Bovine in-vitro production(IVP) of embryos is a growing field for producing offspring with desirable genetics in the cattle industries. While IVP is effective, oocyte cytoplasmic maturation is compromised, and bovine oocytes matured in-vitro have reduced metabolic activity than those matured in-vivo. Mitochondria are the central unit of oocyte metabolism, producing ATP through OXPHOS. Photobiomodulation is a light treatment reported to improve metabolic activity. In five experiments, we measured the effects of photobiomodulation treatment with red LED at 16-(L-16) and 20 h(L-20) of bovine oocyte in-vitro maturation on subsequent embryonic development, mitochondrial activity and nuclear progression. In vitro produced (IVP) bovine embryos provide enhanced genetic value and a means for studying early embryonic development. However, IVP embryos are developmentally compromised and are lower quality compared to in vivo derived (IVD) embryos. Consequently, after transfer pregnancy rates from IVP embryos are consistently lower, as they exhibit the highest amounts of pregnancy failure during the early embryonic phase (prior to day 28). Progesterone (P4) has been shown to influence bovine endometrial epithelial (EPI) and stromal fibroblast (SF) cells to secrete developmentally important molecules that promote early embryo development and conceptus elongation. Changes to the embryonic environment that occur within the first few days of development can have significant effects on subsequent fetal development. The following studies investigate the effects of progesterone-deprivation during the early preimplantation period in the mare. The first study described embryo morphology (diameter, stage, and quality grade) collected from mares with induced aluteal cycles. The second study evaluated the effect of induced aluteal cycles on the future fertility of embryo collections as a potential method for cycle manipulation of embryo donor mares. In the centuries following, dramatic advances have been made in the field of infertility research through the employment of a variety of species. Despite these advances, determination of oocyte and embryo quality remains largely subjective. The focus of this study was to elucidate the molecular and cellular mechanisms whereby fibroblast growth factors (FGFs) mediate outgrowth of the limb. Specifically, we examined the epistatic relationship between FGF and Wnt/Planar cell polarity (PCP) signaling in establishing cell polarity as a mechanism for outgrowth. By implanting beads into embryonic limbs and lateral plate mesoderm, we established that FGF activates Wnt5a in a gradient fashion. Once it was established that Wnt5a was expressed at the right time and place to turn on PCP signaling, we investigated the ability of Wnt5a to influence cell migration and/or cell polarity. BACKGROUND: Runx proteins are developmentally important metazoan transcription factors that form a heterodimeric complex with the non-homologous protein Core Binding Factor beta (CBFbeta). CBFbeta allosterically enhances Runx DNA binding but does not bind DNA itself. We report the initial characterization of SpCBFbeta, the heterodimeric partner of SpRunt-1 from the sea urchin Stronylocentrotus purpuratus.

RESULTS: SpCBFbeta is remarkably similar to its mammalian homologues, and like them it enhances the DNA binding of the Runt domain. SpCBFbeta is entirely of zygotic provenance and its expression is similar that of SpRunt-1, accumulating globally at late blastula stage then later localizing to endoderm and …

BACKGROUND: Runx transcription factors play critical roles in the developmental control of cell fate and contribute variously as oncoproteins and tumor suppressors to leukemia and other cancers. To discover fundamental Runx functions in the cell biology of animal development, we have employed morpholino antisense-mediated knockdown of the sea urchin Runx protein SpRunt-1. Previously we showed that embryos depleted of SpRunt-1 arrest development at early gastrula stage and underexpress the conventional protein kinase C SpPKC1.

RESULTS: We report here that SpRunt-1 deficiency leads to ectopic cell proliferation and extensive apoptosis. Suppression of the apoptosis by pharmacological inhibition of caspase-3 prevents the …

Leukemia inhibitory factor (LIF) is a pleiotropic cytokine which demonstrates perplexing physiological effects. It has been demonstrated that LIF is essential for implantation in mice. Little is known relating to the manner by which LIF effects pre-implantation and post-implantation development. The objectives of this project were to determine the effects LIF on pre-implantation development, to determine the effects that it may have on implantation rates, successful pregnancy rates, and resorption rates, and to determine the effects that LIF has on the skeletal development of mice.
