study using brightfield, epifluorescence, and transmission electron microscopy of the early stages of wall formation in 6491 has been completed, along with a similar study of Arabidopsis thaliana (L.) heynh, ecotype Wassilewskija (WS). Thousand Cankers Disease (TCD) is a disease complex involving the fungal pathogen Geosmithia morbida, an insect vector Pityophthorus juglandis, and the hosts, Juglans spp. and Pterocarya spp. Signs and symptoms of TCD include crown thinning due to branch dieback, yellowing and wilting of the leaves, appearance of epicormic shoots, numerous entrance/exit holes, gallery formation by P. juglandis, and the development of small, dark brown cankers underneath the bark. Unusual monoenoic fatty acids (UMFA’s) and specialized metabolites called anacardic acids (AnAc) are produced in glandular trichomes of Pelargonium ´ hortorum (geranium). The UMFA’s, 16:1∆11 and 18:1∆13 are precursors for the synthesis of unsaturated AnAc 22:1n5and 24:1n5 that contribute to pest resistance in geraniums. UMFAs and their derived AnAc metabolites not only provide a useful biological marker that differentiates the biosynthetic pathway for unusual mononenes from the common fatty acids (i.e. stearic, palmitic, oleic, linoleic and linolenic) but also have industrial, medical and agricultural applications. Protoplasts are appropriate targets for genome editing, DNA functional screens and transgenesis. This project focuses on the comparison of inexpensive mesophyll protoplast isolation via the use of food-grade enzymes and transformation between diploid Panicum hallii Vasey (PAH) and polyploid cellulosic feedstock Panicum virgatum L (switchgrass), a relative of PAH. PAH has great potential as a C4 model species for crop and bioenergy research. Here an inexpensive switchgrass and PAH mesophyll protoplast isolation and transformation system was developed; the first protoplast system for PAH. Invasive species have been recorded to cause large economic and ecological impacts on various ecosystems and their services. Locally, giant reed (Arundo donax L.), a woody grass native to the Mediterranean, adversely impacts riparian ecosystems in southwestern United States by aggressively displacing their native flora and fauna. Giant reed also has become a cause of concern for national water security, especially in water-limited areas of the arid southwestern United States.
