The Role Of Auxin Resistant 1 (Axr1) In Arabidopsis Cytokinin Signaling, Yan Li

The plant hormone cytokinin plays essential roles in many aspects of growth and development. The cytokinin signal is transmitted by a multistep phosphorelay to the members of two functionally antagonistic classes of Arabidopsis response regulators (ARRs): the type-B ARRs (response activators) and type-A ARRs (negative-feedback regulators). Previous studies have shown that mutations in AXR1, encoding a subunit of the E1 enzyme in the related to ubiquitin (RUB) modification pathway, leads to decreased cytokinin sensitivity. This research shows that the cytokinin resistance of axr1 seedlings is suppressed by loss-of-function of type-A ARRs and that the cytokinin resistance caused by …

Headwater stream systems are important components of the overall hydrologic system. Forestry best management practices (BMP) are effective at minimizing non point source pollution from forest harvesting activities. Streamside management zones (SMZ) are one BMP used to protect surface water quality by maintaining shade near streams, filtering runoff, and minimizing soil disturbance near streams. An evaluation of BMP effectiveness on the watershed scale was conducted at the University of Kentucky’s Robinson Forest. Six watersheds were harvested using a two-age deferment harvest with one of three SMZ configurations applied to each watershed. Two unharvested watersheds served as controls.

The normalized difference vegetative index (NDVI) has been correlated with physiological plant parameters and used to evaluate plant growth. There is little information about the use of this technique to detect soybean nutrient deficiencies. The objective of this work was to determine the ability of the NDVI sensor to detect P and K deficiencies, and grain yield reduction, in soybean. During 2010 and 2011, NDVI measurements were made on a soybean field trial site known to exhibit yield responses to both P and K nutrition. Despite the agronomic benefits of exotic grass plantings for pasture and rangeland forage, exotic grass invasions are capable of having dramatic and widespread impacts on native communities and ecosystems. Exotic Old World bluestem grasses (OWBG; Bothriochloa and Dichanthium spp.) have become increasingly invasive throughout the central and southern U.S. Little is known regarding the impacts of OWBG invasion on native grassland and savannah ecology and how to successfully control OWBG invasions in natural areas. Nicotine and nornicotine are the two main alkaloids that accumulate in Nicotiana tabacum L. (tobacco), and nornicotine is the N-demethylation metabolite of nicotine. Nicotine is synthesized in the root, and probably primarily in the root tip. Both nicotine and nornicotine exist as two isomers that differ from each other by the orientation of H atom at the C-2' position on the pyrrolidine ring. (S)-nicotine is the dominant form in tobacco leaf and the enantiomer fraction of nicotine (EFnic), the fraction of (R)-enantiomer over the total nicotine, is approximately 0.002. Twenty-four steers were grazed on endophyte (Neotyphodium coenophialum)-infected [[Lolium arundinaceum (Schreb.) Darbysh]; E+] tall fescue (TF) and exposed to ergot alkaloids for an 106-d grazing period. Cattle were removed from pasture, placed in dry lots, and fed a non-toxic diet to evaluate changes in vascular contraction relative to time-off E+ TF pasture. Lateral saphenous veins (SV) were biopsied from steers at 0-, 21-, 42-, and 63-d off TF pasture and from 6 control steers at 0- and 63-d off bermudagrass (BG) pasture. Tall fescue (Lolium arundinaceum) is the principal cool-season species within pastures of the southeastern USA and is known to have a mutualistic relationship with a fungal endophyte (Neotyphodium coenophialum) that produces the ergot alkaloids responsible for tall fescue toxicosis. Management of the reproductive growth of tall fescue is necessary, as the seedheads contain the highest concentrations of ergot alkaloids, and livestock have been documented to selectively graze these tissues. Corn nitrogen (N) applications are still done on a field basis in Kentucky, according to previous crop, soil tillage management and soil drainage. Soil tests, as well as plant analysis for N, are not very useful in making N fertilizer rate recommendations for corn. Recommended rates assume that only 1/3 to 2/3 of applied N is recovered, variability largely due to the strong affect of weather on the release of soil N and fertilizer N fate. The use of exotic resistance quantitative trait loci (QTL) provides one strategy for breeding wheat cultivars resistant to Fusarium Head Blight (FHB), a devastating disease of wheat. The objective of this study was to evaluate the effectiveness of two QTL, Fhb1 and QFhs.nau-2DL, in diverse genetic backgrounds and to evaluate their effects on agronomic and quality traits. Five populations from crosses between FHB susceptible parents (26R58, KY97C-0574-01, 25R54, KY97C, KY97C-0554-02, 25R78 and KY93C-1238-17-1) and FHB-resistant VA01W-476, were evaluated in the FHB nursery at Lexington, KY in 2010 and 2011. Nineteen bermudagrass (Cynodon dactylon (L.) Pers.) cultivars were evaluated for field emergence, establishment rate, traffic tolerance, post-harvest seed coating, germination velocity, and total germination under varying temperature regimes. Two cultivars were evaluated for thermal modeling, day/night temperature fluctuations, day lengths, and effects of fluridone on speed and percentage of total germination.

The effect of cultivar was highly significant for visible field germination, time to 100% cover, and traffic tolerance in both 2010 and 2011. Riviera was the slowest or equivalent to the slowest for visible germination. Casino Royale was the fastest or equivalent to the fastest for visible germination. …

Field studies examined the strategies of mowing, herbicide, fertility, and all combinations on tall ironweed populations, weed biomass, pasture yield, grazing, economics, and forage quality at three Kentucky locations. Mowing was performed in July 2008 and 2009, herbicide applied in August 2008, and fertilizer applied in September 2008 and 2009 at all locations. Weed populations were measured in 2008, 2009, and 2010, and forage and weed biomass collected in May or June of 2009 and 2010. Herbicide treatments reduced weed biomass at all locations, and reduced tall ironweed stems by 64% or greater in 2009 at all locations. Weed biomass …

Climate change is likely to alter plant species composition and interactions between plants and soil microbes that together dictate the quantity and quality of forage produced in pastures, the base of animal production in central Kentucky. This study assessed the seasonal dynamics of soil microbes and their response to increased temperature (+3oC) and growing season precipitation (+30% of the mean annual). Total soil microbial biomass, community composition, enzyme activities, potential carbon mineralization, and catabolic responses to selected substrates were measured seasonally in the different climate treatments.
